The aim of this study was to compare on an objective basis the results obtained during five classical swine fever (CSF) ring tests conducted in Germany between 1999 and 2003. A novel and simple statistical approach used in behavioural sciences was used. For each ring test, the regional laboratories received a panel of five lyophilized pig sera. The panel contained CSF virus positive and negative samples. The final task of the laboratory was to ascertain if a serum sample was positive for CSF or not. Some sera were very easy to diagnose as CSF positive while some sera had border line values and proved to be challenging. Depending on the degree of difficulty the sera were divided into five categories. The evaluation of the ring test results was performed using a scoring system based on a score from ?3 to +3 which takes into consideration the degree of difficulty to produce a correct diagnosis. To compare the results between different laboratories and/or between different ring tests more easily the total score of one laboratory was expressed in percentage. The final analysis of the data showed that the CSF diagnostic quality improved continuously.  相似文献

Eight groups of altogether 25 goats without neutralizing antibodies against BVD virus, were inoculated either intranasally or intranasally and subcutaneously with two different BVD virus isolates during different stages of gestation. In all 18 goats inoculated within the first 78 days of gestation an abortion and foetal death rate of approximately 100% occurred. Only one goat gave birth to a clinically healthy kid. The other seven goats which were inoculated after the 78th day of gestation showed also a high foetal death rate. Only two of them gave birth to clinically healthy kids. Neutralizing antibodies against BVD virus could be detected in blood samples drawn from 14 kids born at normal term including stillborn and non-viable offsprings. BVD virus was reisolated from different organs taken from seven foetuses. It was not possible to isolate BVD virus from any of the normal offsprings.  相似文献

The Friedrich-Loeffler-Institut, founded in 1910 by Friedrich Loeffler, the discoverer of the first animal virus, foot-and-mouth disease virus, is the oldest virological research facility in the world. Beyond viruses, its area of competence has significantly expanded since its foundation and now also covers bacterial, parasitic and prion diseases of livestock, poultry and aquatic animals. Presently located at four sites within Germany (Insel Riems, Jena,Tübingen,Wusterhausen) the tasks of the institute as delineated in the Animal Disease Act encompass research on infectious animal diseases including zoonoses, import/export examinations, epidemiological studies in case of outbreaks of notifiable animal diseases, acting as reference laboratory for notifiable animal diseases and nationwide quality management of diagnosis of notifiable animal diseases. It is obliged to publish and maintain up-to-date diagnostic regimes for notifiable animal diseases, and it publishes a yearly report on animal health in Germany. With the increasing importance of infectious diseases of animals, in particular those potentially harmful to man (zoonoses), the Friedrich-Loeffler-Institut will be moving into new facilities including laboratories and animal facilities up to the highest biosafety level at its main site Insel Riems on the occasion of its 100th anniversary.  相似文献

Dictyocaulus viviparus cause severe lung infections and are endemic in some areas of temperate and tropical countries. Treatment is based on strategic nematode control programs using potent compounds with no reports of drug failure. Macrocyclic lactones (MLs) are available at different concentrations and combinations and have being used heavily by producers. The objective of this work was to determine the efficacy of the MLs ivermectin, moxidectin, doramectin, and abamectin, and the combination ivermectin plus abamectin in naturally infected calves (n=70). Initial infection was determined by necropsy of tracer animals. Faecal larval counts determined that none of the compounds used was able to eliminate D. viviparus for up to 28 days after treatment. The "Area Nova" strain was isolated for future work.  相似文献

The European Commission intends to change its animal disease control strategy following the slogan "Prevention is better than cure". Vaccination and diagnostics should play a major role in emergency situations. A policy paper regarding the use of vaccines has been discussed with all Member States and the main aspects were evaluated in a questionnaire. In principle, the majority of Member States are in favour of a future strategy in which vaccination is replacing culling. However, questions regarding the pathogen freedom and the trade of vaccinated animals and animal products from vaccinated animals still remain open.  相似文献

The aim of the study was to evaluate the diagnostic quality of the real-time RT-PCR assay described by Hoffmann et al. [Hoffmann, B., Beer, M., Schelp, C., Schirrmeier, H., Depner, K., 2005. Validation of a real-time RT-PCR assay for sensitive and specific detection of classical swine fever. J. Virol. Methods 130, 36-44] for the routine intra vitam diagnosis of classical swine fever (CSF). We compared the assay with conventional diagnostic methods by using defined diagnostic material from an animal experiment with pigs showing different clinical forms of CSF. Compared to virus isolation and antigen ELISA an enhanced sensitivity of the real-time RT-PCR could be shown. We were able to detect all infected pigs regardless of the clinical course of CSF. CSF infection was detected already during the incubation period, during the entire clinical phase as well as at the beginning of convalescence when the first antibodies were detected and no virus could be isolated any more. In most cases, positive PCR results were obtained 2 days earlier than with virus isolation and 2-4 days earlier than with the antigen ELISA.  相似文献

The aim of the studies was to fathom the duration and the role of maternal immunity for Aujeszky's disease (AD) and classical swine fever (CSF) in wild boar offspring. In one experiment, two wild boar sows were infected with a low pathogenic pseudorabies virus (PRV) in 1999. A total of 51 offspring was born between 1999 and 2002 and was monitored for PRV maternal antibodies. In a second experiment, the maternal immunity for CSF was analysed. Therefore, a sow was orally vaccinated against CSF using vaccine baits containing the live-attenuated C-strain vaccine. The vaccination took place in January 1999. The sow gave birth to four piglets in 2001 and to two piglets in 2002. With respect to maternal immunity for AD, some piglets reacted positive in the ELISA up to 27-week post-partum while in the neutralization test antibodies were detected up to 15-week post-partum. The calculated half-life of neutralizing antibodies was 21 days. Regarding CSF, the neutralization titres of maternal antibodies dropped continuously reaching values of < or =10 ND50 20-week post-partum. After the 12th week post-partum, most of the sera reacted negative in the ELISA. However, after the third month, low levels of neutralization titres were still detectable. The results are discussed with respect to the epidemiology and control of both diseases in wild boar populations.  相似文献
