Measurements of movement along 28 boreholes reveal the three-dimensional flow field in a 6 million cubic meter reach of Worthington Glacier, a temperate valley glacier located in Alaska. Sliding at the bed accounted for 60 to 70 percent of the glacier's surface motion. Strain rates in the ice were low from the surface to a depth of about 120 meters, but then increased rapidly toward the bed. Ice deformation was not affected by temporal changes in the sliding rate. The three-dimensional pattern of motion indicates that plane strain, which is often assumed by models, is a poor approximation of this viscous flow.  相似文献

Es wird über die Herstellung eines spezifischen Antiserums gegen Schweine-Zona-pellucida im xenogenen System mit einem möglichst reinen Antigen in Form der isolierten Zona pellucida berichtet. Die Schweine-Eizellen werden durch Follikel-punktion von Schweine-Ovarien gewonnen. Nach Ablösung der Cumuluszellen erfolgt die mechanische Abtrennung der Zona pellucida durch Ansaugen mittels einer im Ver-gleich zur Eizelle im Durchmesser kleineren Kapillare. Dabei bricht die Zona pellucida an einer Stelle auh und die Oozyte entleert sich nach auβen. Das so isolierte Schweine-Zona-pellucida-Material (ca. 700 Zonen) wird einem Kaninchen subcutan injiziert. Nach drei Boosterungen wird das Anti-Zona-Serum nach 25 Tagen gewonnen und von verschiedenen Schweinegeweben absorbiert. Der anschlieβende Antikörpernachweis erfolgt durch die Präzipitatbildung an der Oberfläche der Zona pellucida-Membran einer cumulusfreien Schweine-Eizelle. Als weiterer Antikörpernachweis dient der indirekte Immunfluoreszenztest. In beiden Nachweismethoden fanden sich hohe Antikörperaktivitäten im Anti-Zona-Serum. Zum Nachweis der Kreurantigenität wurde das Anti-Zona-Serum auch gegen menschliche Eizellen und gegen Mäuse-Ei-zellen ausgetestet. Der indirekte Immunfluoreszenztest war an der menscblichen Zona pellucida positiv, die Präzipitatbildung dagegen negativ. Die Mäuse-Eizelle reagierte in beiden Nachweisverfahren negativ .  相似文献

The magnetometer and electron reflectometer investigation (MAG/ER) on the Mars Global Surveyor spacecraft has obtained magnetic field and plasma observations throughout the near-Mars environment, from beyond the influence of Mars to just above the surface (at an altitude of approximately 100 kilometers). The solar wind interaction with Mars is in many ways similar to that at Venus and at an active comet, that is, primarily an ionospheric-atmospheric interaction. No significant planetary magnetic field of global scale has been detected to date (<2 x 10(21) Gauss-cubic centimeter), but here the discovery of multiple magnetic anomalies of small spatial scale in the crust of Mars is reported.  相似文献

The distribution and prevalence of Thelleria buffeli in Queensland cattle were investigated using serum samples and blood films collected primarily for brucellosis surveillance and tick fever diagnosis. Serums from 8654 cattle from 357 farms throughout Queensland were examined by an indirect fluorescent antibody test for antibody to T buffeli. In addition, 347 peripheral blood films collected from 147 farms in south-eastern Queensland were examined for piroplasms of T buffeli. The overall herd and animal prevalences for T buffeli were 75% and 41%, respectively. There was significant variation among regions in both herd and animal prevalences (P less than 0.001). Herd and animal prevalences were highest in the north and east decreasing westward. The results indicate that T buffeli is more widespread in Queensland than previously thought.  相似文献

The cytokine tumor necrosis factor-alpha (TNF-alpha) plays a major role in inflammatory and immune-pathological reactions of the skin. With respect to a possible therapeutic modulation of TNF-alpha mediated activation of Nuclear Factor-kappa B (NF-kappaB) in canine cutaneous inflammation, we investigated the role of NF-kappaB and the involvement of reactive oxygen species (ROS) in the TNF-alpha signalling pathway in dermal fibroblasts of the dog. TNF-alpha treatment resulted in the activation of NF-kappaB as assessed by electrophoretic mobility shift assay (EMSA). Additionally, NF-kappaB translocation was induced with butylhydroperoxide and antimycin A, but not with hydrogen peroxide. TNF-alpha stimulated NF-kappaB activation was partially inhibited by preincubation with the antioxidants alpha-lipoic acid and butylated hydroxyanisol (BHA). No superoxide generation following TNF-alpha stimulation could be detected in the supernatant of canine fibroblasts with the superoxide dismutase-inhibitable cytochrome c reduction test. In contrast, production of TNF-alpha dependent intracellular hydrogen peroxide, the dismutation product of the superoxide radical, was demonstrated spectroscopically by formation of electron dense cerium-hydroperoxide precipitates. With electron energy loss spectroscopy (EELS) significant cerium deposits were detected in the mitochondria, the endoplasmatic reticulum, the cytosol and to a lesser extent on the plasma membrane of canine fibroblasts indicating multiple hydrogen peroxide production sites. Peroxides, therefore, possibly play an important part in the redox-sensitive pathway of TNF-alpha dependent NF-kappaB activation in canine skin. An adjunctive therapy with appropriate antioxidants modulating NF-kappaB overactivation in cutaneous inflammation in the dog is promising.  相似文献

Previous attempts have shown the potential of T cells in immunotherapy of cancer. Here, we report on the clinical activity of a bispecific antibody construct called blinatumomab, which has the potential to engage all cytotoxic T cells in patients for lysis of cancer cells. Doses as low as 0.005 milligrams per square meter per day in non-Hodgkin's lymphoma patients led to an elimination of target cells in blood. Partial and complete tumor regressions were first observed at a dose level of 0.015 milligrams, and all seven patients treated at a dose level of 0.06 milligrams experienced a tumor regression. Blinatumomab also led to clearance of tumor cells from bone marrow and liver. T cell-engaging antibodies appear to have therapeutic potential for the treatment of malignant diseases.  相似文献
