Photoinhibition of photosynthesis and photosynthetic recovery were studied in detached needles of cypress (Cupressus sempervirens L.) Clones 52 and 30 under controlled conditions of high irradiation (about 1900 micromol m(-2) s(-1) for 60 min; HL treatment), followed by 60 min in darkness. The degree of photoinhibition was determined based on the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm), which is a measure of the potential efficiency of photosystem II (PSII), and on electron transport measurements. The Fv/Fm ratio declined in needles of both clones in response to the HL treatment. Minimal fluorescence (Fo) increased in HL-treated needles of both clones. The HL treatment decreased rates of whole-chain and PSII activity of isolated thylakoids more in Clone 52 than in Clone 30. In needles of both clones, PSI activity was less sensitive to photoinhibition than PSII activity. In the subsequent 60-min dark incubation, fast recovery was observed in needles of both clones, with PSII efficiencies reaching similar values to those in non-photoinhibited needles. The artificial exogenous electron donors diphenyl carbazide (DPC), hydroxylamine (NH2OH) and manganese chloride (MnCl2) failed to restore the HL-induced loss of PSII activity in needles of Clone 30, whereas DPC and NH2OH significantly restored PSII activity in photoinhibited needles of Clone 52. Quantification of the PSII reaction center protein D1 and the 33-kDa protein of the water-splitting complex following HL treatment of needles revealed pronounced differences between Clone 52 and Clone 30. The large decrease in PSII activity in HL-treated needles was caused by the marked loss of D1 protein and 33-kDa protein in Clone 30 and Clone 52, respectively.  相似文献

Sewage sludge (SS), a highly heterogeneous semisolid fraction of sewage water (about 1% of the sewage water), contains various amounts of nitrogen (N) and phosphorus (P) as well as trace elements such as cadmium (Cd), lead (Pb), copper (Cu), nickel (Ni) and zinc (Zn) with extremely variable physical and chemical compositions. Application of SS improves soil properties, increases yield and simultaneously increases trace metal content in soil and plants. The difficulty in handling, transporting and applying SS and its adverse effect, especially trace metal content in soil and plant, can be overcome by SS–coir pith pelletization (SSCP) or mixing with sewage sledge–coir pith mixture (SSCM). A study was undertaken to evaluate the prepared SSCM and SSCP (1:1 ratio of SS and coir pith) along with SS on dry matter yield, trace metal content in soil and plant parts. The results showed that increased rates of application of SS or SSCM or SSCP increased the green and dry fodder yield of forage maize. Application of SS as either SSCM or SSCP at 1.2 and 2.4 g pot?1 significantly reduced the trace metal content diethylene triamine pentaacetic acid (DTPA extractable) in soil and plant parts (leaves steam and root) compared to SS application. Therefore, in order to reduce the bioavailability of trace metal in soil and its uptake by plant, application of SSCM or SSCP at 1.2 or 2.4 g pot?1 proved to be a better option than SS application.  相似文献

Monitoring the qualitative and quantitative changes of soil organic carbon (SOC) is very much essential to keep the soil viable and productive for long-term sustainability. Impact of long-term (36 years) enforcement of organic and inorganic nutrient management practices on changes in physio-chemical, microbiological properties and enzymatic activities of flooded rice soil was assessed along with soil carbon pools. The results revealed that the fractions of SOC, microbial biomass carbon, culturable microbial counts and dehydrogenase and β-glucosidase activities increased significantly in green manure amended soil. The population of functional microbial groups, namely denitrifiers and methanotrophs was high in farm yard manure applied soil, while methanogens population was high in green manure application. Higher nitrifier population was evident in inorganic or integrated organic and inorganic fertilizer. Continuous application of recommended levels of chemical fertilizers did not greatly affect biological activities in the soil. Fourier Transform Infrared Spectroscopy analysis of humic acid revealed that organic fertilized soil fractions were more aliphatic with greater diversity than did the inorganic fertilized soil fractions. This study emphasizes the importance of organic manure and underscores the recommended dose of inorganic fertilizer to maintain the soil biological properties in flooded rice soil.  相似文献

Amylose content is a parameter that correlates with the cooking behavior of rice. It is measured at the earliest possible stages of rice improvement programs to enable breeders to build the foundations of appropriate grain quality during cultivar development. Amylose is usually quantified by absorbance of the amylose‐iodine complex. The International Network for Quality Rice (INQR) conducted a survey to determine ways that amylose is measured, reproducibility between laboratories, and sources of variation. Each laboratory measured the amylose content of a set of 17 cultivars of rice. The study shows that five different versions of the iodine binding method are in use. The data show that repeatability was high within laboratories but reproducibility between laboratories was low. The major sources of variability were the way the standard curve was constructed and the iodine binding capacity of the potato amylose used to produce the standard. Reproducibility is much lower between laboratories using a standard curve of potato amylose alone compared with those using calibrated rice cultivars. This study highlights the need to standardize the way amylose is measured, and presents research avenues for doing so.  相似文献

Streptococci are one among the major mastitis pathogens which have a considerable impact on cow health, milk quality, and productivity. The aim of the present study was to investigate the occurrence and virulence characteristics of streptococci from bovine milk and to assess the molecular epidemiology and population structure of the Indian isolates using multilocus sequence typing (MLST) and pulsed-field gel electrophoresis (PFGE). Out of a total of 209 bovine composite milk samples screened from four herds (A–D), 30 Streptococcus spp. were isolated from 29 milk samples. Among the 30 isolates, species-specific PCR and partial 16S rRNA gene sequence analysis identified 17 Streptococcus agalactiae arising from herd A and 13 Streptococcus uberis comprising of 5, 7, and 1 isolates from herds B, C, and D respectively. PCR based screening for virulence genes revealed the presence of the cfb and the pavA genes in 17 and 1 S. agalactiae isolates, respectively. Similarly, in S. uberis isolates, cfu gene was present in six isolates from herd C, the pau A/skc gene in all the isolates from herds B, C, and D, whereas the sua gene was present in four isolates from herd B and the only isolate from herd D. On MLST analysis, all the S. agalactiae isolates were found to be of a novel sequence type (ST), ST-483, reported for the first time and is a single locus variant of the predicted subgroup founder ST-310, while the S. uberis isolates were found to be of three novel sequence types, namely ST-439, ST-474, and ST-475, all reported for the first time. ST-474 was a double locus variant of three different STs of global clonal complex ST-143 considered to be associated with clinical and subclinical mastitis, but ST-439 and ST-475 were singletons. Unique sequence types identified for both S. agalactiae and S. uberis were found to be herd specific. On PFGE analysis, identical or closely related restriction patterns for S. agalactiae ST-483 and S. uberis ST-439 in herds A and B respectively, but an unrelated restriction pattern for S. uberis ST-474 and ST-475 isolates from herds D and C respectively, were obtained. This signifies that the isolates of particular ST may exhibit related PFGE patterns suggesting detection of a faster molecular clock by PFGE than MLST. Since all the isolates of both the species belonged to novel sequence types, their epidemiological significance in global context could not be ascertained, however, evidence suggests that they have uniquely evolved in Indian conditions. Further research would be useful for understanding the role of these pathogens in bovine sub-clinical mastitis and implementing effective control strategies in India.  相似文献

A survey on the endorhizal status of 39 fruit crops of 25 families, indicated that 22 fruit crops had arbuscular mycorrhizal (AM)–, four had dark septate endophyte (DSE)–fungal association, and 13 had dual colonization of AM and DSE fungi. Fruit crops were capable of forming Arum‐, Paris‐, or intermediate‐types of AM morphologies of which intermediate‐type was common. To our knowledge, we report for the first time AM in 10 fruit crops and DSE‐fungal association in 17 fruit crops. The extent of AM‐ and DSE‐fungal colonization ranged from 41% to 98% and < 1% to 89.9%, respectively, in different fruit crops. Arbuscular mycorrhizal–fungal spore numbers in the rhizosphere ranged from 6 to 61 spores per 25 g of soil. Arbuscular mycorrhizal–fungal spores belonging to Acaulospora, Glomus, and Scutellospora were isolated from the rhizosphere soil.  相似文献

A detailed study of electromagnetic shielding effectiveness (EMSE) of woven fabrics made of polyester and stainless steel/polyester blended conductive yarn was presented in this research work. Fabrics with different structures were analyzed and their shielding behavior was reported under different frequencies. Shielding efficiency of fabric was analyzed by vector network analyzer in the frequency range of 300 kHz to 1.5 GHz using coaxial transmission line holder. The effects of different fabric parameters such as weft density, proportion of conductive weft yarn, proportion of stainless steel content, grid openness, weave pattern and number of fabric layers on EMSE of fabrics were studied. The EMSE of fabric was found to be increased with increase in proportion of conductive yarn in the weft way. With increase in overall stainless-steel content in the fabric, the EMSE of fabric was increased. As such weave is considered, it did not have significant effect on EMSE of fabrics. But fabric with lower openness and aperture ratio showed better conducting network, hence better shielding. With increase in number of layers of fabric and ply yarns, EMSE of fabric was increased.  相似文献

Drought resistance is becoming an indispensable character for rice improvement due to the dwindling global water resources. Genetic improvement for drought resistance is achieved through physiological dissection and genetic analysis of independent component traits associated with crop productivity under stress. A subset mapping population of 93 near flowering recombinant inbred lines with uniform phenology was constituted for genetic analysis of reproductive stage drought resistance. The population was phenotyped for 22 physio-morphological traits under two contrasting water regimes imposed at reproductive stage. Broad sense heritabilities of morphological traits were lower under stress than irrigated. Predominant association of plant height, panicle exsertion and harvest index with grain yield were observed under stress. The sustenance of panicle exsertion through maintaining growth during moisture stress was found as a significant trait associated with the grain yield through minimizing spikelet sterility. Selective genotyping was carried out with 23 polymorphic microsatellite markers of the established target genomic regions for drought resistance. The study validated the association of a QTL region on the long arm of chromosome 1 with plant height, panicle length, panicle exsertion, biological yield and stomatal conductance under stress. This region, flanked by markers RM246 and RM315, was known to possess the semi-dwarf gene, sd-1. Role of another major interval lying between RM256 and RM149 on chromosome 8 in defining the drought resistance could be established through identification of QTLs associated with leaf rolling, panicle exsertion, plant height, panicle length, senescence and biological yield under moisture stress condition.
