Fracture-related infections remain a leading cause of morbidity and mortality. We aimed to establish a simple contaminated radial osteotomy model to assess the efficacy of a biodegradable polymer poly(sebacic-co-ricinoleic acid) [p(SA-RA)] containing 20% w/w gentamicin. A unilateral transverse osteotomy was induced in Sprague-Dawley (SD) rats, followed by application of Staphylococcus aureus suspension over the fracture. After successfully establishing the contaminated open fracture model, we treated the rats either systemically (intraperitoneal cefuroxime), locally with p(SA-RA) containing gentamicin, or both. Control groups included non-contaminated group and contaminated groups that were either untreated or treated with the polymer alone. After 4 weeks, the bones were subjected to micro-CT scanning and microbiological and histopathology evaluations. Micro-CT analysis revealed similar changes in the group subjected to both local and systemic treatment as in the non-contaminated control group. Lack of detectable bacterial growth was noted in most animals of the group subjected to both local and systemic treatment, and all samples were negative for S. aureus. Histopathological evaluation revealed that all treatment modalities containing antibiotics were highly effective in reducing infection and promoting callus repair, resulting in early bone healing. While p(SA-RA) containing gentamicin treatment showed better results than cefuroxime, the combination of local and systemic treatment displayed the highest therapeutic potential in this model.  相似文献

Middle East respiratory syndrome coronavirus, MERS‐CoV, was identified in Saudi Arabia in 2012, and as of January 29, 2018, there were 2,123 laboratory‐confirmed MERS‐CoV cases reported to WHO (WHO, 2018, https://www.who.int/emergencies/mers-cov/en/ ). Multiple studies suggest that dromedary camels are a source for human MERS‐CoV infection. MERS‐CoV‐specific antibodies have been detected in the serum of dromedary camels across Northern Africa and across the Arabian Peninsula. Israel's geographic location places Israel at risk for MERS‐CoV infection. To date, MERS‐CoV‐related illness has not been reported and the burden of MERS‐CoV infection in the Israeli population is unknown. The seroprevalence of MERS‐CoV‐specific antibodies in Israeli dromedary camels is unknown. The objective of this study was to determine the prevalence of MERS‐CoV seropositivity in dromedary camels in Israel. The prevalence of MERS‐CoV antibodies in Israeli camels was examined in 71 camel sera collected from four farms across Israel by MERS‐CoV‐specific microneutralization (Mnt) assay and confirmed by MERS‐CoV‐specific immunofluorescence assay (IFA). Although this study cannot rule out potential antibody cross‐reactivity by IFA, the presence of bovine coronavirus‐specific antibodies do not appear to impact detection of MERS‐CoV antibodies by Mnt. MERS‐CoV neutralizing antibodies were detectable in 51 (71.8%) camel sera, and no association was observed between the presence of neutralizing antibodies and camel age or gender. These findings extend the known range of MERS‐CoV circulation in Middle Eastern camels. The high rate of MERS‐CoV‐specific antibody seropositivity in dromedary camels in the absence of any reported human MERS cases suggests that there is still much to be learned about the dynamics of camel‐to‐human transmission of MERS‐CoV.  相似文献

Bleeding during surgical procedures is a common complication. Therefore, hemostatic agents have been developed to control bleeding, and fibrin sealants have several benefits. sFilm-FS is a novel fibrin sealant that comprises a biodegradable co-polymeric film embedded with human fibrinogen and thrombin. Herein, the safety and efficacy of sFilm-FS were compared using a liver and spleen puncture model of Göttingen minipigs with those of the standard hemostatic techniques (control animals) and EVARREST®, a reference fibrin sealant. Hemostasis and reduced blood loss were more effectively achieved with sFilm-FS than with the standard techniques in the control animals and comparable to those achieved with EVARREST®. No treatment-related adverse effects were observed in any of the groups. Histopathological evaluation indicated that sFilm-FS was slightly and moderately reactive at the liver puncture site and spleen, respectively, compared with the standard techniques in the control animals. These changes are expected degradation reactions of the co-polymeric film and are not considered as adverse events. No treatment-related abnormalities were noted in the other evaluated organs. Additionally, no evidence of local or systemic thromboses was noted. These results support the use of sFilm-FS for hemostasis in humans.  相似文献

We investigated the effect of small-scale overstory gaps on the ecophysiology and growth of Quercus ithaburensis saplings. The study aim was to characterize how changes in daily exposure to direct beam radiation affect photosynthetic performance in the short term and growth and biomass partitioning in the long term. Using individual net-houses, the following treatments were applied: (a) Unshaded (daily irradiance = 100 %), (b) shading net with no gap (Shade-daily irradiance = 6 %), (c) shading net with 1 h gap allowing direct beam radiation (11:00 am–12:00 pm, Shade+1-irradiance = 20 %), (d) shading net with 3 h gap (11:00 am–2:00 pm, Shade+3-irradiance = 44 %). The experiment was performed in an irrigated field. We measured growth, biomass allocation, leaf traits, daily courses of leaf gas exchange and water potential. Oak dry-weight increased while height to dry-weight ratio and specific leaf area decreased with increasing daily exposure to direct beam radiation. Leaf chlorophyll content was less affected. Higher net carbon assimilation rates (A), stomatal conductance (gs) and A/gs were associated with higher instantaneous photosynthetic photon flux density (PPFD) throughout the entire experimental PPFD range. However, during gap-hours, while exposed to saturating radiation levels of similar level (ca. 1,800 µmol photon m?2 s?1), A in the Shade+1 oaks was about half that of the Shade+3 oaks and nearly one-third that of the Unshaded oaks. Patterns of gs, intercellular CO2 (Ci) and quantum efficiency of photosystem II pointed towards the possibility of a metabolic limitation. In conclusion, oaks benefited significantly from small scale overstory gaps though their capacity to utilize transient saturating radiation levels decreased with decreasing gap duration.  相似文献

Profitability of recirculating systems depends in part on the ability to manage nutrient wastes. Nitrogenous wastes in these systems can be eliminated through nitrifying and denitrifying biofilters. While nitrifying filters are incorporated in most recirculating systems according to well-established protocols, denitrifying filters are still under development. By means of denitrification, oxidized inorganic nitrogen compounds, such as nitrite and nitrate are reduced to elemental nitrogen (N2). The process is conducted by facultative anaerobic microorganisms with electron donors derived from either organic (heterotrophic denitrification) or inorganic sources (autotrophic denitrification). In recirculating systems and traditional wastewater treatment plants, heterotrophic denitrification often is applied using external electron and carbon donors (e.g. carbohydrates, organic alcohols) or endogenous organic donors originating from the waste. In addition to nitrate removal, denitrifying organisms are associated with other processes relevant to water quality control in aquaculture systems. Denitrification raises the alkalinity and, hence, replenishes some of the inorganic carbon lost through nitrification. Organic carbon discharge from recirculating systems is reduced when endogenous carbon sources originating from the fish waste are used to fuel denitrification. In addition to the carbon cycle, denitrifiers also are associated with sulfur and phosphorus cycles in recirculating systems. Orthophosphate uptake by some denitrifiers takes place in excess of their metabolic requirements and may result in a considerable reduction of orthophosphate from the culture water. Finally, autotrophic denitrifiers may prevent the accumulation of toxic sulfide resulting from sulfate reduction in marine recirculating systems. Information on nitrate removal in recirculating systems is limited to studies with small-scale experimental systems. Packed bed reactors supplemented with external carbon sources are used most widely for nitrate removal in these systems. Although studies on the application of denitrification in freshwater and marine recirculating systems were initiated some thirty years ago, a unifying concept for the design and operation of denitrifying biofilters in recirculating systems is lacking.  相似文献

Reduced fishery harvests and increased consumer demand for seafood have precipitated an increase in intensive fish farming, predominantly in coastal and open ocean net-pens. However, as currently practiced, aquaculture is widely viewed as detrimental to the environment and typical operations are vulnerable to environmental influences, including pollution and endemic diseases. Here we report the development of a land-based, marine recirculating aquaculture system that is fully contained, with virtually no environmental impact as a result of highly efficient biological waste treatment and water recycling. Over 99% of the water volume was recycled daily by integrating aerobic nitrification to eliminate toxic ammonia and, for the first time, simultaneous, anaerobic denitrification and anaerobic ammonium oxidation, to convert ammonia and nitrate to nitrogen gas. Hydrogen sulfide generated by the separated endogenous organic solids was used as an electron source for nitrate reduction via autotrophic denitrification and the remaining organic solids were converted to methane and carbon dioxide. System viability was validated by growing gilthead seabream (Sparus aurata) from 61 g to 412 g for a total of 1.7 tons in a record 131 days with 99% fish survival. Ammonia nitrite and nitrate did not exceed an average daily concentration of 0.8 mg/l, 0.2 mg/l and 150 mg/l, respectively. Food conversion values were 16% lower than recorded levels for net-pen aquaculture and saltwater usage of less than 16 l/every kg of fish produced. The system is site-independent, biosecure, devoid of environmental contaminants and is not restricted to a single species.  相似文献

Until the 1980s, at least 31 pairs of long-legged buzzards (Buteo rufinus) nested along the streams of the Judean Mountains in Israel, mostly on rocky cliffs, which – according to existing literature – is the common nesting style of this bird. During the past 40 years, however, nesting in these areas has substantially decreased, with many pairs of buzzards now nesting on trees in the Judean Foothills.We suggest that the geographical shift in nesting area, and with it the dramatic change from nesting on cliffs to nesting on trees, is probably due to the increase in land cover (as a result of afforestation, expansion of human settlements and recovery of the Mediterranean chaparral) that has taken place in the Judean Mountains during the last four decades. Buzzards forage in open habitats and the change in land cover has hindered their ability to locate prey. Since there are no cliffs appropriate for nesting in the Judean Foothills, the buzzards were thus forced to adapt to a new style of nesting. This hypothesis is further supported by our observations that within their new nesting grounds in the Judean Foothills, breeding success was significantly related to the area of the open habitat within the territory.These findings have important scientific and ecological implications. We recommend that foresters should take into consideration the effect of afforestation on open-landscape raptors, and while planning new forests will consider also their needs, particularly leaving large open swaths of land as foraging grounds for such raptors.  相似文献

Cattle grazing and overstory cover restrict understory growth and interact in shaping the understory community structure in Mediterranean conifer plantations.

Context

Understanding how silvicultural manipulations drive understory structure and function in Mediterranean pine plantations is essential for their multifunctional management.

Aims

This paper aims to study the interactive effects of cattle grazing and overstory thinning on understory structure and function.

Methods

Ten plots (0.25 ha) were selected in East Mediterranean mature Pinus brutia plantation (rainfall = 600 mm year?1) representing thinned (≈100 trees ha?1, leaf area index (LAI) ≈ 1.6) and non-thinned (≈230 trees ha?1, LAI ≈ 3.5) areas. Two subplots (100 m2) within each plot were fenced in 2000 and 2006 while a third one remained grazed. Understory growth and species composition were measured in 2010.

Results

Thinning and grazing exclusion both positively influenced woody growth with their combined effect during 10 years leading to 20-fold increase in vegetation volume. An increase (15-fold) in herbaceous biomass was recorded 4 years after grazing exclusion but disappeared 10 years after exclusion due to increased woody cover. Species richness was not influenced by grazing but was positively affected by thinning. Understory composition was affected by grazing × thinning interaction with herbaceous ephemerals and short woody species being more frequent in grazed, thinned areas while larger woody species were more associated with ungrazed, non-thinned areas.

Conclusion

Grazing impacts on forest understories depend on overstory cover. We propose variable grazing-thinning combinations to meet multiple management objectives.

Intensive recirculating aquaculture systems (RAS) produce high volumes of biosolid waste. The high salinity of brackish/marine sludge limits its use in landfill sites and waste outflows and it is a source of pollution. A reduction in sludge mass would therefore minimize the potential environmental hazard and economic burden stemming from its disposal. The aims of the current study were: 1) to characterize brackish aquaculture sludge (BAS) from three RAS in order to test for potentially suitable treatments, and 2) to test the BAS's suitability for anaerobic digestion in an upflow anaerobic sludge blanket reactor (UASB). Brackish sludge from three intensive RAS was collected periodically and analyzed for a variety of physical and chemical parameters. The mean sludge electrical conductivity and pH values ranged from 4.0 to 8.6 mS cm− 1 and 7.0 to 7.7, respectively. A low sludge redox potential averaging − 80 mV and dissolved oxygen concentrations of less than 1 mg l− 1 indicated the existence of anaerobic conditions. Volatile solids comprised 56 to 76% of the dry weight and the sludge volume index ranged from 44 to 69 ml g − 1. High concentrations of total nitrogen and total carbon were also observed, resulting in a C:N ratio ranging between 8.1 and 10.3. Toxic and/or inhibitory compounds for methanogenesis such as nitrites, nitrates and sulfides were almost absent. Sludge BOD5 ranged from 10 to 30% dry weight. These data suggest that BAS may be used in anaerobic digestion and methanogenesis without pretreatment. This concept was tested by digesting aquaculture sludge in UASB reactors. Despite the high sulfate and phosphate concentrations in the BAS, these were found not to be inhibitory to methanogenesis. Up to 70% sludge-mass reduction and an average of 40% methane production were demonstrated.  相似文献

Summary

Fruit of Cereus peruvianus (L.) Miller, a newly developed cactus crop, is subject to cracking. Fruit growth follows a double sigmoid curve, with a significant size increase in the first and third stages and moderate increase in the middle stage. Peel and seed grow in the first two stages, while pulp growth and maturation (e.g. peel colour change and stretching, sugar accumulation, fruit softening) occur in the final stage. Sensitive, moderately sensitive, and tolerant clones are subject to 100, 50–60 and 6–9% cracking, respectively, correlated to relative growth rate of the pulp in the third stage. Fruit cracking, lengthwise splitting of the peel, and exposure of the pulp occurred in fully mature fruits towards the end of their growth. Continuous monitoring of fruit growth revealed expansion and cracking during the day and shrinkage during the night, as expected for CAM plants. Cracking was not correlated with the threshold cracking turgor, which declined during ripening in all clones. GA3 sprays during colour break reduced cracking in the two most susceptible clones to 20% and 60%, while paclobutrazol increased cracking. Cracking rates were the same in two ecozones differing in salinity, temperatures and relative humidity. It is suggested that breeding and hormone treatments could reduce cracking significantly.  相似文献
