The rates at which (E)- and (Z)-mevinphos are hydrolysed in aqueous solutions in the pH range 8.7–11.5, and in the temperature range 30–50°C, have been measured. The overall activation parameters have been calculated, and equations to allow calculation of the rates in any basic conditions are given. The complicated routes by which (E)- and (Z)-mevinphos are hydrolysed to simple molecules have been deduced. The rates for the individual steps in each route have been either measured experimentally, or have been calculated through analogue simulation of all the reactions, by matching the calculated to the observed ultraviolet spectral changes during hydrolysis. It is proposed that (Z)-2-carboxyl-1-methylvinyl dimethyl phosphate is not observed as a hydrolysis product of (Z)-mevinphos because it decomposes by an extremely fast intramolecular reaction. The reasons for the greater lability of (E)-mevinphos are also discussed.  相似文献

Wheat flour from plants deficient in sulfur has been shown to contain substantially higher levels of free amino acids, particularly asparagine and glutamine, than flour from wheat grown where sulfur nutrition was sufficient. Elevated levels of asparagine resulted in acrylamide levels up to 6 times higher in sulfur-deprived wheat flour, compared with sulfur-sufficient wheat flour, for three varieties of winter wheat. The volatile compounds from flour, heated at 180 degrees C for 20 min, have been compared for these three varieties of wheat grown with and without sulfur fertilizer. Approximately 50 compounds were quantified in the headspace extracts of the heated flour; over 30 compounds were affected by sulfur fertilization, and 15 compounds were affected by variety. Unsaturated aldehydes formed from aldol condensations, Strecker aldehydes, alkylpyrazines, and low molecular weight alkylfurans were found at higher concentrations in the sulfur-deficient flour, whereas low molecular weight pyrroles and thiophenes and sugar breakdown products were found at higher concentrations in the sulfur-sufficient flour. The reasons for these differences and the relationship between acrylamide formation and aroma volatile formation are discussed.  相似文献

The objective of the study reported here was to determine the onset, duration, and degree of analgesia achieved with a combination of romifidine (50 microg/kg body weight [BW]) and morphine (0.1 mg/kg BW) administered epidurally. Ten adult Holstein Friesen cows were assigned to either a treatment group receiving the romifidine and morphine combination or a control group receiving 0.9% saline in a randomized, blinded, crossover design. Cows were assessed for degree of flank analgesia and systemic sedation at various time intervals over a period of 24 hours. The romifidine and morphine combination, compared with saline, provided significant analgesia for at least 10 minutes (P = 0.016) and up to 12 hours (P = 0.004) after epidural administration. Treated cows were sedate between 10 minutes (P = 0.016) and 6 hours (P = 0.002) after epidural administration. These results provide evidence for a potential cost-effective intra- and postoperative method of analgesia; however, the sedation seen in this study could be detrimental to patients expected return to the farm shortly after surgery. Further research into withdrawal times, systemic effects, and potential adverse effects are needed before an opiod and alpha2-adrenergic agonist combination can be used safely in a clinical setting.  相似文献

The objectives of this study were to investigate the determinants of the anion gap (AG) in cattle and to evaluate the utility of AG in detecting hyperlactatemia in sick neonatal calves and adult cattle. The AG was calculated as AG = ([Na+] + [K+]) - ([Cl-] + [HCO3]), with all values in mEq/L. The AG of healthy neonatal calves (n = 16) was 29.6 ± 6.2 mEq/L (mean ± SD), and the blood L-lactate concentration ranged from 0.5 to 1.2 mM/L. The AG was significantly (P > .05) correlated with serum phosphate (r = .66) and creatinine (r = .51) concentrations. The AG of neonatal calves with experimentally induced diarrhea (n = 16) was 28.6 ± 5.6 mEq/L, and the blood L-lactate concentration ranged from 1.1 to 2.9 mM/L. The AG was significantly correlated with blood L-lactate concentration (r = .67), serum phosphate concentration (r = .63), creatinine concentration (r = .76), and blood pH (r = -.61). The AG of adult cattle with abomasal volvulus (n = 41) was 20.5 ± 7.8 mEq/L, and the blood L-lactate concentration ranged from 0.6 to 15.6 mM/L. The AG was significantly correlated with blood L-lactate concentration (r = .60), serum phosphate concentration (r = .71), creatinine concentration (r = .65), albumin concentration (r = .47), total protein concentration (r = .54), blood pyruvate concentration (r = .67), and blood pH (r = -.41) but not plasma β-OH butyrate concentration. The results indicate that the AG in cattle is only moderately correlated with blood L-lactate concentration and is similarly correlated with serum phosphate and creatinine concentrations in neonatal calves and adult cattle, as well as with serum albumin and total protein concentrations in adult cattle. Anion gap determination is of limited usefulness in predicting blood L-lactate concentration in sick cattle, whereas the correlation between AG and serum creatinine concentration in sick cattle suggests that an increased AG should alert the clinician to the potential presence of uremic anions.  相似文献

ObjectiveTo quantify induction time, reliability, physiological effects, recovery quality and dart volume of a novel formulation of alfaxalone (40 mg mL?1) used in combination with medetomidine and azaperone for the capture and handling of wild bighorn sheep.Study designProspective clinical study.AnimalsA total of 23 wild bighorn sheep (Ovis canadensis) in Sheep River Provincial Park, AB, Canada.MethodsFree-ranging bighorn sheep were immobilized using medetomidine, azaperone and alfaxalone delivered with a remote delivery system. Arterial blood was collected for measurement of blood gases, physiologic variables (temperature, heart and respiratory rates) were recorded and induction and recovery length and quality were scored.ResultsData from 20 animals were included. Administered dose rates were alfaxalone (0.99 ± 0.20 mg kg?1; 40 mg mL?1), azaperone (0.2 ± 0.04 mg kg?1; 10 mg mL?1) and medetomidine (0.16 ± 0.03 mg kg?1; 30 mg mL?1). The mean drug volume injected was 1.51 mL. The median (range) induction time was 7.7 (5.8–9.7) minutes, and recovery was qualitatively smooth.Conclusions and clinical relevanceAn increased concentration formulation of alfaxalone was administered in combination with medetomidine and azaperone, and resulted in appropriate anesthesia for the capture and handling of bighorn sheep. A novel proximity ligation assay (PLA) using a pan-serotype reactive monoclonal antibody was developed and evaluated for the detection of foot-and-mouth disease virus (FMDV) in clinical samples collected from field cases of disease. The FMDV-specific PLA was found to be 100 times more sensitive for virus detection than the commonly used antigen capture-ELISA (AgELISA). As few as five TCID50 were detected in individual assays, which was comparable with the analytical sensitivity of real-time RT-PCR. Although this assay was capable of detecting diverse isolates from all seven FMDV serotypes, the diagnostic sensitivity of the PLA assay was lower than real-time RT-PCR mainly due to a failure to detect some SAT 1, SAT 2 and SAT 3 FMDV strains. In conclusion, this new PLA format has high analytical sensitivity for the detection of FMDV in clinical samples and may prove valuable as a rapid and simple tool for use in FMD diagnosis.  相似文献

A cross-sectional study was designed to investigate the prevalence and risk factors for Cryptosporidium infection in young calves. Forty-one farms in a discrete, densely farmed 100 km2 area of North West England were visited over a 3-week period and 215 faecal samples were collected from young calves. Farms were not selected on the basis of existing scour problems. At the time of sampling, several investigator-observed variables were recorded at the pen, animal and stool levels. Samples were screened and 60/215 were confirmed as positive by PCR of the 18S rRNA gene. Risk factors for infection were explored using multilevel multivariable logistic regression with farm as a random effect. Age was significant in the final model, with a higher risk of infection in calves aged 8-21 days, when compared to those aged 0-7 days. The depth of the bedding was also significant in the final model, with calves housed in bedding 11-15 cm deep being at lower risk of infection than those on beds 0-5 cm deep. Consistency of the faeces was highly correlated with age and colour of the faeces and was not significantly associated with infection when these variables, and clustering at farm-level, were accounted for. This is interesting as Cryptosporidium is considered to be a primary enteropathogen. The results suggest that intervention strategies should be targeted at calves under 21 days old. These animals represent a significant reservoir of infection on the farm and may also pose a risk to public health, assuming that the species and genotypes shed are zoonotic pathogens.  相似文献
