Black snook, Centropomus nigrescens, have been identified as a promising candidate for aquaculture although, like many of the Centropomid species, high mortality associated with early larval stages presents a significant bottleneck to their commercialization. The digestive capacity of black snook larvae throughout the first 37 d after hatch (d.a.h.) was evaluated by quantifying digestive enzyme activities using biochemical techniques. Results showed that black snook larvae have alkaline proteases at hatching, which are known to be important during the first days of feeding for digestion. Toward the end of the study, acid proteases concentration increased (37 d.a.h.). Enzymes for lipid digestion, pancreatic lipase and bile salt‐activated lipase, were already present in the larvae before exogenous feeding commenced, and their activity increased with age and growth (length). Intracellular digestion, measured as the activity of leucine‐alanine peptidase, was high early on (5 d.a.h.) and decreased as development progressed (next 32 d). In contrast, alkaline phosphatase activity was lowest at first feeding and subsequently increased with age. Overall patterns in enzyme activity suggest the possibility of live feed weaning before 32 d.a.h. if artificial diets can be properly balanced. 相似文献

Piscirickettsia salmonisis the causative bacterial pathogen of piscirickettsiosis, a salmonid disease that causes notable mortalities in the worldwide aquaculture industry. Published research describes the phenotypic traits, virulence factors, pathogenicity and antibiotic‐resistance potential for various P. salmonisstrains. However, evolutionary and genetic information is scarce for P. salmonis. The present study used multilocus sequence typing (MLST) to gain insight into the population structure and evolution of P. salmonis. Forty‐two Chilean P. salmonisisolates, as well as the type strain LF‐89T, were recovered from diseased Salmo salar, Oncorhynchus kisutchand Oncorhynchus mykissfrom two Chilean Regions. MLST assessed the loci sequences of dnaK, efp, fumC, glyA, murG, rpoD and trpB. Bioinformatics analyses established the genetic diversity among P. salmonis isolates (H = 0.5810). A total of 23 sequence types (ST) were identified, 53.48% of which were represented by ST1, ST5 and ST2. Population structure analysis through polymorphism patterns showed few polymorphic sites (218 nucleotides from 4,010 bp), while dN/dS ratio analysis indicated purifying selection for dnaK, epf, fumC, murG, and rpoD but neutral selection for the trpB loci. The standardized index of association indicated strong linkage disequilibrium, suggesting clonal population structure. However, recombination events were detected in a group of seven isolates. Findings included genogroups homologous to the LF‐89T and EM‐90 strains, as well as a seven‐isolate hybrid genogroup recovered from both assessed regions (three O. mykiss and four S. salar isolates). The presented MLST scheme has comparative potential, with promising applications in studying distinct P. salmonis isolates (e.g., from different hosts, farms, geographical areas) and in understanding the epidemiology of this pathogen. 相似文献

Many estimators and algorithms have been developed to infer the genealogical relationships from molecular marker data when there is no pedigree information. Most pairwise methods provide estimates in a continuous range that needs to be converted into genealogical relationships (namely full-sibs, half-sibs and unrelated) if there is a previous knowledge of the population structure. Transformations are usually based on arbitrary thresholds, but the possibility of correcting the coancestry estimates via explicit pedigree reconstruction methods has been suggested. Using molecular data for ten highly polymorphic microsatellite loci on a population of turbot (Scophthalmus maximus) with a known genealogy, the behaviour of four pairwise marker-based coancestry estimators and the molecular coancestry has been evaluated. The population consisted on 138 families with 4 full-sib individuals each and one family with 8 full-sib individuals (up to 15 half-sib families were also present due to the sharing of parents between some of the full-sibs families). Our results suggested that transforming the pairwise estimators and the molecular coancestry to family relationships through the establishment of thresholds performs slightly better than the explicit pedigree reconstruction method, when accuracy is measured in a pairwise basis. However, if joint relationships between more than two individuals were tested at a time, the threshold methods led to a high percentage of incongruous triads of full-sib individuals, with Mendelian incompatibilities appearing in congruous full-sib families (more than 70% and 60% in our study, respectively). The explicit pedigree reconstruction approach, due to its nature, is free from such problems. Therefore, the pedigree reconstruction approach seems to be a valuable tool to provide a congruent and compatible pedigree when the pairwise marker-based coancestry matrices or the molecular coancestry need to be transformed. 相似文献

The Mexican silverside, Menidia estor, is a species with great regional importance and with very high prices in local markets. Unfortunately, due to high fisheries pressure, environmental degradation and pollution, the species has become endangered. Recently, there has been much progress in the biotechnology of this species, aimed at its culture, and the present paper describes the advances in feeding and nutrition of those important fish. M. estor is a stomachless, zooplantophagous fish, which also occasionally feeds on small fish and crustaceans in the adult stage. Studies on the digestive enzymatic activities show high proteolytic capacity and a late or different model of digestive maturation from that described for marine stomach fish. Nutritional studies on M. estor have shown that juveniles have dietary requirements of about 400 g kg?1 protein and 80 mg kg?1 vitamin C. The upper level of dietary carbohydrate for good growth and survival for juveniles is about 150 g kg?1. Based on the high docosahexaenoic acid (DHA) content in flesh when fed on diets low in DHA, it is believed that the species has the capacity to biosynthetize >20‐carbon fatty acids from 18‐carbon fatty acids. The high levels of DHA in flesh makes this fish a very significant potential component of human nutrition. Using these findings the first practical diets for commercial culture of the species have been developed. 相似文献

Flower initiation date and readiness to flowering in buds of different age were studied in ‘Fino de Jete’ cherimoya (Annona cherimola) cultivar in order to establish the limits for the manipulation of its flowering date. Flower initiation was analyzed by light and scanning electron microscopy (SEM) collecting axillary buds from May to the following February, whereas the bud readiness to produce perfect flowers was determined by forcing buds of different age to sprout by means of leaf removal and tipping the new growth. SEM images confirm that cherimoya buds are differentiated into flowers almost a year before blooming. In this regard, axillary buds have already formed the sepals when the subtending leaf has just begun unfolding (week 0), while the petals are clearly visible in 1-week-old buds. Sectioning of paraffin-embedded buds illustrate that cherimoya buds are in fact a bud complex that 1 week after its inception comprises 4–5 buds of different size of which the two largest ones are reproductive, while the 2–3 smallest buds often remain undifferentiated at that time. The high capacity of flowering expressed by young buds that have been forced to grow proves that cherimoya meristems are early competent for flowering. No differences in fertility or in the time needed to reach anthesis after leaf removal were found among buds of different ages. Node position had no effect on bud break and flowering potential. The early flower initiation in cherimoya deduced from this work opens a wide temporal window for the experimental manipulation of flowering and harvest dates in this crop. 相似文献

The effect of semi-continuous culture on the nutritional value of microalgae was tested in the rotifer Brachionus plicatilis in short-term enrichment experiments. Isochrysis aff. galbana clone T-ISO was cultured semi-continuously with renewal rates from 10 to 50% of the volume of the culture per day and used to feed the rotifers. After 24 h, dramatic differences in dry weight and protein, lipid and carbohydrate contents were observed in the rotifers depending on the renewal rate applied to the microalgal culture. Rotifers fed T-ISO cultured with low renewal rates showed low dry weight and organic content, whereas rotifers fed microalgae from nutrient-sufficient, high renewal rate cultures showed higher dry weight and increases up to 60% in protein, 35% in lipid and 100% in carbohydrate contents. Feed conversion rate (FCR) and organic FCR decreased with increasing renewal rates, indicating a more efficient assimilation of the microalgal biomass obtained at high growth rates. The fatty acid profile of rotifers reflected that of T-ISO, with maximum content of polyunsaturated fatty acids (PUFAs), n-3 fatty acids and docosahexaenoic acid (DHA) being found in the rotifers fed microalgae from the renewal rate of 40%. Results demonstrate that the biochemical composition of B. plicatilis is strongly modified through the use of semi-continuous cultures of microalgae in short-term enrichment processes. In the past two decades spaces for user participation have been opened within water governance structures at many scales. In this contribution, based on a case study of the Provincial Water Users Federation Interjuntas‐Chimborazo in the Ecuadorian Highlands, I explore how and why formal participation of water users in itself is problematic in terms of democracy. The case shows that for organised peasant water users to work on more democratic water governance, the creation of upscaled federations, alliances, networks and sometimes street protests is crucial to open up spaces in which their interests are represented. This suggests that democracy is not merely about participation, but more importantly, it is about the politics of how democracy is made through conflicts, protests, negotiations and the creation of strategic alliances that challenge the structures and processes through which decisions are taken. 相似文献
