The polypyrimidine tract binding protein (PTB) is a 58-kilodalton RNA binding protein involved in multiple aspects of messenger RNA metabolism, including the repression of alternative exons. We have determined the solution structures of the four RNA binding domains (RBDs) of PTB, each bound to a CUCUCU oligonucleotide. Each RBD binds RNA with a different binding specificity. RBD3 and RBD4 interact, resulting in an antiparallel orientation of their bound RNAs. Thus, PTB will induce RNA looping when bound to two separated pyrimidine tracts within the same RNA. This leads to structural models for how PTB functions as an alternative-splicing repressor. 相似文献

It has been proposed that hydrogen produced from basalt-ground-water interactions may serve as an energy source that supports the existence of microorganisms in the deep subsurface on Earth and possibly on other planets. However, experiments demonstrated that hydrogen is not produced from basalt at an environmentally relevant, alkaline pH. Small amounts of hydrogen were produced at a lower pH in laboratory incubations, but even this hydrogen production was transitory. Furthermore, geochemical considerations suggest that previously reported rates of hydrogen production cannot be sustained over geologically significant time frames. These findings indicate that hydrogen production from basalt-ground-water interactions may not support microbial metabolism in the subsurface. 相似文献

In the Bay of Saint-Brieuc, a 3,110-ha tidal bay in NW France, English Channel, the cockle Cerastoderma edule has been collected by traditional fishing methods for many decades without any evaluation or management of this resource
taking place. Since 2001, the National Natural Reserve of the bay of Saint-Brieuc has carried out an evaluation of the stock
and a mapping of the cockle fishing grounds each year. Analysis of the spatial structure of the population is approached by
cartography through interpolation of the data using the kriging method. The recruitment zones are geographically quite well
defined and located in areas limited to the mean-tide zone. The distribution of the population was affected by passive displacement
of juvenile cockles. Evaluation using the matrix of individual numbers for each age group found the inter-annual mortality
rates to be about 60%. Somatic production was estimated and expressed in ash-free dry weight. The average annual production
ranged from 7.4 to 14.5 g/m2. In the autumn of 2006, the minimum legal fishing size changed from 30 to 27 mm (corresponding
to individuals aged about 2.5 years). The model developed shows that this change has led to a doubling of the fishable stock. 相似文献

A nation-wide survey was carried out in 370 randomly chosen French commercial broiler chicken flocks from October 2005 to September 2006 to determine Salmonella spp. prevalence and to identify risk factors for contamination, at the end of the rearing period. The Salmonella status of the flocks was assessed from five faecal samples (litter swabs) analysed by classical bacteriological methods. A flock with at least one contaminated sample was classified as a Salmonella-positive flock. The apparent prevalence of Salmonella was 8.6% (95% CI: 5.7, 11.5%). The most prevalent serovar was S. hadar followed by S. anatum and S. mbandaka. Logistic regression methods were used to analyse the associations between husbandry practices, farm characteristics, general hygiene and the Salmonella status of the sample. The risk for Salmonella contamination of the flock at the end of the rearing period increased when neighbours helped in the placement of day-old chicks. On the contrary, the risk decreased when mobile equipment was dismantled before cleaning and disinfection, when the farm had a specific container for dead-bird disposal and when acetic acid was added to the drinking water. 相似文献

1. The aims of this study were (i) to evaluate the prevalence of welfare-related cutaneous lesions in turkeys at the end of the rearing period and (ii) to study the correlations between lesions. 2. The percentage of lesions in each flock was determined by observation at the slaughterhouse. Sixty flocks were investigated in 13 slaughterhouses between April and July 2006. 3. All flocks showed a high proportion of turkeys with footpad lesions, with 40·7% of turkey flocks having severe footpad dermatitis. A significant number of feet had swelling of the footpad and deviated toes (60·0% and 21·4% respectively). Arthritis was observed in 25·4% of the carcases and an average of 30·1% had breast buttons on the keel. 4. Positive correlations were found between evidence of feather pecking and arthritis, toe deviations and swelling of the footpad. Deviated toes, swelling of the feet and feather pecking were negatively linked to the most severe scratches (P?0·001). 5. A method is proposed to assess turkey welfare at the slaughterhouse based on several criteria: footpad lesions, arthritis, severe scratches, deviated toes, swelling of the footpad and breast buttons. 相似文献

The chief aims of this paper were the following: (i) to describe the demography and genetic structure in two divergent selected lines for total fleece weight (TFW) of French Angora rabbits with overlapping generations; (ii) to describe the effects of inbreeding during an experiment of divergent selection. A study of longevity with the survival kit showed that there was no significant difference in the risk of death or culling between the low line (LL) and high line (HL). A significant effect of inbreeding (p < 0.05) was observed with a 30% higher risk factor in the highest class of inbreeding coefficient compared with the other classes. The means of generation interval were 562 and 601 days in LL and HL, respectively. The numbers of generations for LL and HL were 3.90 and 3.64, respectively. Generation intervals decreased significantly from 1995 to 2000 (p < 0.05). The number of daughters in HL was very variable. The number of animals per generation was higher in HL than in LL. Each buck left nearly three daughters to the next generation (2.52 in LL, 3.24 in HL). In both lines, the effective number of ancestor genomes still present in the genetic pool of the generation was around eight from the reference population of 1995 to that of 2001. Inbreeding in HL was always higher than in LL. The effect of inbreeding was also significant (p < 0.05) on TFW and live weight. The animals with the lowest inbreeding category produced a higher TFW (p < 0.05) than the others. The observed selection differentials were lower than that expected owing to the breeding animal management rules in order to control inbreeding increase. 相似文献

The relationship between anchovy ( Engraulis encrasicolus ) recruitment in the Bay of Biscay and environmental variables during their planktonic phase (March to July) was investigated from 1986 to 1997. Meteorological variables (wind and temperature) are forcing effects on the sea, but they are not thought to be processes that govern larval survival directly. Food-web dynamics are believed to be more closely linked to larval survival and are related to the physical vertical water column structure. Therefore, we used a three-dimensional (3D) hydrodynamic model to characterize three major physical mesoscale processes affecting vertical structure in south-east Biscay: stratification, upwelling and river plume extent. Indices were estimated from the model outputs to characterize and quantify the space/time evolution of these structures during the period March to July. A multiple linear regression analysis was then used to analyse hierarchy in the explanatory power of the physical indices. Coastal upwelling and shelf stratification breakdown indices were the most significant explanatory variables, with positive and negative effect on recruitment, respectively. A model with these two indices explains 75% of the recruitment variability of anchovy observed in the period 1987–96. 相似文献
