Grassland is one of the most important terrestrial ecosystems for carbon (C) and nitrogen (N) cycling. However, while CO2 fixation by phototrophic bacteria is relatively well studied, little is known about microbial CO2 fixation without light by chemoautotrophic bacteria in grassland soils. Therefore, in this study, the isotope 14C-CO2 was used to investigate the CO2-fixing process in grassland soils. Soil samples were collected from both fenced and adjacent continuous grazing grassland sites in Inner Mongolia and then incubated for 120 days under dark conditions. Meanwhile, the cbbL genes (red- and green-like) were analyzed to isolate chemoautotrophic bacteria, which are responsible for CO2 fixation. After incubation, 14C was fixed into soil organic carbon (14C-SOC) and microbial biomass carbon (14C-MBC) were found in both the fenced and grazing soils, and the fixation rate of 14C-SOC in the fenced soils (48.55‰) was significantly higher than in the grazing soils (22.11‰). The fixation rate of 14C-MBC in the fenced soils (14.05‰) was higher than in the grazing soils (7.08‰), but the difference was not significant. The red-like cbbL genes could be detected in all the soil samples, but the green-like cbbL genes could not be amplified. A greater number of identified operational taxonomic units were observed in the fenced soils compared with the grazing soils. The chemoautotrophic bacteria were mainly affiliated with Alphaproteobacteria and Actinobacteria. However, Chloroflexi was detected in only the fenced soils. The results suggested that CO2 fixation by chemoautotrophic bacteria might be significant in carbon cycling in grassland. 相似文献

We conducted two feeding experiments to evaluate the effects of supplementation with either cellooligosaccharide or kraft pulp on growth performance in grazing beef calves (Japanese Black) from 4 weeks pre‐weaning to 12 to 16 weeks post‐weaning. In Experiment 1 (20‐week duration), nine calves (2.9‐month‐old females) were assigned to either a control group (CON) or an experimental group (CEL) fed cellooligosaccharide at a rate of 10 g/day mixed with concentrate. Average daily weight gain tended to be greater in CEL than in CON, especially after 1 month of weaning. In Experiment 2 (16‐week duration), 10 calves (2.0‐month‐old females) were assigned to either a control group or an experimental group (KRA) fed kraft pulp at a rate of 10% replacement of total digestible nutrients with concentrate. The proportion of fibrolytic bacteria increased and that of methanogenic Archaea decreased in the rumen microbial community composition of KRA calves in Experiment 2, whereas the decrease in Fibrobacter and Archaea was observed in CEL calves at first 4 weeks in Experiment 1. We conclude that beta‐glucan prebiotic supplementation to grazing calves at pre‐weaning would affect rumen microbial composition and modified rumen fermentation characteristics, leading to a better rumen environment via different means. 相似文献

Feeding dried distillers grains with solubles (DDGS) during the late stage of the fattening period of Holstein steers was studied in regard to the influence on meat quality. Sixteen Holstein steers approximately 18 months old were used in this study. Eight animals were fed commercial concentrated feed for the entire fattening period. The other eight animals were fed 15% DDGS in the concentrated feed for 3 months before slaughtering. The moisture, ether extract and crude protein from both groups was approximately the same. The thiobarbituric acid reactive substance (TBA) value of storage for 7 days at 5°C from the animals fed DDGS showed a tendency to be lower (P = 0.059). The change in the TBA value during storage was also lower for the animals not fed DDGS (P < 0.05). There were no differences in the subcutaneous fat color between the two groups. The a*(measure of redness) and b*(measure of yellowness) of the M. longissimus from the animals fed DDGS showed a tendency to be lower (P = 0.051, 0.070). The fatty acid composition of the M. longissimus, subcutaneous and perirenal fat were not widely influenced by the feeding of DDGS. It is suggested that feeding 15% DDGS during the late stage of the fattening period for Holstein steers reduced the oxidation of the beef. 相似文献

Pseudomonas cichorii is the major causal agent of bacterial rot disease in lettuce, and apoptosis-like programmed cell death is closely associated with disease development. Depletion of cellular ATP and expression of the alternative oxidase gene was observed in lettuce leaves inoculated with P. cichorii suggesting mitochondrial dysfunction. Cytochemical observation showed production of reactive oxygen species in mitochondria of P. cichorii-inoculated lettuce leaf cells. Release of cytochrome c into the cytosol was also induced by inoculation with the bacterium. Superoxide production was observed in the mitochondrial fraction isolated from P. cichorii-infected leaves much more intense than water-treated leaves. Loss of swelling ability was also observed in the mitochondrial fractions following inoculation with P. cichorii. Intriguingly, inhibitors of the mitochondrial respiratory complex III prevented loss of swelling ability, whereas superoxide generation was scarcely affected by inhibitors of mitochondrial permeability transition in the mitochondrial fractions. Inhibitors of the mitochondrial respiratory complex III and mitochondrial permeability transition delayed not only P. cichorii-induced cell death, but also disease development. In contrast, P. cichorii-induced DNA fragmentation was not inhibited in the presence of either type of inhibitor. The findings suggest that mitochondria may play a crucial role in DNA fragmentation-independent cell death pathway(s). 相似文献

An 11-month-old Japanese Black steer with chronic bloat underwent clinical and histological analyses. During the observation period, it showed normal appetite and fecal volume but persistent chronic bloat symptoms. Compared to controls, the steer's feces contained undigested large straws. Necropsy revealed normal rumen, reticulum, and abomasum but a small omasum. The rumen, reticulum, and abomasum mucosa was normal, with well-developed ruminal papillae. However, severe hypoplasia of the omasal laminae was observed along with hypoplasia reticular groove and ruminoreticular fold. The contents of the reticulum, omasum, and abomasums comprised undigested large sized hay particles. The omasum papillae showed no pathological abnormalities. This is a rare case of a steer with chronic bloat probably caused by severe hypoplasia of the omasal laminae. 相似文献

A multigene family encoding HLK effectors is well conserved in all Ralstonia solanacearum strains. The HLK family contains three paralogous members. We constructed single, double, and triple deletion mutants of hlk genes in R. solanacearum strain OE1-1 and characterized HLK effectors by comparing the phenotypes of the strains. HLK effectors exhibited some functional redundancy. While the triple deletion mutant was significantly impaired in virulence on tomato and multiplied less efficiently than the wild type in the leaves, the mutant was as virulent as the wild type on eggplant and tobacco. 相似文献

We conducted two feeding experiments to evaluate the effects of cellooligosaccharide (CE) supplementation on growth performance in grazing beef calves. Calf sex and age and duration of the experimental period differed between the experiments. Experiment 1 (10 weeks) used 5.6‐month‐old castrated males; Experiment 2 (13 weeks) used 3.9‐month‐old females. Eight Japanese Black calves were assigned to either a control group (CON) or an experimental group (CE) fed CE at a rate of 10 g/day mixed with concentrate. Calves were stocked in fields in which Kentucky bluegrass was dominant. In both experiments, average daily gain tended to be greater in CE than in CON, especially in late stages of experiments, but no significant differences in body weight changes were observed. In Experiment 2, final heart girth was larger in CE than in CON calves. We monitored rumen microbial community composition in Experiment 2 and found increases in fibrolytic bacteria and methanogenic archaea in CE calves, but the overall microbial composition did not differ between the groups. Our results suggest that supplementation with CE may positively affect growth performance in weaned calves on pasture, but longer‐term CE supplementation may be required to exhibit the effect. 相似文献
