Adult Mesenchymal Stem Cells (MSC) are cells that can be defined as multipotent cells able to differentiate into diverse lineages, under appropriate conditions. These cells have been widely used in regenerative medicine, both in preclinical and clinical settings. Initially discovered in bone marrow, MSC can now be isolated from a wide spectrum of adult and foetal tissues. Studies to evaluate the therapeutic potential of these cells are based on their ability to arrive to damaged tissues. In this paper we have done a comparative study analyzing proliferation, surface markers and OCT4, SOX9, RUNX2, PPARG genes expression in MSC cells from Bone marrow (BMMSC) and Adipose tissue (ASC). We also analyzed the role of Stem Cell Factor (SCF) on MSC proliferation and on ASCs metalloproteinases MMP-2, MMP-9 secretion. Healthy dogs were used as BMMSC donors, and ASC were collected from omentum during elective ovariohysterectomy surgery. Both cell types were cultured in IMDM medium with or without SCF, 10% Dog Serum (DS), and incubated at 38 °C with 5% CO2. Growth of BMMSCs and ASCs was exponential until 25–30 days. Flow citometry of MSCs revealed positive results for CD90 and negative for CD34, CD45 and MCH-II. Genes were evaluated by RT-PCR and metalloproteinases by zymografy. Our findings indicate morphological and immunological similarities as well as expression of genes from both origins on analyzed cells. Furthermore, SCF did not affect proliferation of MSCs, however it up-regulated MMP-2 and MMP-9 secretion in ASCs. Two experiments were conducted to evaluate reduced supplementation frequencies for grazing beef cattle in rainy season. In experiment 1, evaluating the nutritional parameters, four rumen-cannulated Nellore bulls (BW = 410 kg) were used. In experiment 2, evaluating animal performance, 48 Nellore bulls (BW = 358 kg) were used. The treatments were as follows: mineral supplement (MS) alone and MS plus protein-energy supplement provided 3×, 5× and 7×/week. Supplementation frequency did not affect (P > 0.05) intake and digestibility. Average daily gain was greater (P < 0.001) to supplementation compared with MS. The supplementation 5×/week resulted in greater weight gain per hectare (9.24) and higher economic returns during the study period (1.64%) compared to other supplementations. Supplementation 5×/week increased animal performance and positively influenced economic returns. 相似文献

Mixtures of wet vegetable wastes (Brassica, carrot or onion) and dry onion waste were composted at 50 °C for 7 days. The incorporation of the raw or composted vegetable waste mixtures into sandy loam, silt and peat soils reduced the viability of sclerotia of S. cepivorum in glasshouse pot bioassays. The reduction in viability was dependent on waste type, rate of incorporation, duration of exposure and soil type. Onion waste was the most effective waste type in reducing sclerotia viability in all three soils. The Brassica and carrot wastes were as effective as the onion waste in silt soil but less effective in sandy loam and peat soil. A 50% w/w incorporation rate of the wastes gave the largest reduction in viability, with an increase in reduction over time. Composted onion waste reduced sclerotia viability under glasshouse and field conditions although the effect was smaller in the field. Composted onion waste incorporated into soil at 50% w/w reduced the incidence of Allium white rot on onion seedlings in glasshouse pot tests. Incidence and control of the disease differed with soil type. The most consistent control was achieved in peat soil whereas no control was observed in silt soil. Incorporation of the waste 2 months prior to sowing or transplanting reduced seedling emergence in sandy loam soil and growth in all three soil types. The potential for field application of composted vegetable wastes as a sustainable method for control of Allium white rot and waste disposal is discussed. 相似文献

Assemblages of plants were studied at 14 sites in northern Patagonia corresponding to localities at which we (Monjeau et al. 1997) earlier studied the relationship between small mammal assemblages and landscape classifications. This allowed us to test predictions that both plants and small mammals correspond to the more inclusive hierarchical landscape divisions but that plants track better than small mammals the less inclusive divisions. Species presence or absence of plants at each locality was used in a series of multivariate analyses and compared by correlation analysis with those generated from small mammal species data. Assemblages of both plants and small mammals corresponded to the upper divisions, which are based on climatic and geomorphological features, but small mammal assemblages did not correspond to the lower divisions of the landscape classifications. Three factors are considered as explanations for the observed differences between plants and small mammals: a) small mammal habitat is determined more by plant growth form than by plant species; b) trophic level differences between the two groups; and c) species pool size affects the resolution of microhabitat correspondence. Our data indicate that both plant assemblages and small mammal assemblages respond to climatic and geomorphological features, which is in contrast to the paradigm that mammal assemblages simply follow plant assemblages. We also attempted to reconcile classification systems in Patagonia by proposing a nomenclatural system based on a hierarchical classification. In the system proposed, ecoregion is the lowest division small mammal assemblages can recognize in Patagonia. Finally, we conclude that the hierarchical nature of landscapes based on a holistic view of environments reflects real entities that are not just the perceptions of landscape ecologists. 相似文献

The use of live animals for educational purposes is an old practice that is still employed in teaching and research institutions. However, there are several objections to this practice, whether for ethical or humanitarian reasons. Surgical techniques teaching using anatomical pieces and/or preserved cadavers promotes greater learning efficiency, provides exercise repetition and increases the confidence and satisfaction of the students when compared to the use of live animals. The current work aimed to analyse the feasibility of using fresh swine urinary bladder and small intestines (jejunum), obtained from slaughterhouses, fixed in 99.8% ethyl alcohol (EA) and preserved in sodium chloride hypersaturated solution (SCHS) at 30%, for 7, 14 and 21 days, as an alternative method for surgical skills training (SST). Swine viscera, fixed in EA and preserved in SCHS, presented a realistic appearance, absence of odour and maintained the viable morphological characteristics during the performance of the operative techniques. Preservation solutions had low cost, were easy to acquire and did not offers risks to human health. Therefore, urinary bladders and small intestines fixed in 99.8% EA for 30 days and maintained in 30% SCHS at different periods were demonstrated as a good viable option as a preservation method for surgical skills training. 相似文献

We determined the prevalence of diseases and pathogens associated with mortality in beef cattle in the State of Rio Grande do Sul, Brazil, based on pathology laboratory submissions. Postmortem examinations were conducted on 1,277 beef cattle that died between 2008 and 2018. Information regarding age, time of the year, breed, and regional location were analyzed statistically. Most cattle were from the surrounding region of Porto Alegre, and 78.7% of the analyzed cases had diagnostic value. The diagnostic category with most cases was infectious and/or parasitic diseases (60%), followed by toxic and toxicoinfectious (25%). Most cases occurred in the fall. Major disease conditions identified included hemoprotozoal infection (18.2%), rabies (8.2%), and plant intoxications by Senecio spp. (8.5%) and Pteridium arachnoideum (4.6%). Hemoprotozoal infection occurred at a higher frequency in young cattle, mainly in animals up to 1 y old. Intoxication by Senecio spp. was more frequent in cattle 2–3 y old. 相似文献

Reducing the antinutritional components of soybean (Glycine max) seed is necessary for producing animal feed with increased metabolizable energy and nutrient availability. The objective of this study was to evaluate marker-assisted and phenotypic selection for the high inorganic phosphorus (Pi) and low stachyose traits controlled by the Lpa and MIPS1 mutations in six breeding populations. Phenotypic selection for low phytate individuals carrying Lpa mutations using Pi was reliable, but the assay should include parental genotypes to construct thresholds before performing selections, as the effect of Lpa on Pi depends on genetic background. Phenotypic selection for low phytate individuals carrying MIPS1 using Pi should be complemented with sugar data because selection based solely on Pi may be inefficient, as this trait is not environmentally stable in MIPS1 germplasm. Marker-assisted selection for simple sequence repeat (SSR) linked to Lpa2 (Satt527, Satt561) depended on the pedigree of the population suggesting that perfect markers (e.g., single nucleotide polymorphism) should become the molecular tool for screening a large number of progenies with high efficiency. 相似文献
