Wolbachia (Rickettsiales: Alphaproteobacteria) infections induce abnormalities in the reproductive system and affect various biological traits of the host insects. The density of Wolbachia is one of the major parameters that influence induced phenotypes and interactions with the hosts. Wolbachia occurs naturally in populations of the leafhopper Yamatotettix flavovittatus Matsumura (Hemiptera: Cicadellidae), which transmits phytoplasma that cause white leaf disease in sugarcane. However, the quantity and dynamics of Wolbachia in this leafhopper are not well understood. In the current study, we estimated the number of Wolbachia by absolute quantification of the copy number of wsp, which encodes the outer surface protein, using real-time quantitative polymerase chain reaction (PCR). This investigation was performed using natural populations and laboratory colonies from three lineages of leafhoppers (designated as UD, KK, and SK). There was no significant difference in the number of wsp copies in most of field-collected adults. During the immature developmental stages, there were differences in the dynamics of Wolbachia infection between the UD lineage and the other two lineages. However, the number of wsp copies increased in the early instar and plateaued in the later nymphal instars. Sex had no influence on the number of Wolbachia within the same lineages. The number of Wolbachia was relatively constant during the adult stage in the UD lineage but fluctuated in the other two lineages. In conclusion, the present data provide a framework for exploring the relationship between Wolbachia and the leafhopper and could facilitate future research into management strategies using Wolbachia.  相似文献

When fungal germlings, after forming haustoria of Blumeria graminis f. sp. hordei (B. graminis), were removed from the surfaces of barley coleoptiles by cellulose acetate, followed by challenge inoculation with the non-pathogen Erysiphe pisi, they infected the nonhost barley coleoptile cells. This phenomenon was not observed on the coleoptile surface when the fungal germlings of B. graminis were removed before the formation of haustoria. Also, when the surface was inoculated with the pathogen of barley B. graminis as a challenger, after removing the fungal germlings of inducer post haustorial formation, the penetration efficiency of the fungi increased significantly compared with that of the control. Furthermore, when we extracted the crude-susceptibility inducing factor (suppressor) from coleoptiles before and after the formation of haustoria of B. graminis, suppressor activity against infection with E. pisi was observed only in the extract of barley coleoptiles that included haustoria of B. graminis about 18 h or later after inoculation. Surprisingly, however, the extract did not increase the penetration efficiency of B. graminis significantly. Thus, we hypothesize that the suppressor extracted from barley coleoptiles in which B. graminis had formed haustoria has no effect on increasing the penetration efficiency of the primary infection from the appressorium of B. graminis but has an effect on the infection with non-pathogen E. pisi.  相似文献

As soil engineers, earthworms and ants play major roles in soil functioning, especially in modifying soil physical and chemical properties. This study was conducted in a very constraining environment, i.e., paddy fields which have anoxic conditions (approximately four months per year), and which are affected by soil salinisation during the dry period (approximately eight months per year). This study points out that despite those very adverse conditions, soil invertebrates must be taken into account in the dynamic of soil organic and mineral properties in paddy fields. The effects of one earthworm species, Glyphodrillus sp., and an ant species, Epelysidris sp., on soil physical and chemical properties were determined through elemental soil physical and chemical properties (texture, pH, conductivity, C and N contents) and near infrared reflectance spectroscopy (NIRS) in order to evaluate their ability to influence soil organic matter quality. PCA processed with NIRS data clearly showed that biogenic structures (ant sheetings and earthworm casts) were separated from the control surrounding soil. Earthworms and ants affected differently soil properties. Glyphodrillus sp. increased the SOM content and decreased the pH on the surface of the soil. These effects were attributed to an increase in fine particle content (clay). Conversely, Epelysidris sp. only increased the content of coarse particles (sand) and did not influence either soil pH or SOM content. Soil conductivity was found to be very variable but was not significantly affected by soil invertebrates. These results show the potential of soil macro-fauna to create heterogeneity at small spatial scale and to modify the quality of surface soils even under adverse conditions like saline paddy fields.  相似文献

Abstract

The rapid expansion of mandarin (Citrus reticulata L.) production areas with high agrochemical input in the highland areas of northern Thailand has resulted in negative effects in terms of production, environment, soil quality, and public health. The use of microorganisms as plant growth promoters is an alternative method to reduce agrochemical input. Thus, we studied the diversity of endophytic actinomycetes in mandarin and their potential as plant growth promoters. A total of 252 endophytic actinomycete isolates were recovered from mandarin. Based on spore chain morphology, cell wall type, and 16S rRNA gene sequence, the isolates were classified into six genera: Streptomyces, Nocardia, Nocardiopsis, Spirillospora, Microbispora and Micromonospora. The most frequent isolates recovered were members of Streptomyces (85.3%). Selected isolates (64 isolates) from these genera were evaluated for their indole-3-acetic acid (IAA) production potential in a medium with 2 mg mL?1 tryptophan, and all the selected isolates showed the potential to produce IAA, with average values of IAA production of 13.34, 3.36, 140.38, 12.55, 1.40, and 6.19 µg IAA mL?1, respectively. Isolates of genus Nocardiopsis showed a very high ability to produce IAA that was the highest among all the genera, with values ranging from 62.23 to 222.75 µg mL?1. Twelve isolates selected from these genera were inoculated onto mandarin seedlings, and the results indicated that the shoot height, fresh shoot weight and fresh root weight of the seedlings were promoted by the inoculation of endophytic actinomycetes, with values ranging from 20.2 to 49.1%, 14.9 to 53.6%, and 1.6 to 102% over the control, respectively.  相似文献
