In Portugal, the oak pinhole borer Platypus cylindrus and its mycobiota have been associated with cork oak (Quercus suber) death, but no knowledge exists regarding the associated bacterial community. However, it is known that some bacteria are important for ambrosia beetle symbiosis and play a role in oak tree health. To explore the bacteria associated with this beetle and its host, with the ultimate goal of highlighting potential roles in oak decline, this study used a culture-dependent approach for strain isolation and phylogenetic identification using 16S rRNA gene sequencing and multilocus sequence analysis (MLSA). The bored galleries of different cork oak trees from a cork stand in Alentejo, together with the body and mycangia of adult beetles, were investigated. The samples revealed a diverse community comprising 500 isolates with 64 distinct types of bacterial colonies. Sixty-eight strains were selected for sequencing and used for phylogenetic analysis, 40 from wood galleries and 28 from beetles. Thirty-two genera of bacteria were identified, 18 of which were described for the first time within oak–beetle interactions. Major taxonomic groups were Actinobacteria in beetles and Enterobacterales in wood galleries. Although specific oak bacterial pathogens were not detected, a group of distinct strains detected in wood galleries, potentially belonging to a new Pectobacteriaceae species, were able to produce mild symptoms on cork oak plantlets. This study reports for the first time the biodiversity of culturable bacteria associated with the Q. suber–P. cylindrus interaction, their relevance to both organisms and the possible contribution to oak decline.  相似文献

Leaves of nine green manure (GM) species were surface applied to a tropical volcanic-ash soil at a rate of 100 kg N ha-1 in order to evaluate their N-fertilizer value in a glasshouse experiment. GM treatments were compared to urea at two rates, 50 kg N ha-1 (FN50) and 100 kg N ha-1 (FN100), and to a control with no fertilizer application (FN0). Two weeks after treatment application, upland rice seedlings were sown in order to conduct N uptake studies. Soil volumetric moisture content was maintained close to 50%. In general, soil showed an initial increase in inorganic N followed by a rapid decline with time. After 2 weeks of evaluation FN100, FN50 and leaves of Mucuna pruriens var. Tlaltizapan and Indigofera constricta presented higher values of inorganic N (157-109 mg N kg-1 soil); while, FN0 and leaves of Mucuna deerengianum, Cratylia argentea and Calliandra calothyrsus presented lower values (75-89 mg N kg-1 soil). N recovery by rice, at 20 weeks after planting, was highest for FN100 (59.9%) followed by Canavalia brasiliensis (54.6%), Calliandra calothyrsus (47.4%) and M. pruriens var. IITA-Benin (32.4%); while, M. pruriens var. Tlaltizapan, FN50, Tithonia diversifolia and I. constricta presented lower N uptake (13-20%). Significant relationships were found between some quality parameters of GM evaluated (i.e. total N, fibers, lignin and polyphenol content), soil N availability and rice N uptake. These results suggest that GM that decomposed and released N slowly resulted in high N uptake when they were used at pre-sowing in a tropical volcanic-ash soil.  相似文献

From the genome of a Japanese field isolate of the rice blast fungus, Magnaporthe oryzae, we newly identified Inago1 and Inago2 LTR retrotransposons. Both elements were found to be Ty3/gypsy-like elements whose copies were dispersed within the genome of Magnaporthe spp. isolates infecting rice and other monocot plants. Southern hybridization patterns of nine re-isolates derived from conidia of the strain Ina168 produced after a methyl viologen treatment were not changed, indicating that the insertion pattern of Inago elements is relatively stable.  相似文献

Azuche Hymenachne amplexicaulis (Rudge) Nees is a C3 grass native to tropical flooded areas in South and Central America with 2n = 2x = 24 chromosomes. During 2002 and 2003, the Graduate College for Agricultural Sciences (CP) and the Mexican Institute for Forestry, Agriculture and Animal Production Research (INIFAP) organized an expedition to collect the genetic diversity of this species within tropical Mexico, in order to preserve and study it under nursery conditions. The collection was performed from Puerto Vallarta to Tapachula on the Pacific and from Palizada, Campeche to Veracruz on the Gulf of Mexico coast, and also crossing sections through the Tehuantepec isthmus and through the Sierra Madre de Chiapas. Ninety sites were sampled, on each, 25 tillers were collected, tagged and stored in a commercial soil mix until establishment of individual plants within 6 l pots. The largest Azuche prairie observed had a 10 ha surface, with the grass limited to small surfaces of up to 0.25 ha. It was not found on places with salty water, even seasonally. The Pacific coastal side is less abundant in niches for this species. Producers’ experience indicated that this grass is beneficial for milk production compared with other forage grasses, mainly C4, growing in flooded areas. Initial studies of genetic diversity showed wide morphological variation for sprouting ability, leaf width and length among samples. Azuche diversity showed good potential production attributes for farm and wild animals, under flooding tropical savanna conditions.  相似文献

Six entomopathogenic fungal isolates were tested under controlled conditions, as biological control agents against Monochamus galloprovincialis (Coleoptera: Cerambycidae) (Olivier), the vector of the pine wood nematode Bursaphelenchus xylophilus (Nematoda: Parasitaphelenchidae) (Steiner and Buhrer). The fungi Beauveria bassiana (Bals.-Criv.) Vuill, Metarhizium anisopliae (Metchnikoff) Sorokin, Metarhizium sp. and Fusarium sp. were tested against larvae and adults of the insect, by comparing two exposure techniques: continuous contact and spraying. Regarding the larvae, only Fusarium sp. and Metarhizium sp. killed more than 40% of the individuals, while B. bassiana and M. anisopliae were found to kill 50% of the adults (LT50) within five days. Exposure to B. bassiana reduced adult longevity by 19 days, when compared to untreated beetles, and had a direct efficacy of 46%. Continuous contact with the fungi proved to be more effective than spraying. A larval disinfection protocol was developed to eliminate other fungal isolates from the wood-collected tested individuals. The implications of the results and possible applications of selected strains as bio-control agents against M. galloprovincialis are discussed.  相似文献

In 2000 Guarnera et al. proposed using ELISA in canine faeces collected from the ground to detect dogs infected with Echinococcus granulosus, thus determining sheep farms with active transmission. The objective was to evaluate the prevalence of E. granulosus infection in sheep farms of the Patagonia. Sheep farms were randomly selected in the Provinces of Río Negro, Chubut, Neuquén, Santa Cruz and Tierra del Fuego (areas with control programs) and La Pampa (comparison area). From one to three samples of fecal matter were obtained for each sheep farm, which were processed by means of copro-ELISA test with confirmation of positive samples by copro-Western blot. A total of 1042 samples were obtained from 352 sheep farms, 26 (7.3%) proving positive. Of these 5 (6.3%) were from La Pampa, 9 (13.8%) from Neuquén, 4 (4.7%) from Río Negro, 2 (2.9%) from Chubut, 1 (5.9%) from Santa Cruz and 5 (13.9%) from Tierra del Fuego. The identification of parasitized dogs is an essential activity upon which rests the strategy of control and surveillance. Arecoline tests or coproantigen test with fecal matter obtained directly from the dog contribute information on individual prevalence, while the use of coproantigens detected in ground-collected samples transfers the dog unit of observation to units of greater epidemiological value. In the present experience, the technique employed seems promising for its application in systems of epidemiological surveillance of cystic echinococcosis and in drawing a baseline on which to measure the progress of control programs in the Argentine Patagonia in subsequent years.  相似文献

Cystic echinococcosis (CE), caused by Echinococcus granulosus, is a chronic and debilitating zoonotic larval cestode infection in humans, which is principally transmitted between dogs and domestic livestock, particularly sheep. Human CE occurs in almost all pastoral communities and rangeland areas of the underdeveloped and developed world. Control programmes against CE have been implemented in several endemic countries to reduce or eliminate the disease. New Zealand and Tasmania are examples of some of the first programmes to be undertaken (in insular territories) and which were very successful in the elimination of CE. The advent and proven effectiveness of praziquantel, plus the experience of insular models, produced high expectations for rapid advances in a second generation of control programmes undertaken in continental areas (Argentina, Uruguay and Chile). Nevertheless, only moderate gains in CE control have been made and the impact on prevalence among humans has been slight. A major impediment to the adoption of procedures that were successful in New Zealand and Tasmania has been the requirement to administer praziquantel to dogs in rural areas eight times per year over numerous years. In addition, there have been clear technological improvements made in the diagnosis of canine echinococcosis for surveillance, the genetic characterization of parasite strains and in vaccination against CE infection in livestock. In order to establish new paradigms and appropriate combinations of control strategies, we have carried out a review and discussion of the available control tools and control models. Control strategies must be suitable and sustainable to benefit the Echinococcosis–endemic areas primarily, which at the same time are the poorest regions of the world.  相似文献
