Short basal internodes are important for lodging resistance of rice(Oryza sativa L.).Several canopy indices affect the elongation of basal internodes,but uncertainty as to the key factors determining elongation of basal internodes persists.The objectives of this study were(1)to identify key factors affecting the elongation of basal internodes and(2)to establish a quantitative relationship between basal internode length and canopy indices.An inbred rice cultivar,Yinjingruanzhan,was grown in two split-plot field experiments with three N rates(0,75,and 150 kg N ha−1 in early season and 0,90,and 180 kg N ha−1 in late season)as main plots,three seedling densities(16.7,75.0,and 187.5 seedlings m−2)as subplots,and three replications in the 2015 early and late seasons in Guangzhou,China.Light intensity at base of canopy(Lb),light quality as determined from red/far-red light ratio(R/FR),light transmission ratio(LTR),leaf area index(LAI),leaf N concentration(NLV)and final length of second internode(counted from soil surface upward)(FIL)were recorded.Higher N rate and seedling density resulted in significantly longer FIL.FIL was negatively correlated with Lb,LTR,and R/FR(P<0.01)and positively correlated with LAI(P<0.01),but not correlated with NLV(P>0.05).Stepwise linear regression analysis showed that FIL was strongly associated with Lb and LAI(R2=0.82).Heavy N application to pot-grown rice at the beginning of first internode elongation did not change FIL.We conclude that FIL is determined mainly by Lb and LAI at jointing stage.NLV has no direct effect on the elongation of basal internodes.N application indirectly affects FIL by changing LAI and light conditions in the rice canopy.Reducing LAI and improving canopy light transmission at jointing stage can shorten the basal internodes and increase the lodging resistance of rice. 相似文献

The objective of this study was to investigate the structural changes of whey proteins during exposure in a continuous-flow UV reactor. Varying UV irradiation dosages were obtained by controlling the flow rate and the mixing speed. Whey protein isolate (WPI) solutions at concentrations of 1% and 5% (w/v) were circulated at flow rates ranging from 30 to 800 mL·min(-1), and changes in physicochemical properties of the proteins were investigated. Intrinsic fluorescence spectra and surface hydrophobicity measurements suggested changes in the tertiary structure of the proteins with UV exposure. The UV treatment also increased the concentration of total and accessible thiol groups in 1% WPI solutions, while no change was measured in 5% WPI solutions. Size-exclusion chromatography demonstrated the formation of UV-induced aggregates and oxidation products (N-formylkynurenine and dityrosine) of aromatic amino acids. Furthermore, the UV-induced changes in protein conformation increased the susceptibility of whey proteins to pepsin hydrolysis. 相似文献

南方低甲烷排放的高产水稻品种筛选与评价 总被引:3,自引:1,他引:3
为综合评价高产和低甲烷(CH4)排放兼具的水稻品种,应对我国粮食安全和环境保护问题,于2013年晚季和2014年早季,以南方稻区50个代表品种为供试材料,在盆栽条件下采用静态箱-气相色谱法测定品种的CH4排放通量,并对生育期的CH4排放量、稻谷产量及植株农艺性状进行了综合评价。结果表明,早、晚季供试品种间的CH4排放量和产量均存在显著差异。早季全生育期的CH4排放量在7.37~21.10 g·m-2之间,晚季分蘖期至幼穗分化期的CH4排放量在1.37~13.97 g·m-2之间;早季产量在0.40~0.65kg·m-2之间,晚季在0.59~0.87 kg·m-2之间。根据系统聚类法的分析结果,将供试品种划分成具有不同CH4排放量和产量水平的类型,从中筛选出了适合推广应用的CH4低排高产、中排高产和低排中产品种,如黄华占、五优308、丰华占、特三矮、茉莉占选、广超丝苗、珍桂矮、广恢998和七桂早25等。其中,恢复系广恢998组配品种、黄华占和五优308已在我国南方稻区大面积推广应用。相关分析结果表明,株高、叶面积指数与CH4排放通量呈显著正相关,这2个性状可作为高产低CH4排放水稻新品种选育的辅助筛选指标。此外,稻谷产量与CH4排放量无显著相关。研究结果为培育高产与低CH4排放兼具的新品种提供了亲本材料和理论依据。 相似文献

The stone marten (Martes foina) is a carnivorous mammal that often consumes fleshy fruits, thus potentially promoting seed dispersion. The present study was developed in Bussaco National Forest, central Portugal, and aimed to assess the potential role of the stone marten in dispersing native and exotic plants in different forest landscape types. Seeds from stone marten scats and fleshy fruits were collected monthly and were thereafter identified and sowed in a nursery, following a randomized experimental setup. Plant emergence was monitored fortnightly. Generalized linear models were used to test for differences in time and success of emergence between seeds from scats and fruits of 3 native species (Rubus ulmifolius, Arbutus unedo and Celtis australis) and 1 exotic plant species with invasive behavior (Prunus laurocerasus). Fruit consumption by the stone martens significantly increased and accelerated the germination of the native R. ulmifolius but had no effect on the other 2 native species or on the invasive species. This suggested that stone martens contribute to gene flow and forest regeneration by dispersing native plant seeds. However, although the germination was not enhanced in the invasive species, the preference of stone martens for these fruits may potentially contribute to the proliferation of P. laurocerasus. Despite the recognised importance of oxidative stress in the health and immune function of dairy cows, protein oxidation markers have been poorly studied in this species. The current study aimed to characterise markers of protein oxidation generated by activated bovine neutrophils and investigate the biological effects of advanced oxidation protein products (AOPP) on bovine neutrophils. Markers of protein oxidation (AOPP, dityrosines and carbonyls) were measured in culture medium containing bovine serum albumin (BSA) exposed to neutrophils. The effect of AOPP-BSA on generation of reactive oxygen species (ROS) was assessed by chemiluminescence. Activation of caspases-3, -8 and -9 and the presence of DNA laddering were used as apoptosis markers.Greater amounts of AOPP were generated by phorbol myristate acetate (PMA)-activated than non-activated neutrophils (1.46 ± 0.13 vs. 0.75 ± 0.13 nmol/mg protein, respectively; P < 0.05). Activated neutrophils and hypochlorous acid generated slightly different patterns of oxidized protein markers. Exposure to AOPP-BSA did not stimulate ROS production. Activated neutrophils generated a lesser amount of ROS when incubated with AOPP-BSA (P < 0.001). Activation with PMA induced a loss of viable neutrophils after 3 h, which was greater with AOPP-BSA incubation (P < 0.05). Detectable amounts of active caspases-3, -8 and -9 were found in nearly all samples but differences in caspase activation or DNA laddering were not observed comparing treatment groups. Apoptosis was unlikely to be responsible for the greater loss of PMA-activated neutrophils cultured in AOPP-BSA and it is possible that primary necrosis occurred. The results suggest that accumulation of oxidized proteins at an inflammatory site might result in a progressive reduction of neutrophil viability. 相似文献

The report demonstrates that the induction of apoptosis in peripheral blood granulocytes and lymphocytes of rabbits infected with three non-haemagglutinating RHDV strains (English Rainham, German Frankfurt, and Spanish Asturias) is a crucial determinant of the pathogenesis of rabbit haemorrhagic disease. Apoptosis was measured by flow cytometric detection of caspase activity. These studies demonstrated that the investigated RHDV (rabbit haemorrhagic disease virus) viral strains affected leukocyte apoptosis to varying degrees. Enhanced leukocyte apoptosis was detected between 4 and 36h after infection and was more pronounced in lymphocytes than in granulocytes. The data presented here thus provide a preliminary understanding of the kinetics of apoptosis in leukocytes of rabbits infected with RHDV. 相似文献

Summary Functional markers (FMs) are derived from polymorphic sites within genes causally involved in phenotypic trait variation (Andersen,
J.R. & T. Lübberstedt, 2003. Trends Plant Sci 8: 554–560). FM development requires allele sequences of functionally characterized
genes from which polymorphic, functional motifs affecting plant phenotype can be identified. In maize and other species with
low levels of linkage disequilibrium, association studies have the potential to identify sequence motifs, such as a few nucleotides
or insertions/deletions, affecting trait expression. In one of the pioneering studies, nine sequence motifs in the dwarf8 gene of maize were shown to be associated with variation for flowering time (Thornsberry, J.M., M.M. Goodman, J. Doebley,
S. Kresovich, D. Nielsen & E.S. Buckler, 2001. Nat Genet 28: 286–289). Proof of sequence motif function can be obtained by
comparing isogenic genotypes differing in single sequence motifs. At current, the most appropriate approach for this purpose
in crops is targeting induced local lesions in genomes (TILLING) (McCallum, C.M., L. Comai, E.A. Greene & S. Henikoff, 2000.
Nat Biotechnol 18: 455–457). In central Europe, maize is mainly grown as forage crop, with forage quality as major trait,
which can be determined as proportion of digestible neutral detergent fiber (DNDF). Brown midrib gene knock out mutations have been shown to be beneficial for forage quality but disadvantageous for overall agronomic performance.
Two brown midrib genes (bm1 and bm3) have been shown to be involved in monolignol biosynthesis. These two and additional lignin biosynthesis genes have been
isolated based on sequence homology. Additional candidate genes putatively affecting forage quality have been identified by
expression profiling using, e.g., isogenic bm lines. Furthermore, we identified an association between a polymorphism at the COMT locus and DNDF in a collection of European

An Agrobacterium-mediated transformation procedure for soybean [Glycine max L. Merrill] proliferating somatic embryos is here described. The Agrobacterium tumefaciens LBA4404 strain harboring pTOK233, pCAMBIA1390-olp or pH7WG2Dwrky plasmids was used to mediate gene transfer into the plant genome. Prior to Agrobacterium inoculation, proliferative soybean embryogenic clusters were microwounded by DNA-free tungsten particle bombardment. Three
independent transformation experiments were performed. In Experiment I, 26 transgenic plants were obtained from a unique clone
of cv Bragg, while 580 plants were recovered from 105 clones of cv IAS5. In Experiment II, a single hygromycin-resistant clone
of cv BRSMG68 Vencedora was recovered and gave rise to five plants. In Experiment III, 19 plants of cv Bragg and 48 plants
of IAS5 were recovered, representing five and 14 independent transformation events, respectively. PCR and Southern analyses
confirmed the transgenes’ integration into plant genomes. Transgenic plants were fertile. They flowered, set pods and seeds.
Transgene segregation in two T1 progenies fits the Mendelian pattern (3:1 transgenic:non-transgenic plants). This is the first report of transgenic fertile
soybean plants obtained from somatic embryogenic tissues transformed by the system that combines DNA-free particle bombardment

【目的】研究施氮对水稻茎秆抗倒伏能力的影响及其与茎秆形态性状和力学性状的关系及其作用机理,为水稻抗倒高产栽培调控提供依据。【方法】以常规籼稻银晶软占为材料,设置4个氮水平,进行大田试验,研究施氮对水稻茎秆基部节间形态和力学性状的影响及其与倒伏指数的关系。【结果】施氮影响水稻茎秆的形态和力学性状。随着施氮量的增加,株高增加,重心上移,基部节间长度增加,节间充实度下降,抗折力和弹性模量减小,茎秆倒伏指数增加,抗倒伏能力下降。倒伏指数与株高、重心高度及基部节间长度呈正相关,而与基部节间充实度、抗折力及弹性模量呈负相关,且相关系数大多达到显著或极显著水平。倒伏指数与单位面积有效穗数和每穗总粒数呈显著或极显著正相关,而与结实率和千粒重呈负相关趋势。【结论】株高、重心高度、基部节间长度和基部节间充实度等形态性状,以及弯曲力矩、抗折力等力学性状,是影响水稻茎秆抗倒伏能力的主要因素。 相似文献
