OBJECTIVE: To evaluate diagnostic utility of a commercially available immunoassay for assessing adequacy of passive transfer of immunity in neonatal calves. DESIGN: Prospective study. ANIMALS: 123 calves. PROCEDURE: Blood and serum samples were obtained from the calves prior to 2 weeks of age. The immunoassay was performed, along with refractometry and an 18% sodium sulfite turbidity test. Serum IgG concentration was determined with a radial immunodiffusion assay. Sensitivity and specificity of the immunoassay, refractometry, and the sodium sulfite test were calculated by comparing results with results of the radial immunodiffusion assay. RESULTS: Sensitivity and specificity of the blood IgG immunoassay were 0.93 and 0.88, respectively, compared with 1.00 and 0.53 for the sodium sulfite test. For refractometry, sensitivity and specificity were 0.71 and 0.83, respectively, when a serum total solids concentration of 5.2 g/dl was used as the cutoff between positive and negative test results. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that the immunoassay performs well in detecting calves with inadequate passive transfer of immunity.  相似文献

OBJECTIVE: To determine the effects of pasteurization of colostrum on serum lactoferrin concentration and neutrophil oxidative function by comparing values from calves given pasteurized (76 C, 15 minutes) colostrum versus calves given fresh frozen colostrum. ANIMALS: 8 Holstein bull calves were used to study the effects of pasteurization of colostrum on the absorption of lactoferrin and neutrophil oxidative burst. Three additional calves were used to study the effect of exogenous lactoferrin on neutrophil oxidative burst. METHODS: Calves were fed fresh frozen or heat pasteurized colostrum (76 C for 15 minutes) via esophageal feeder within 4 hours of birth. Neutrophils were isolated from whole blood samples. Neutrophil oxidative burst was induced by phorbol ester (300 ng/ml) stimulation of cells (1 X 10(6) cells) at 37 C. Serum lactoferrin concentrations were compared, using immunoblot analysis. Serum IgG concentrations were determined by radial immunoassay. Comparisons were made between the use of the 2 types of colostrum in calves by measuring subsequent serum IgG and lactoferrin concentrations and neutrophil superoxide production. RESULTS: Serum IgG and lactoferrin concentrations increased more in calves receiving fresh frozen colostrum. Neutrophil superoxide production was higher in neutrophils prepared from calves receiving fresh frozen colostrum. Colostral lactoferrin addition to neutrophil incubations resulted in increased oxidative burst. CONCLUSIONS AND CLINICAL RELEVANCE: Compared with calves given fresh frozen colostrum, calves given pasteurized colostrum had decreased serum IgG and lactoferrin concentrations and neutrophil superoxide production 24 hours after administration. These results suggest that pasteurizing bovine colostrum at 76 C for 15 minutes has substantial effects on passive transfer of proteins and neutrophil function.  相似文献

Passive transfer of colostral immunoglobulins has long been accepted as imperative to optimal calf health. Many factors, including timing of colostrum ingestion, the method and volume of colostrum administration, the immunoglobulin concentration of the colostrum ingested, and the age of the dam have been implicated in affecting the optimization of absorption. The practice of colostrum pooling, the breed and presence of the dam, and the presence of respiratory acidosis in the calf also may affect passive transfer. Various tests have been reported to accurately measure passive transfer status in neonatal calves. The radial immunodiffusion and the enzyme-linked immunosorbent assay (ELISA) are the only tests that directly measure serum IgG concentration. All other available tests including serum total solids by refractometry, sodium sulfite turbidity test, zinc sulfate turbidity test, serum gamma-glutamyl transferase activity, and whole blood glutaraldehyde gelation estimate serum IgG concentration based on concentration of total globulins or other proteins whose passive transfer is statistically associated with that of IgG. This paper presents a comprehensive review of the literature of passive transfer in calves including factors that affect passive transfer status, testing modalities, effects of failure of passive transfer on baseline mortality, consequences of failure of passive transfer, and some treatment options. Many previously accepted truisms regarding passive transfer in calves should be rejected based on the results of recent research.  相似文献

The objectives of this study were to compare glycerol and ethylene glycol at different concentrations as cryoprotectants and lycopene or cysteamine (with/without) as antioxidants in Tris extender for bull semen. Twenty‐four ejaculates were obtained from three bulls. Each ejaculate was split into four equal aliquots and diluted using both of the Tris extenders with glycerol (5% or 7%) or ethylene glycol (3% or 5%). After that, each extenders were split into three equal aliquots and added using both of the cysteamine 5 mm or lycopene 500 μg/ml, and control (without additives). The addition of 7% glycerol with cysteamine, 5% ethylene glycol with cysteamine and 3% ethylene glycol with cysteamine groups gave the lowest CASA motility than the other groups. However, 7% glycerol and 7% glycerol with lycopene resulted in a better rate of CASA progressive motility compared with that of other groups. Generally, all the lycopene groups signed better protective effects on acrosome and total morphology than the other groups. Glycerol 7% and 3% ethylene glycol with lycopene groups yielded to slight higher percentages of membrane integrity assessed by HOST than that of the other groups, but 7% glycerol with cysteamine and 3% ethylene glycol with cysteamine showed the worst percentages of membrane integrity. Glycerol 7% and 5% glycerol with lycopene gave rise to a higher value of VAP, VSL and VCL compared with that of the other groups. On the contrary, adding to 5% glycerol with cysteamine showed negative effect for VAP, VSL, VCL and ALH values. All cryoprotectant groups with lycopene decreased chromatin damage than the other groups. Ethylene glycol 3% led to lower non‐return rates of inseminated cows. However, this result was not considered to be statistically important.  相似文献

This paper provides baseline information on the involvement of women in small-scale coastal aquaculture such as the farming of slipper oyster, Crassostrea iredalei (Faustino 1933), and green mussel, Perna viridis (Linnaeus 1758). Data were gathered during a socio-economic survey of oyster and mussel farming in Western Visayas, Philippines conducted from July to December 1991.There were 175 respondents interviewed; 18.5% and 2.4% of oyster and mussel farmers were females. Fishing and related activities were the major source of livelihood for the majority of oyster and mussel farmers. In 1990, 79.4% and 69.8% of oyster and mussel farmers earned an annual income of less than P30 000 (1 USS P23). Based on the 1988 (latest data available) poverty threshold of P2654 (monthly) for Western Visayas, 80.4% and 71.7% of oyster and mussel farmers lived below poverty level in 1990. of 175 respondents. 79.3% and 48.2% of oyster and mussel farmers indicated that their household members participated in farming activities. More female household members were involved in oyster farming compared with mussel farming. Harvesting in mussel farming and both harvesting and marketing in oyster farming generated the widest participation among female household members.  相似文献

A flea larval bioassay was developed by an international team of scientists to monitor the susceptibility of fleas (Ctenocephalides felis) to imidacloprid (Advantage, Bayer HealthCare). The assay was validated using laboratory and field isolates of C. felis. Flea eggs representing different field isolates of C. felis were collected by veterinarians in the United States, United Kingdom, and Germany. Of the 972 flea isolates obtained during the 5-year study, 768 contained sufficient numbers of eggs to conduct the larval bioassay. Greater than 5% survival occurred for only six of the field isolates evaluated. Further evaluation and analysis of these isolates demonstrated that they did not differ significantly in their susceptibility to imidacloprid from the reference strains used to develop the assay. Collections of field flea isolates will continue in an attempt to detect and document any change in the susceptibility of field flea populations to imidacloprid.  相似文献

Abstract

AIM: To determine what, if any, changes have taken place in the optimum time, for undertaking faecal egg count reduction tests (FECRT) in sheep in New Zealand.

METHODS: A comparison was made between the numbers and types of nematode genera adequately represented for testing purposes (faecal nematode egg count (FEC) of >50 epg) in initial FECRT case submissions to a veterinary laboratory in New Zealand, during two 4-year periods, in 1992–1995 and 2006–2009.

RESULTS: Although there were some minor differences between them, the seasonal patterns of occurrence remained basically the same for all parasite genera in both datasets, with their individual peak periods of representation occurring during February to May in all instances. Not surprisingly, this period of maximum seasonal occurrence for each parasite genus also coincided with those months of the year when the greatest numbers of worm genera were adequately represented for faecal nematode egg count reduction (FECR) testing.

CONCLUSIONS: The results of this study indicate that the optimum time for conducting FECRT in sheep in New Zealand continues to be during the late summer-autumn months of February to May. However, they also reaffirmed that even during this optimal period there are still likely to be many occasions when relatively few nematode genera may be sufficiently well represented for satisfactory FECR testing. Accordingly, veterinary practitioners ought to be aware that, in order to obtain a complete picture of the resistance status of all worm genera on a particular property, such testing may need to be carried out on more than one occasion.  相似文献

Following the recent discovery of Libyostrongylus douglassii in ostriches in New Zealand, information relating to this nematode is reviewed. Amongst some of the topics considered are the parasite's distribution and prevalence, its life cycle and survival, and its diagnosis and control. Of particular interest from a New Zealand perspective is the potential for the parasite to infect other ratites, especially the kiwi. While the current evidence suggests that this may be unlikely to occur, the possibility that it might be able to do so cannot be ruled out entirely.  相似文献
