Plasmid, protein and restriction endonuclease analysis (REA) profiles and multilocus enzyme electrophoresis were used to effect a molecular discrimination of twenty-seven Campylobacter coli serogroup 20, biotype 1 (Lior) strains. These strains were not outbreak-associated but were isolated from a number of different countries and different animal and environmental sources. Each of the techniques was able to discriminate, to various degrees, between the serogroup 20, biotype 1 strains. The choice of a particular technique depends to a large extent on the level of discrimination desired, the previous experiences of the investigator and on the laboratory facilities at hand. REA profiles demonstrated the greatest degree of discrimination between these strains. Plasmid and protein profiles could discriminate reasonably well. Multilocus enzyme electrophoresis (allozyme typing) and protein profiles may prove effective in subgrouping serogroup 20, biotype 1 strains.  相似文献

A periphyton biofiltration system of mariculture effluents was studied to identify ecological processes and management procedures that strongly affect the biofilter functioning, in order to attain optimal biomass production and nutrient removal. The multivariate statistical technique of factor analysis allowed reducing the large amount of data available into three main factors. The first factor, which accounted for 49% of the overall data variability, was herein called “biological activity” as it represents the joint effects on the variables measured of photosynthesis, N and P uptake, respiration and decomposition of organic matter. The second and third factors were “autotrophic biomass density” and “nitrite and phosphate uptake/release balance”, which, respectively, accounted for further 20% and 14% of the data variability. A conceptual model, describing the functioning of the periphyton biofilters, revealed a delicate equilibrium among the different processes, whose understanding help to manage the biofilters towards optimal production of periphytic biomass and nutrient removal. Raising flow rate raised the overall nutrient uptake rate but reduced uptake efficiency and diluted nitrifying particles. A reduced flow rate led to sedimentation of organic particles, decomposition and nutrients re‐mineralization. Apparently, control of water flow and nutrient content, periphyton substrate area and the cleaning of the effluent supply system are key management elements for the operation of a periphyton‐based biofilter system that maximizes both periphyton biomass production and nutrient removal.  相似文献

The roots of Rumex bucephalophorus, collected in Israel, were analyzed for trans-stilbenes. Two stilbene-O-glycosyl derivatives were identified, in addition to 3,5,4'-trihydroxystilbene (1) (resveratrol). The stilbene-O-glycosyl derivatives were 5,4'-dihydroxystilbene-3-O-beta-d-glucopyranoside (2) (piceid) and the new 5,4'-dihydroxystilbene-3-O-alpha-arabinopyranoside (3), which is being named rumexoid. The structure of rumexoid was elucidated by using spectroscopic data. The antioxidant capacities of stilbenoids 1-3 were determined and expressed as trolox equivalent antioxidant capacity (TEAC). TEAC value for trans-resveratrol was highest (2.7) and for rumexoid lowest (1.5). In vitro, trans-resveratrol and rumexoid demonstrated a potent inhibitory effect on alpha-glucosidase activity (IC50 < 0.1 and < 0.5 mM, respectively). The commercial antidiabetic agent acarbose was shown to inhibit only 35% of the enzyme activity at 0.5 mM. The addition of piceid to the reaction mixture did not inhibit alpha-glucosidase in vitro in the range of concentrations used. These findings extend the range of reported beneficial effects of stilbene derivatives, and demonstrate the multifaceted activities that dietary polyphenols may exert in the intestine, where their concentrations are highest in the body.  相似文献

Nonhuman primates represent the most relevant model organisms to understand the biology of Homo sapiens. The recent divergence and associated overall sequence conservation between individual members of this taxon have nonetheless largely precluded the use of primates in comparative sequence studies. We used sequence comparisons of an extensive set of Old World and New World monkeys and hominoids to identify functional regions in the human genome. Analysis of these data enabled the discovery of primate-specific gene regulatory elements and the demarcation of the exons of multiple genes. Much of the information content of the comprehensive primate sequence comparisons could be captured with a small subset of phylogenetically close primates. These results demonstrate the utility of intraprimate sequence comparisons to discover common mammalian as well as primate-specific functional elements in the human genome, which are unattainable through the evaluation of more evolutionarily distant species.  相似文献

Jojoba Israel is a world-leading producer of Jojoba products, whose orchards are covered with sensors that collect soil moisture data for monitoring plant needs at real-time. Based on these data, the company’s agronomist defines a weekly irrigation plan. In addition, data on weather, irrigation, and yield are recorded from other sources (e.g. meteorological station and irrigation-plan records). However, so far, there has been no attempt to use the entire set of collected data to reveal insights and interesting relationships between different variables, such as soil, weather, irrigation characteristics, and resulting yield. By integrating and utilizing data from different sources, our research aims at using the collected data not only for monitoring and controlling the crop, but also for predicting irrigation recommendations. In particular, a dataset was constructed by integrating data collected over almost two years from 22 soil-sensors spread in four major plots (which are divided into 28 subplots and eight irrigation groups), from a meteorological station, and from actual irrigation records. Different regression and classification algorithms were applied on this dataset to develop models that were able to predict the weekly irrigation plan as recommended by the agronomist. The models were developed using eight different subsets of variables to determine which variables consistently contributed to prediction accuracy. By comparing the resulting models, it was shown that the best regression model was Gradient Boosted Regression Trees, with 93% accuracy, and the best classification model was the Boosted Tree Classifier, with 95% accuracy (on the test-set). Data that were not contributing to the model prediction success rate were identified as well. The resulting model can significantly facilitate the agronomist’s irrigation planning process. In addition, the potential of applying machine learning on the company data for yield and disease prediction is discussed.  相似文献

Cultural monitoring was used to determine the incidence and sources of salmonellae in a 4160-bird broiler flock raised on litter in 32 pens. Twenty-five of the pens remained apparently free of salmonellae during the 49-day growing period. Salmonella johannesburg, first detected in the meat meal component of the starter ration, was recovered from the litter of seven pens and from the intestines of dead or culled chicks from two pens. Salmonella alachua was also recovered from two of these pens. Culture of swabs collected from the plastic crates used to transport this flock for processing showed that 97/112 (86.6%) were contaminated with salmonellae (15 serovars) before the birds were loaded. The crate washer at the plant did not remove salmonellae from these crates: 97/132 (73.5%) crates sampled after washing yielded salmonellae. Eleven serovar were recovered, including S. johannesburg and S. alachua introduced by the infected flock. Twelve of 31 chickens (38.7%) collected when the birds were unloaded at the processing plant were intestinal carriers of S. johannesburg and/or S. alachua and 29 (93.5%) were external carriers. Salmonella johannesburg, S. alachua and four other serovars were isolated from the feathers of these birds. Eleven of 25 (44%) carcasses tested from this flock yielded salmonellae. Salmonella johannesburg or S. alachua, first isolated from the infected flock, were recovered from five carcasses and S. haardt and S. Typhimurium, first isolated from the transport crates were recovered from six carcasses.  相似文献

Geosmin and 2-methylisoborneol (MIB) are semi-volatile terpenoid compounds produced as secondary metabolites by benthic and planktonic cyanobacteria, several genera of fungi, and various actinomycetes. These off-flavor compounds pose a heavy economic burden in the aquaculture industry rendering fish unmarketable unless purified by purging with large quantities of clean water. In the present study, the presence of off-flavor compounds was examined in a recirculating aquaculture system (RAS) for tilapia culture. In this zero-discharge system, where water from the fish basins is recirculated through parallel aerobic (drum filter and a trickling filter) and anaerobic treatment loops (sedimentation/digestion basin), concentrations of geosmin and, in particular, MIB were highest in the aerobic treatment loop. Lowest concentrations were detected in the anaerobic treatment loop. This latter finding pointed toward a possible reduction of these compounds in this basin. Two bacterial strains of the streptomycetes family were isolated from the aerobic, organic-rich, drum filter and the nitrifying trickling filter. In vitro tests with these isolates, closely related to Streptomyces roseoflavus and Streptomyces thermocarboxydus, revealed that MIB production exceeded geosmin production under all conditions tested and was significantly higher under aerobic than under anoxic conditions. Under the latter conditions, with nitrate as an electron donor, the S. roseoflavus-like isolate was capable of denitrification. Based on the results obtained in this study, it was concluded that aerobic, organic-rich conditions stimulate the growth of actinomycetes and subsequent production of geosmin and MIB in the system. The observed reduction of these compounds in the anaerobic water treatment component may serve in designing treatment steps aimed at alleviating the problem of geosmin and MIB accumulation in recirculating systems.  相似文献

Cultural monitoring was used to study the incidence and sources of salmonellae in a 4160 bird broiler flock during the growing period, transport and processing in a commercial plant. No salmonellae were isolated from any of 132 litter samples of 189 chickens cultured during the seven-week growing period, even though nest litter samples from four of the eight parent flocks yielded salmonellae and Salmonella worthington was isolated from the meat meal component of the grower ration. On arrival at the plant, 2/23 birds sampled carried S. infantis on their feathers, although intestinal cultures failed to yield salmonellae. Three of 18 processed carcasses samples yielded salmonellae (S. infantis, S. heidelberg, S. typhimurium var copenhagen). The most likely source of these salmonellae was the plastic transport crates, since 15/107 sampled before the birds were loaded yielded salmonellae (S. infantis, S. typhimurium). The crate washer at the plant did not reduce the incidence of Salmonella-contaminated crates, since 16/116 sampled after washing yielded salmonellae (S. infantis, S. typhimurium, S. heidelberg, S. schwarzengrund, S. albany).  相似文献
