The many recent dry-grind plants that convert corn to ethanol are a potential source of substantial amounts of corn oil, if an economical method of separating it can be developed. Oil separation from corn germ by aqueous extraction (AE) was studied. Batches of 5–20% germ in a buffer solution were preheated in a pressure cooker, ground in a blender or colloid mill, churned in an incubator/shaker and centrifuged to separate a free oil fraction. The highest oil yields were obtained when the germ mass fraction was between 0.1 and 0.16. Addition of an enzyme solution to the ground germ suspension prior to churning, aqueous enzymatic extraction (AEE) gave an oil yield twice as high as the AE yield, carried out using similar conditions. Both types of extraction produced the most oil after a 122 °C cook followed by churning at 160 rpm and 70 °C. Oil yield increased linearly with dispersion loading, from 0.5 up to 1.0 g/cm2 based on beaker cross-section (0.09–0.12 germ mass fraction). Over a several month period the germ used for the extractions became more difficult to extract, despite storage at 4 °C.  相似文献

Thin interdigitated films composed of a long-chain, water-insoluble chiral acid (p-pentadecylmandelic acid of absolute configuration R) and a water-soluble chiral base (phenylethylamine, R') were constructed at the air-solution interface. The (R, R') structure was characterized to near-atomic resolution by grazing-incidence x-ray diffraction (GIXD). The two diastereomeric systems, (R, R') and (R, S'), demonstrate similar surface pressure-molecular area isotherms, but their structures are completely different on the molecular level, as monitored by GIXD. Complementary data on these two architectures were provided by atomic force microscopy.  相似文献

Science and technology studies (STS) research challenges the concept of technological determinism by investigating how the end users of a technology influence that technology’s trajectory. STS critiques of determinism are needed in studies of agricultural technology. However, we contend that focusing on the agency of end users may mask the role of political-economic factors which influence technology developments and applications. This paper seeks to mesh STS insights with political-economic perspectives by accounting for relationships between availability of diverse technologies, variations in political-economic structures, and farmer interests and characteristics. We present the results of an analysis on the recent development of three wheat varieties: (a) a wheat variety that was modified genetically to tolerate the herbicide glyphosate, (b) wheat varieties with characteristics selected to serve specific markets, (c) and emerging research and development of perennial wheat varieties. Using data obtained through a survey of wheat growers in Washington State, we analyzed whether farmer interest in these three clusters of wheat varieties was associated with distinct individual characteristics and attitudes and whether those characteristics and attitudes are consistent with political economic structures. Although our analysis did not allow us to assess the degree of direct influence that farmers have on the technological development trajectory for these types of wheat, we were able to document variation in technological alternatives and farmer characteristics related to different political-economic trends.  相似文献

Feral pigs are one of the most abundant free‐roaming ungulates in the United States, yet their role in the ecology and transmission of foodborne pathogens is poorly understood. Our objectives were to estimate the prevalence of Salmonella shedding among feral pigs throughout Texas, to identify risk factors for infection, and to characterize the isolates. Faecal samples were collected from feral pigs in Texas from June 2013 through May 2015. Standard bacteriologic culture methods were used to isolate Salmonella from samples, and isolates were characterized via serotyping and anti‐microbial susceptibility testing. The prevalence of faecal Salmonella shedding among sampled pigs was 43.9% (194/442), with positive pigs originating from 50 counties. Pigs sampled during fall and summer were significantly more likely to be shedding Salmonella than pigs sampled during winter. High serovar diversity was evident among the isolates, and many of the detected serovars are leading causes of human salmonellosis. The most common serovars were Montevideo (10.0%), Newport (9.1%), and Give (8.2%). Resistance to anti‐microbial agents was rare. The burgeoning feral pig population in the United States may represent an emerging threat to food safety.  相似文献

CDV is a highly contagious viral pathogen causing a lethal systemic disease in dogs and other carnivores. Several lineages or genotypes of CDV exist that are variously distributed throughout several continents. Legal or uncontrolled trading of animals may modify the epidemiology of CDV, introducing novel strains in CDV-na?ve areas or accounting for the resurgence of CDV in areas where vaccine prophylaxis was effective and successful to control the disease. A hemi-nested PCR system was developed to genotype strains of the major CDV lineages, America-1, Europe, Asia-1, Asia-2 and Arctic. The assay was tested using a collection of 27 laboratory and vaccine strains and of 36 field CDV strains. Distinct lineages could be differentiated by specific primers targeted to the H gene. The method could be useful for molecular epidemiological studies of CDV, providing a tool for large-scale studies, and for the diagnosis of vaccine-related disease.  相似文献

Kjeldahl analysis is commonly used to measure zein proteins in corn maize (N × 5.7) with no attempt to eliminate contribution from other nitrogen sources. In this study, dry milled corn was extracted with 70% ethanol or 0.1N NaOH and the zein content of the extract measured using capillary electrophoresis in the presence of sodium dodecyl sulfate. The amount of zein protein in alcohol extracts, using this method, was in good agreement with that determined by Kjeldahl nitrogen analysis. However, less than half of the Kjeldahl nitrogen in the alkaline extracts could be attributed to corn zein. Reproducibility expressed as relative standard deviation for migration time and peak area was 0.10 and 1.05, respectively. The technique permits rapid analysis of a large number of samples without interference from other compounds present in the extracts.  相似文献
