In the adult brain, neuroblasts born in the subventricular zone migrate from the walls of the lateral ventricles to the olfactory bulb. How do these cells orient over such a long distance and through complex territories? Here we show that neuroblast migration parallels cerebrospinal fluid (CSF) flow. Beating of ependymal cilia is required for normal CSF flow, concentration gradient formation of CSF guidance molecules, and directional migration of neuroblasts. Results suggest that polarized epithelial cells contribute important vectorial information for guidance of young, migrating neurons. 相似文献

The generation of pluripotent stem cells from an individual patient would enable the large-scale production of the cell types affected by that patient's disease. These cells could in turn be used for disease modeling, drug discovery, and eventually autologous cell replacement therapies. Although recent studies have demonstrated the reprogramming of human fibroblasts to a pluripotent state, it remains unclear whether these induced pluripotent stem (iPS) cells can be produced directly from elderly patients with chronic disease. We have generated iPS cells from an 82-year-old woman diagnosed with a familial form of amyotrophic lateral sclerosis (ALS). These patient-specific iPS cells possess properties of embryonic stem cells and were successfully directed to differentiate into motor neurons, the cell type destroyed in ALS. 相似文献

Four lichen species, Parmelia sulcata Taylor, Hypogymnia physodes (L.) Nyl., Cetraria islandica (L.)Ach., and Bryoria fuscescens (Gyelnik) Brodo andHawksworth were exposed during autumn and winter at differentsites of the Erzgebirge (Ore Mountains), the Fichtelgebirge andcontrol sites. All lichens tested became visibly damaged withtime. Thallus bleaching started from the edges and went on tothe centre of the thallus. Sites of facilitated gas exchangelike the soralia of P. sulcata and the pseudocyphelles ofC. islandica became preferentially bleached. The sulfateconcentration increased with exposure time reaching 200% ofunpolluted controls. In contrast to coniferous trees (e.g. Picea abies), further exposure lead to a reduction in the S-concentration in the lichens, as sulfate and otherintracellular metabolites were leached from the thalli due tomembrane damage. The changes in the K-concentration proved tobe an excellent measure for membrane leakiness; it wascorrelated with time of exposure and with SO2 concentrations at the different sites. Photosynthetic capacityand respiration were also strongly affected. Depending on theSO2-dose, the Bryoria species were unable tophotosynthesize as early as 4–8 weeks after exposure, whereasCetraria and Hypogymnia showed clear reduction intheir ability to photoreduce CO2 within 8–10 weeks ofexposure in the field. Parmelia sulcata was found to bethe most tolerant species. After 3–4 months, photosynthesis wasreduced by only 30%. The bioindicative value of theseobservations is discussed. 相似文献

The duration of the occultation was 11 minutes, 4.4 seconds, and mid-occultation occurred at 14 hours, 25 minutes, 9 seconds U.T. ObjectiveTo establish an accurate anaesthetic dose for chemical restraint of African mole-rats using ketamine and xylazine.Study designProspective nonrandomized laboratory study.AnimalsSixteen adult Ansell’s mole-rats (Fukomys anselli) and eight giant mole-rats (F. mechowii).MethodsFukomys anselli of different ages, sexes and reproductive status were systematically anaesthetized starting with an intramuscular injection of ketamine (2.5 mg kg−1) and increasing the doses in steps of 0.5 mg kg−1 until loss of the righting reflex (LRR) was observed. Xylazine was added to a constant dose of ketamine, starting at 0.5 mg kg−1 that was increased by 0.5 mg kg−1 in further trials. Once an effective combination was established and evaluated in F. anselli, it was also tested in F. mechowii. Heart and respiratory rates and rectal temperatures were measured during anaesthesia. anova for repeated measures and Student’s t-test were used to compare means.ResultsChemical restraint was accomplished at a dose of 6 mg kg−1 ketamine combined with 2.5 mg kg−1 xylazine. LRR lasted on average mean 56 ± SD 19 minutes (F. anselli) and 140 ± 41 minutes (F. mechowii). Loss of pedal withdrawal reflex (LPR) lasted for 20 ± 15 minutes (F. anselli) and for 29 ± 2 minutes (F. mechowii), respectively. All animals recovered satisfactorily. Heart and respiratory rates were stable during anaesthesia, but rectal temperature fell significantly in F. mechowii after losing the righting reflex (LRR) from T1 (32.6 ± 0.6 °C) to T3 (30.4 ± 0.9 °C).Conclusions and Clinical relevanceAfrican mole-rats (Bathyergidae) live in closed burrow systems under particular conditions (hypercapnia, hypoxia, stable temperature, humidity, darkness) and show several physiological adaptations. Injectable anaesthetics in the dose rates used in other rodents are not appropriate for use in these subterranean species. Here, a reliable protocol for chemical restraint is provided. 相似文献

Loading of the lithosphere of Mars by the Tharsis rise explains much of the global shape and long-wavelength gravity field of the planet, including a ring of negative gravity anomalies and a topographic trough around Tharsis, as well as gravity anomaly and topographic highs centered in Arabia Terra and extending northward toward Utopia. The Tharsis-induced trough and antipodal high were largely in place by the end of the Noachian Epoch and exerted control on the location and orientation of valley networks. The release of carbon dioxide and water accompanying the emplacement of approximately 3 x 10(8) cubic kilometers of Tharsis magmas may have sustained a warmer climate than at present, enabling the formation of ancient valley networks and fluvial landscape denudation in and adjacent to the large-scale trough. 相似文献
