Tilmicosin(TM), a macrolide antibiotic and active ingredient in formulated Micotil 300TM (Eli Lilly and Co., Indianapolis, IN, USA), is the active ingredient in a formulated animal product used for the treatment of respiratory tract infections in cattle. Owing to the concern of governmental regulatory agencies over the possibility of an accidental injection of the antibiotic to a livestock handler, the cardiovascular effects of sub lethal doses of TM were evaluated in conscious mixed-breed dogs. Left ventricular function, systemic arterial blood pressure, and heart rate (HR) responses to TM alone and in combination with propranolol(P) or dobutamine HCl(DOB) were evaluated. Dogs were instrumented with indwelling micromanometers implanted in the left ventricular chamber and in the thoracic aorta. Cardiovascular variables were recorded, and the peak value of the first derivative of left ventricular pressure (dp/dt(max)) was used as an index of left ventricular inotropic state. Six treatments were randomly assigned to each of the six dogs using a Latin square design. The six treatments were vehicle, TM alone (2.5 mg/kg of body weight), TM immediately followed by P. and TM immediately followed by 1 of 3 dosages of DOB infused for ?45 min. Additionally, doses of TM alone (0.25, 1.0, 2.5, and 5.0 mg/kg) were administered to complete a dose-response curve. TM caused dose dependent decreases in (dp/dt(max)) and aortic pulse pressure. HR increased dose-dependently. Left ventricular end-diastolic pressure increased at the 2.5 and 5.0 mg/kg dosages. Left ventricular systolic pressure was reduced dose-dependently at the 2.5 and 5.0 mg/kg dosages. Treatment with P exacerbated the negative inotropic effect and the decrease in left ventricular systolic pressure, but did not attenuate the tachycardia associated with TM treatment. DOB attenuated the changes in ventricular inotropic state in a dose-dependent manner. DOB infusion also restored left ventricular systolic pressure at dosages of 3 or 10 μg/min/kg. Our data indicate that toxic doses of TM may have a negative inotropic effect in conscious dogs. HR increased in a dose-dependent manner and was not the result of β1-receptor stimulation. DOB reversed some, but not all, of the effects caused by TM administration.  相似文献

A modification of the intravenous regional anaesthesia technique was used to assess the peripheral antinociceptive effect of remoxipride, clonidine and fentanyl. Drugs administered intravenously via peripheral catheters were restricted to the distal limb and nociceptive threshold test site by prior inflation of a tourniquet proximal to both the catheter and a threshold-testing device. Lignocaine (1 mg/kg) induced peripheral antinociception during tourniquet inflation. Clonidine (6 μg/kg) only induced significant elevations in thresholds after tourniquet deflation. A low dose of remoxipride (2 mg/kg), which had no systemic antinociceptive effect, produced antinociception after its restriction to the periphery. Peripheral administration of saline and tourniquet-induced restriction of blood flow to the distal limb did not alter threshold values. Peripheral administration of fentanyl was used to test a further modification of the injection protocol designed to reduce the incidence of leakage into the systemic circulation. Fentanyl administration (11.2 μg/kg) failed to elicit an increase in thresholds when it was restricted to the distal limb test site. The contribution of a peripheral mechanism to the antinociception induced by systemic administration of a higher remoxipride dose (7.5 mg/kg) was investigated using an inflated tourniquet to exclude remoxipride from the periphery. Exclusion of remoxipride from the periphery reduced its antinociceptive effect, i.e. threshold values were lower than if remoxipride was allowed free access to the limb prior to tourniquet inflation.
The technique described here was effective in demonstrating that the increase in noninflammatory nociceptive thresholds seen with clonidine and fentanyl is not peripherally mediated whilst that seen with remoxipride has a peripheral component.  相似文献

SUMMARY Chronic fluoride toxicosis caused lameness, dental lesions and illthrift in an extensive beef cattle herd in northern Australia. Up to 15% of the herd was lame and the disease forced the culling of large numbers of cows. The source of fluoride was fertiliser-grade monoammonium and diammonlum phosphate fed as part of a mineral supplement. Large quantities of mineral supplement were provided to the cattle because lameness was attributed to phosphorus deficiency, which is endemic in the area. Most lameness developed in the late dry season in the post-lactation phase. Severe lameness was caused by fractured pedal bones.  相似文献

The degradation of imazapyr, flumetsulam and thifensulfuron applied at 500.40 and 30 g active ingredient (a.i.) ha-1, respectively, to silt loam soil was studied under laboratory and field conditions. Herbicide residues were analysed by a lentil ( Lens culinarits L.) bioassay. Results showed that temperature had a significant effect on herbicide degradation, whereas the impact of soil organic matter ami pH were less well defined. Half-lives for imazapyr, flumetsulam and thifensulfuron in soil samples from the 0-5 cm layer (6.4% organic carbon) at 15 °C were 125, 88 and 5.4 days, respectively, and 69, 30 and 3.9 days at 30°C. In soil sampled from the 15-20 cm layer (3.5% organic carbon) half-lives were 155. 70 and 6.4 days, respectively, at 15 °C and 77, 24 and 4.8 days at 30 °C, A field experiment investigated the degradation and teaching of each herbicide under two precipitation regimes [natural precipitation (208 mm), and natural precipitation plus 75 mm irrigation (283 mm) over 4 months to a soil depth of 25 cm. Thifensulfuron degraded rapidly, whereas residues of flumetsulam and imazapyr leached below 25 cm in both the low-and high-precipitasion treatments after 4 months. Significant imazapyr residues were still present in the soil to 25 cm depth after 3 months, A multi-component model for herbicide dissipation was developed and evaluated using data from the laboratory and field experiments.  相似文献
