One-year-old immature green turtles (500–900 g) were kept at 25°C in recirculating seawater. When deprived of food for 108 h and then offered floating trout pellets, they consumed a satiation meal (S g) described by the equation S = 0.025 W0.96 where W is wet body weight in g. When food was offered at different deprivation times after satiation, maximum meals were taken after 60–72 h and corresponded to 2.5% body weight. Longer deprivation (108 h) led to reduced intake of 1.9–2% body weight. Gastric emptying was measured using X-radiography or chromic oxide to label the faeces. Complete stomach emptying required 110 ± 11 h S.D. and maximum appetite occurred when the stomach was estimated to be 80–95% empty. The average time for a meal to be voided from the alimentary tract was 176 ± 16 h S.D. Diets containing 40–50% protein and 4.2–5 kcal/gram were assimilated with apparent efficiencies of 76 ± 6% S.D. and 86 ± 6% for energy and protein nitrogen, respectively. Effective management of potato cyst nematodes (PCNs) requires simple, rapid and accurate identification and quantification of field populations. Soil samples from a survey of 484 fields in potato rotations in England and Wales were used to compare the identification and quantification of PCNs using IEF, PCR, ELISA and bait plant tests. The cyst counts and bait plant test revealed that 64.3% of field samples contained PCNs. Bait plant tests increased the detection rate of PCNs in field samples by 4–6.4%. This means that some infestations are cryptic and would not normally be detected by standard counts. IEF, PCR and ELISA methods distinguished between Globodera rostochiensis and G pallida and were able to register mixed populations; however they were not in full agreement. All methods suggested that G pallida is the dominant species in the field samples tested. The PCR results indicated that 66% of field samples contained pure G pallida, 8% contained pure G rostochiensis and 26% contained mixtures of the two species. ABSTRACT Seven hundred forty-nine isolates of Phytophthora spp. were obtained from irrigation canals in eastern Washington State during the 1992 to 1995 and 1999 growing seasons. Isolates were retrieved using pear baiting techniques. All isolates were pathogenic to pear and were present in irrigation water beginning early in fruit development. Over the course of the 5 year study, 10 and 5% of isolates were identified as P. cactorum and P. citricola, respectively, using morphological criteria. The remaining isolates could not be identified using morphological criteria. Colony morphology of these isolates was characterized during all years of the study. In 1999, more detailed studies utilizing polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) analysis of entire internal transcribed spacer (ITS) regions (ITS1, 5.8S, and ITS2) of ribosomal DNA for 180 isolates, and sequence analysis of ITS2 for 50 isolates, were used to investigate genetic variation and phylogenetic relationships among isolates. Isolates were divided into 12 groups based on their growth type on corn meal agar. Restriction digestion of the entire ITS region with three enzymes revealed 11 restriction digestion patterns among 180 isolates. PCR-RFLP and sequence data were obtained for 12 reference Phytophthora spp. (two species in each of Waterhouse's six morphological groups). Phylogenetic analysis of ITS2 regions revealed nine clades, each with strong bootstrap support. Molecular analyses revealed 23 isolates that were in the P. gonapodyides clade, 9 in the P. parasitica clade, 1 in the P. cactorum clade, 7 in the P. citricola/capsici clade, and 4 in the P. cambivora/pseudotsugae clade. The three isolates comprising clade 5 were significantly distinct from all other Phytophthora spp. in the databases and may represent a new Phytophthora sp. Colony morphology was not consistently correlated to PCR-RFLP pattern or ITS2 phylogeny, suggesting that the former criterion is insufficient for species identification. The results of this study indicate that at least nine phylogenetically distinct taxa of Phytophthora pathogenic to pear are present in irrigation water in North Central Washington.  相似文献

A single dose of 5 x 10(8) bacilli of Pseudomonas pseudomallei by intratracheal injection resulted in acute (21 cases) or chronic (19 cases) melioidosis in 40 of 48 pigs. Fifteen (10 acute and 5 chronic) had been immunosuppressed by cyclophosphamide before inoculation. The major clinical signs were initial fever, marked neutrophilia and, in the acute cases, respiratory distress. There were no signs of the nasal and ocular discharge, paresis or diarrhoea seen in acute cases in south-east Asia. The cyclophosphamide treatment caused a significant decrease in the neutrophil count by 7 d after inoculation in all 15 immunosuppressed pigs, and all were culture positive at necropsy. Eight of the 33 non-treated pigs were culture negative at necropsy. Pigs overcoming the initial phase of infection had more abscess-like nodules that were bacteriologically sterile at necropsy than the pigs with acute cases of melioidosis. P. pseudomallei was isolated predominantly from the spleen, lungs and the injection site. Although only one strain was used in this study, it is likely that Australian strains of P. pseudomallei are not as virulent as the south-east Asian isolates.  相似文献

In south-western Australia, plantations of Eucalyptus globulus are being established on land that has previously been used for conventional agriculture. Sustaining the productivity of these tree plantations in second and subsequent rotations will depend partly on maintenance of soil fertility, especially soil nitrogen (N) supply rates. We compared soil N status and supply rates between adjacent pasture and 6-11 year old first-rotation eucalypt plantations at 31 paired sites in south-western Australia. Total soil N varied widely among sites (0.07-0.68% in the fraction <2 mm of the 0-10 cm soil layer), but concentrations averaged over all sites did not differ between land-use types. However, measurements of the indices of mineralization (mineral N produced during incubation of intact cores), potentially available N (from short-term anaerobic incubation) and model-predicted mineralization rates during 28-day aerobic incubations were generally lower in afforested soils than in pasture soils. This finding was supported by in situ field estimates of N mineralization over a full year at two contrasting paired pasture-plantation sites. At each site there was a marked reduction (2-3-fold) in net annual mineral N flux rates in soils under eucalypt plantations. Reduced N mineralization associated with tree plantations was due to both changes in soil organic matter quality and the generally lower soil moisture content under trees in comparison with pasture. These results suggest that N supply rates of pasture soils are likely to decline when the land is planted to successive crops of eucalypts. Eucalypt plantation managers will need to take account of this and implement management strategies to maintain adequate N nutrition to sustain tree growth in future rotations.  相似文献

Two genetic sources of variation influence the percentage of sickle cell hemoglobin found in heterozygotes. Growth and soil N supply in young Eucalyptus tereticornis stands at two sites in Kerala, India, were examined in response to cover cropping with three legume species (Pueraria phaseoloides, Stylosanthes hamata, and Mucuna bracteata). The effects of legume residues on soil N supply were investigated in a long-term (392 day) laboratory incubation using leaching micro-lysimeters. Residues from the eucalypt and legume species had different rates of net N release during the laboratory incubation. Net N release was significantly related to residue N concentration (R2 =0.94), the C:N ratio (R2 =0.91), the lignin:N ratio (R2 =0.83), and the (lignin + soluble polyphenol):N ratio (R2 =0.95). Nitrogen release rates declined in the order Mucuna > Pueraria > Eucalyptus > Stylosanthes. There was no net N release from Stylosanthes residues during the 392-day laboratory incubation, whereas Mucuna and Pueraria released N throughout the incubation period. Net N release from mixtures of legume and eucalypt residues was not additive in the early phase of the incubation, probably because eucalypt residues initially immobilized a portion of the legume-derived N in addition to the soil-derived N. Legume establishment had no significant effect on tree growth at one site (Kayampoovam), but resulted in depressed tree growth at the lower rainfall site (Punnala) at 18 months. There were no significant treatment effects on growth at Punnala after that time. Cover cropping with legumes during the early phase of forest plantation growth may be a useful mechanism to enhance soil N supply and optimize the synchrony between N supply and tree N uptake. Although these effects did not translate into improved plantation growth in the 3 years of this study, improved soil organic matter and N fertility may help ensure sustainable productivity over several rotations in the future. This study showed that the effect of legumes on N dynamics varies markedly with legume species. This, together with other factors (e.g. competition with trees, N fixation capacity), will be important in selecting suitable species for cover cropping in forest plantations.  相似文献
