The excretion rate of [3H]prednisolone from clinically normal and experimentally infected udders of 10 lactating cows was studied. Each quarter of 6 cows was injected with a single dose of [3H]prednisolone mixed with non-radioactive prednisolone equivalent to 10 mg in 10 ml of peanut oil base. Each of the remaining 4 cows was given 40 mg of nonradioactive prednisolone and [3H]prednisolone in 60% ethanol IV. Control and postadministration samples of blood, milk, and urine were examined for radioactivity. The effects of [3H]prednisolone were evaluated in the same cows, first in clinically normal udders, then 2 weeks later in udders experimentally infected with Streptococcus agalactiae. Absorption and elimination of prednisolone were the same before and after induced infection. Within 3 hours after intramammary injection, 95% of the labeled prednisolone was absorbed systemically, less than 5% of this dose was recovered in milk, and 29% was excreted in urine. After IV injection of [3H]prednisolone, less than 0.2% of the total radioactivity was recovered in milk and less than 46% was excreted in urine. Clinical mastitis induced by S agalactiae was moderate. Circulating blood leukocytes and somatic cells in the milk of normal cows remained essentially unchanged. The leukocyte response to induced infection was rapid in blood and milk. Large numbers of leukocytes were noticed in the milk and a severe leukopenia occurred. Prednisolone treatment did not alter the number of somatic cells in milk or reduce the inflammatory response of experimentally infected cows.  相似文献

A comparative study on the response of four indigenous cattle breeds of Ethiopia, namely Abigar, Horro, Sheko and Gurage, to natural challenge of trypanosomosis in the Tolley-Gullele area of the Ghibe valley has been undertaken from August 2000 until August 2004. Fifty female yearlings each of Horro, Sheko and Abigar and 31 of the Gurage were purchased from their natural habitats and introduced in to medium to high tsetse-trypanosomosis challenge area of the Ghibe valley. While the natural habitats of first three breeds are naturally infested with tsetse flies and trypanosomosis, that of the Gurage is known to be very minimal, if any, and hence the Gurage breed was used in this study as the known susceptible breed. During the study animal health, production performance and tsetse fly situation were monitored monthly. The Sheko breed has manifested very significantly (p<0.001) high overall average packed cell volume (PCV) values (25%) compared to that of Abigar (24%), Horro (23%) and Gurage (22%). It also had the lowest mean trypanosome prevalence rate of 9% against 23% of Horro, 26% of Abigar and 27% of Gurage, and the least number of Berenil treatments (1.36) compared to Abigar (4.0), Horro (4.6) and Gurage (6.7). While the Abigar manifested high sensitivity and frequent death to PCV depression, the Horro showed strong resilience to PCV depression and better response to Berenil treatment assistance. At this stage the Sheko breed was also found to be equal to the other breeds in its reproductive performance. These results need to be substantiated with further in-depth investigation including immune response, animal behavior and environmental influences.  相似文献

Sorghum (Sorghum bicolor (L.) Moench) is an important food crop in arid and semi-arid tropics where it is grown mainly due to its drought tolerance. In Ethiopia, sorghum is grown in different agro-ecological zones primarily for food. Landraces are widely cultivated in the region due to their adaptation to the growing conditions and other characteristics such as food quality, quantity, and quality of by-products. The objective of this research was to determine the genetic variation in protein, mineral composition, total starch and its components in sorghum landraces collected from the western regions of Ethiopia. Sorghum whole grains were analyzed for crude protein, total starch and its components, and mineral profile (calcium, magnesium, potassium, phosphorus, iron, manganese, zinc, and sodium). The analysis of variance showed significant differences among the sorghum landraces in nutritional composition. There was highly significant correlation between phosphorus and magnesium, and between zinc and phosphorus and protein which indicated that some interaction existed in absorption and translocation between minerals and protein content. The genetic variation found among the sorghum landrace accessions in chemical composition can be exploited in the breeding program.  相似文献

Countering the economic hurdle caused by coffee leaf rust disease is most appealing at this time as it has posed a major threat to coffee production around the world. Establishing differential expression profiles at different times following pathogen invasion in both innate and acquired immunities unlocks the molecular components of resistance and susceptibility. Suppression subtractive hybridization (SSH) was used to identify genes differentially over-expressed and repressed during incompatible and compatible interactions between Coffea arabica and Hemileia vastatrix. From 433 clones of expressed sequence tags (ESTs) sequenced, 352 were annotated and categorized of which the proportion of genes expressed during compatible interaction were relatively smaller. The result showed upregulation and downregulation of various genes at 12 and 24 h after pathogen inoculation in both interactions. The use of four different databases in searching for gene homology resulted in different number of similar sequences. BLASTx against EMBL-EBI (European Molecular Biology Laboratory-European Bioinformatics Institute) database being with the maximum (100%) hits for all the annotated sequences. RT-qPCR analysis of seven resistance-signaling genes showed similar expression patterns for most of the genes in both interactions, indicating these genes are involved in basal (non-specific) defense during which immune reactions are similar. Using SSH, we identified different types of resistance related genes that could be used for further studies towards resistant cultivar development. The potential role of some of the resistance related proteins found were also discussed.  相似文献

Anchote (Coccinia abyssinica (Lam.) Cogn.) is a perennial root crop belonging to Cucurbitaceae family. It is endemic to Ethiopia and distributed over wide range of agro-ecologies. For further improvement and efficient conservation of this crop, characterization of its genetic diversity and its pattern of distribution is a vitally important step. Expressed sequence tags-simple sequence repeats (EST-SSRs) markers were developed from publicly available watermelon [Citrullus lanatus (Thunb.) Matsum. & Nakai] ESTs in the GenBank database. Among those novel markers, eight were polymorphic and subsequently used for genetic diversity and population structure analyses of 30 anchote accessions collected from western Ethiopia. A total of 24 alleles were obtained across the eight polymorphic loci and 30 accessions that revealed moderate level of genetic diversity in this minor crop. Among the eight loci, locus CA_06 was the most informative with six alleles and polymorphic information content (PIC) of 0.76. The accessions showed about threefold variation in terms of genetic diversity, with expected heterozygosity (He) ranging from 0.15 (accession An) to 0.44 (accession Dg). Other accessions with higher genetic diversity include Ar and Gu (He?=?0.43 and 0.41, respectively). Analysis of molecular variance (AMOVA) revealed that the variation within accessions and among accessions accounted for 84.7% and 15.3% of the total variation, respectively. The study revealed low but significant population differentiation in this crop with no clear pattern of population structure. The EST-SSR markers developed in this study are the first of their kind for anchote and can be used for characterization of its wider genetic resources for conservation and breeding purposes.

Niger (noug; Guizotia abyssinica) is an economically important edible oilseed crop. This review sought to demonstrate the significance of niger for sustainable food security in Ethiopia, where it is produced exclusively by smallholders. In addition to its edible oil, niger seed is an important source of proteins, carbohydrates, vitamins and fiber that significantly contribute to the human diet. The crop has a wide genetic basis that is reflected in the form of a high variation in desirable traits, including seed yield, seed oil content, seed oil quality and photoperiod sensitivity. However, the wealth of niger genetic diversity has so far remained largely unexploited and research efforts have not yet yielded satisfactory results in the form of new and superior cultivars. The recent molecular and nutritional quality studies coupled with pre-breeding work have opened up new opportunities for the improvement of niger. A high yielding niger cultivars with oil content of up to 60 % and/or oleic acid content of up to 70 % can easily be bred based on Ethiopian niger gene pool through the combined use of novel genomic tools, traditional breeding and farmer-participatory approaches. The improvement of niger will have a significant contribution towards Ethiopia’s food security and sustainable development in general and self-sufficiency in edible oil in particular. Overall, an investment in the niger improvement programs will likely be of benefit far beyond Ethiopia’s borders and could potentially lead to the expansion of the crop outside the regions where it is currently grown.  相似文献

Cultivated sorghum [Sorghum bicolor (L.) Moench] is an important food security crop in the semi-arid regions of the world including Asia and Africa. Its genetic diversity is contained mostly in traditional varieties and modern cultivars used by farmers. In this study, agro-morphological traits and molecular markers were used to assess genetic diversity in 22 accessions of cultivated sorghum from five countries (Botswana, Namibia, Swaziland, Zambia and Zimbabwe) in the Southern African Development Community (SADC) region. The study revealed a significant variation among 22 accessions in both qualitative and quantitative morphological traits, indicating the accessions’ promising potential as breeding material. For molecular analysis, 11 microsatellite primer-pairs were used, and generated a total of 70 alleles across 20 accessions. Analysis of molecular variance revealed a high level of genetic variation; 67 % among the accessions and 10 % among the five countries. The patterns of genetic diversity and the relationships observed in this study should provide insights for genetic resource conservation and utilization of sorghum germplasm in the SADC region.  相似文献
