Fluconazole resistance is becoming an important clinical concern. We studied the in vitro effects of cinnamaldehyde against 18 fluconazole-resistant Candida isolates. MIC90 of cinnamaldehyde against different Candida isolates ranged 100–500 μg/ml. Growth and sensitivity of the organisms were significantly affected by cinnamaldehyde at different concentrations. The rapid irreversible action of this compound on fungal cells suggested membrane-located targets for its action. Insight studies to mechanism suggested that cinnamaldehyde exerts its antifungal activity by targeting sterol biosynthesis and plasma membrane ATPase activity. Inhibition of H+-ATPase leads to intracellular acidification and cell death. Toxicity against H9c2 rat cardiac myoblasts was studied to exclude the possibility of further associated cytotoxicity. The observed selectively fungicidal characteristics against fluconazole-resistant Candida isolates signify a promising candidature of this essential oil as an antifungal agent in treatments for candidosis.  相似文献

The possibility of manufacturing rubberwood and kenaf (Hibiscus cannabinus L.) stem medium density fibreboard (MDF) panels at different pressure and resin content were investigated. The effect of mechanisms of interacted independent variables (resin content and pressure) on MDF properties was analyzed. The board performance was evaluated by measuring internal bond (IB) strength, modulus of rupture (MOR), modulus of elasticity (MOE), water absorption (WA) and thickness swell (TS). The test results were statistically analyzed by using response surface method (RSM) to determine the significant independent variables that influenced MDF properties. A mathematical simulation or response surface models were developed to predict the MDF properties (MOR, MOE, IB, WA and TS). The obtained results showed that MDF density and all interactions between the experimental variables were significant factors that influenced the mechanical properties of MDF. At 8 bar and 14 % resin content, the MDF recorded WA of 83.12 % and TS of 20.2 %. It can be inferred that two parameters (resin content and pressure) had positive effect on physical and mechanical properties of MDF. We concluded that resin content show more significant effects on MDF manufacturing as compared to pressure parameters.  相似文献

Burns and other skin injuries are growing concerns as well as challenges in an era of antimicrobial resistance. Novel treatment options to improve the prevention and eradication of infectious skin biofilm-producing pathogens, while enhancing wound healing, are urgently needed for the timely treatment of infection-prone injuries. Treatment of acute skin injuries requires tailoring of formulation to assure both proper skin retention and the appropriate release of incorporated antimicrobials. The challenge remains to formulate antimicrobials with low water solubility, which often requires carriers as the primary vehicle, followed by a secondary skin-friendly vehicle. We focused on widely used chlorhexidine formulated in the chitosan-infused nanocarriers, chitosomes, incorporated into chitosan hydrogel for improved treatment of skin injuries. To prove our hypothesis, lipid nanocarriers and chitosan-comprising nanocarriers (≈250 nm) with membrane-active antimicrobial chlorhexidine were optimized and incorporated into chitosan hydrogel. The biological and antibacterial effects of both vesicles and a vesicles-in-hydrogel system were evaluated. The chitosomes-in-chitosan hydrogel formulation demonstrated promising physical properties and were proven safe. Additionally, the chitosan-based systems, both chitosomes and chitosan hydrogel, showed an improved antimicrobial effect against S. aureus and S. epidermidis compared to the formulations without chitosan. The novel formulation could serve as a foundation for infection prevention and bacterial eradication in acute wounds.  相似文献

Eugenol (1) and thymol (2) exhibit excellent fungicidal activity against pathogenic yeasts, including isolates resistant to azoles. The rapid irreversible action of compound-1 and compound 2 on fungal cells suggested a membrane-located target for their action. We investigated their effect on H+-ATPase mediated H+-pumping by various Candida species. Both compounds inhibit H+-ATPase activity at their respective MIC values — 500 and 100 μg/ml. Glucose stimulated H+-extrusion was also inhibited significantly by compound 1 and compound 2. Inhibition of H+-ATPase leads to intracellular acidification and cell death. Inhibition of cell growth and H+-efflux by test compounds suggests that their antifungal properties are related to their inhibitory effects on H+-ATPase.  相似文献

Legume plants are an essential component of sustainable farming systems. Phosphorus (P) deficiency is a significant constraint for legume production, especially in nutrient-poor soils of arid and semi-arid regions. In the present study, we conducted a pot experiment to evaluate the effects of a phosphorus-mobilizing plant-growth promoting rhizobacterial strain Bacillus cereus GS6, either alone or combined with phosphate-enriched compost (PEC) on the symbiotic (nodulation-N2 fixation) performance of soybean (Glycine max (L.) Merr.) on an Aridisol. The PEC was produced by composting food waste with addition of single super phosphate. The bacterial strain B. cereus GS6 showed considerable potential for P solubilization and mobilization by releasing carboxylates in insoluble P (rock phosphate)-enriched medium. Inoculation of B. cereus GS6 in combination with PEC application significantly improved nodulation and nodule N2 fixation efficiency. Compared to the control (without B. cereus GS6 and PEC), the combined application of B. cereus GS6 with PEC resulted in significantly higher accumulation of nitrogen (N), P, and potassium (K) in grain, shoot, and nodule. The N:P and P:K ratios in nodules were significantly altered by the application of PEC and B. cereus GS6, which reflected the important roles of P and K in symbiotic performance of soybean. The combined application of PEC and B. cereus GS6 also significantly increased the soil dehydrogenase and phosphomonoesterase activities, as well as the soil available N, P, and K contents. Significant positive relationships were found between soil organic carbon (C) content, dehydrogenase and phosphomonoesterase activities, and available N, P, and K contents. This study suggests that inoculation of P-mobilizing rhizobacteria, such as B. cereus GS6, in combination with PEC application might enhance legume productivity by improving nodulation and nodule N2 fixation efficiency.  相似文献
