The first genetic linkage map of macadamia (Macadamia integrifolia and M. tetraphylla) is presented. The map is based on 56 F1 progeny of cultivars ‘Keauhou’ and ‘A16’. Eighty-four percent of the 382 markers analysed segregated as Mendelian loci. The two-way pseudo-testcross mapping strategy allowed construction of separate parental cultivar maps. Ninety bridging loci enabled merging of these maps to produce a detailed genetic map of macadamia, 1100 cm in length and spanning 70–80% of the genome. The combined map comprised 24 linkage groups with 265 framework markers: 259 markers from randomly amplified DNA fingerprinting (RAF), five random amplified polymorphic DNA (RAPD), and one sequence-tagged microsatellite site (STMS). The RAF marker system unexpectedly revealed 16 codominant markers, one of them a putative microsatellite locus and exhibiting four distinct alleles in the cross. This molecular study is the most comprehensive examination to date of genetic loci of macadamia, and is a major step towards developing marker-assisted selection for this crop. Purpose

Sulfamethazine (SMT) is increasingly detected in environmental matrices due to its versatile use as antibiotics. We aimed to investigate the benefits and roles of steam activation of biochars with respect to SMT sorption kinetics and equilibrium sorption.

Materials and methods

Biochars were produced from burcucumber plant and tea waste using a pyrolyzer at a temperature of 700 °C for 2 h. The biochar samples were treated with 5 mL min?1 of steam for an additional 45 min for post-synthesis steam activation. The SMT sorption on the unmodified and steam activated biochars were compared.

Results and discussion

The time taken to reach equilibrium was significantly less for steam activated biochars (~4 h) than non-activated biochars (>24 h). Up to 98 % of SMT could be removed from aqueous solutions by steam activated biochars. The sorption kinetic behaviors were well described by the pseudo-second model and SMT sorption rates of steam activated biochars (k 2?~?1.11–1.57 mg g?1 min?1) were significantly higher than that of the unmodified biochars (k 2?~?0.04–0.11 mg g?1 min?1) because of increased availability of accessible porous structure with averagely larger pore diameters. Moreover, the equilibrium sorption on the unmodified biochars was significantly influenced by increasing solution pH (~30–50 % reduction) because of speciation change of SMT, whereas steam activated biochars manifested much stronger sorption resilience against pH variation (~2–4 % reduction only) because the enhanced porosity offset the effect of unfavorable electrostatic repulsion.

Conclusions

The observed features of steam activated biochars would render their applications more versatile and reliable in field throughout changeable environmental conditions.

Summary

The effect of temperature on inflorescence development and sex expression in two mono-embryonic (`Irwin' and `Sensation'), and two poly-embryonic (`Nam Dok Mai' and `Kensington') mango cultivars was studied. Trees were subjected to natural winter temperatures to induce flowering prior to transfer into controlled environment glasshouse rooms under day/night temperature regimes of 15/5, 20/10, 25/15 and 30/208C for 20 weeks. Inflorescence development did not progress when trees were held at 15/58C. Cooler temperatures (20/108C) delayed the start of anthesis (42.4.d) compared with trees grown at 25/158C (23.d) and 30/208C (16.1.d). At 20/108C, the delay in the start of anthesis was greatest for `Sensation' (55.5.d) and least for `Nam Dok Mai' (25.5.d) while at other temperatures there was little difference between cultivars. The distribution of hermaphrodite flowers within the inflorescence was independent of temperature with the highest percentage found in the apical half of the inflorescence. There was an inverse relationship between the length of the anthesis period and temperature, with anthesis occurring over 30.d at 20/108C and reducing to fewer than 10.d at 30/208C. Temperature also had an inverse effect on the total number of flowers per inflorescence with 619.6.6.108.0 (mean for all cultivars) at 20/108C decreasing to 431.3.6.80.5 at 30/208C.  相似文献

Summary

The effect of a constant (10, 15, 20 or 25°C) or a diurnal maximum/minimum (15/5, 20/10, 25/15 or 30/20°C) incubation temperature on in vitro pollen germination and pollen tube growth in the pistils of two poly-embryonic (‘Kensington’ and ‘Nam Dok Mai’), and two mono-embryonic (‘Irwin’ and ‘Sensation’) mango cultivars was studied. In in vitro experiments where pollen was incubated in a liquid germination medium for 24 h in darkness, little difference was found between pollen germination of mono- and poly-embryonic cultivars. Averaged over the four cultivars, 53.9% of pollen germinated at 10°C, this increased to 76.2% when the incubation temperature was increased to 15°C, thereafter up to 25°C the percentage germination remained stable but germination decreased slightly to 68.2% at 30°C. Similarly, there was no difference in percent germination between cultivars when pollen was incubated under diurnal temperature regimes. Mean pollen germination of all four cultivars was 52.3% at 15/5°C and pollen germination increased by 10% when the temperature was raised to 30/20°C. When self-pollinated flowers were incubated for 24 h on a semi-solid agar medium at 10°C, pollen tube growth of the four cultivars was retarded and no pollen tubes reached the ovaries. As the temperature was increased from 15 to 25°C, the mean number of pollen tubes in ovaries increased from 0.04 to 0.25. At 30°C, the mean number of pollen tubes that entered ovaries decreased to 0.04. After incubation under diurnal temperature regimes, the mean number of pollen tubes in ovaries of all four cultivars at 15/5°C was 0.23 and increased to 0.42 when the temperature increased to 30/20°C. At each incubation temperature, there were significant differences in pollen tube growth between cultivars, but there were no differences between the temperature response of pollen from mono- and poly-embryonic cultivars.  相似文献

Purpose

In this study, we investigated the effect of biochar (BC) and fungal bacterial co-inoculation (FB) on soil enzymatic activity and immobilization of heavy metals in serpentine soil in Sri Lanka.

Materials and methods

A pot experiment was conducted with tomatoes (Lycopersicon esculentum L.) at 1, 2.5, and 5 % (w/w) BC ratios. Polyphenol oxidase, catalase and dehydrogenase activities were determined by idometric, potassium permanganate oxidisable, and spectrophotometric methods, respectively. Heavy metal concentrations were assessed by 0.01 M CaCl2 and sequential extraction methods.

Results and discussion

An increase in BC application reduced polyphenol oxidase, dehydrogenase, and catalase activity. The application of FB increased soil dehydrogenase activity, with the maximum activity found in 1 % BC700?+?FB treatment. Moreover, the CaCl2 extractable metals (Ni, Mn, and Cr) in 5 % BC700 amended soil decreased by 92, 94, and 100 %, respectively, compared to the control. Sequential extraction showed that the exchangeable concentrations of Ni, Mn, and Cr decreased by 55, 70, and 80 % in 5 % BC700, respectively.

Conclusions

Results suggest that the addition of BC to serpentine soil immobilizes heavy metals and decreases soil enzymatic activities. The addition of FB to serpentine soil improves plant growth by mitigating heavy metal toxicity and enhancing soil enzymatic activities.

In most arid and semiarid soils, naturally occurring phosphorus(P) is a major yield-limiting plant nutrient. In this study, to investigate the effects of organic(OP) and inorganic P(IP) sources on P fractionation, a calcareous sandy loam alkaline soil was fertilized with OP and IP fertilizers at low(80 mg P kg~(-1) soil) and high(160 mg P kg~(-1) soil) application rates. Three combinations of OP and IP(i.e., 75% OP + 25% IP, 50% OP + 50% IP, and 25% OP + 75% IP) were applied at low and high application rates,respectively, followed by soil aging for 21 d. Soil samples were collected after 1, 2, 3, 7, and 21 d and subjected to sequential extraction to analyze soluble and exchangeable, Fe-and Al-bound, Ca-bound, and residual P fractions. The soluble and exchangeable P fraction significantly increased up to 24.3%, whereas the Ca-bound fraction decreased up to 40.7% in the soils receiving 75% OP + 25% IP and 50% OP + 50% IP, respectively, compared with the control(receiving no P fertilizer). However, the transformation of P fractions was influenced by aging time. Addition of P sources caused instant changes in different P fractions, which then tended to decline with aging time. Change in soil p H was the limiting factor in controlling P availability. At high application rate, the OP source significantly increased soil P availability compared with the IP source with soil aging. Depending on P fractionation, a proper combination of OP and IP fertilizers, as long-term slow and instant P-releasing sources for plant uptake, respectively, may be a sustainable strategy to meet crop P requirements in the arid and semiarid soils.  相似文献

Summary

Exposure to ethylene gas elicits flower abscission from cut stems of Geraldton waxflower (Chamelaucium uncinatum Schauer). Ethylene response rates in plants are mediated by temperature. At 20°C, flower abscission from waxflower ‘Purple Pride’ occurred upon 12 h exposure to 1 µ11–1 ethylene. This ethylene treatment did not cause flower abscission at either 10 or 2°C. Moreover, flowers held at 2°C were insensitive to 48 h exposure to 1, 10 and 100 µ11–1 ethylene. However, increasing the duration of treatment with 1 µ11–1 ethylene at 10 and 2°C to 48 and 144 h, respectively, induced flower abscission. When flowers were held at 20°C in air without exogenous ethylene following continuous exposure to 1 µ11–1 ethylene at 2°C, the duration required to elicit flower abscission was reduced from 144 to 72 h. Collectively, these responses show that maintaining harvested waxflower at low temperature (e.g. 2°C) is an effective means to minimise ethylene-mediated flower abscission.  相似文献
