Microsatellite (SSR) polymorphism was assessed across 90 diploid Avena strigosa Schreb. and tetraploid Avena barbata Pott ex Link accessions obtained from the USDA-ARS National Small Grains Collection using 105 genomic SSRs. Eleven polymorphic SSRs that detected 69 different alleles were identified and used to genotype the 90 accessions, which were chosen from a larger set of 385 accessions based on geographical source-diversity and variable reaction responses to five Australian pathotypes of the crown rust pathogen Puccinia coronata Corda f. sp. avenae Eriks. Eight diploid and eight tetraploid clades were identified among the 90 accessions. Diploid accessions displayed the lowest genetic diversity, with all accessions being at least 86 % similar, and included accessions from countries in the Americas such as Canada, USA, Argentina, Uruguay and Brazil, and European accessions from France, Romania and Poland. Although both species formed distinct clusters in the dendrogram, a few instances of diploids showing high similarity with tetraploids and vice versa were observed. An AMOVA analysis revealed 86 % of the total genetic variation to be distributed within the two oat species, while between-species differences accounted for only 14 %. Heterozygosity (H) index values of 0.32 and 0.40 were obtained for diploids and tetraploids respectively. Our study effectively differentiated A. strigosa and A. barbata, and identified 11 SSRs suitable for future characterisation of accessions of the two species.  相似文献

Summary To elaborate on off-flavour development in dehydrated potato granules, lipids in subcellular particles and membrane systems of the tuber were investigated. Lipid acyl-hydrolase and lipoxygenase activities were suppressed in tuber homogenates by a buffer isolation medium of pH 7.8 containing nupercaine which minimized the breakdown of phospho- and galactolipids. Phospholipid, glycolipid and neutral lipid constitutents, their fatty acid composition, and unsaturation ratios were reported for amyloplasts, cell wall, microsomes, mitochondria, peroxisomes, and plasmalemma.
Zusammenfassung Analysen von Kartoffelknollen wurden durch lipidabbauende Enzyme, Lipidacylhydrolase (LAH) und Lipoxygenase (LOX) behindert. Die LOX-Aktivit?t in handelsüblicherzugten Kartoffeln war hoch (Tabelle 1) und konnte durch gew?hnliche Inhibitoren nicht beeinflusst werden (Tabelle 2). Die LAH-Aktivit?t, die mit Phospholipiden (PL) und Galactolipiden als Substrate hoch war, wurde durch Nupercaine in verschiedenem Ausmass unterdrückt (Tabelle 3), dessen Weglassung w?hrend der Lipidanalyse von Kartoffelmembranen und subzellularen Partikeln hohe Lipidverluste verursachen k?nnte (Tabelle 4). Lipidergebnisse für mikrosomale Pr?parate, Mitochondrien, Peroxisomen, Plasmalemmen und St?rkek?rner (Tabellen 5–9) wurden wiedergegeben: nach Isolation bei pH 7,2, wenn die LOX-Aktivit?t nicht gehemmt war (Verfahren A); in einer Puffersubstanz von pH 7,8 ohne Nupercaine, wenn wohl die Aktivit?t von LOX unterdrückt war, nicht aber jene von LAH (Verfahren B); und bei pH 7,8 mit Nupercaine, wenn die Aktivit?t beider Enzyme unterdrückt war (Verfahren C). In den meisten Analysen zeigte die Lipidzusammensetzung, wenn die Aktivit?t beider Enzyme nicht unterdrückt wurde, eine entsprechende Anreicherung von PL, Sterollipiden und freien Fetts?uren (FFA) und ihrer Abbauprodukte sowie eine Abnahme im Gehalt an Galactolipiden. Kleine Mengen von Lipiden wurden in Form von Lipidpartikeln und verbunden mit Knollenzellw?nden gefunden (Tabelle 10). Die Fetts?urezusammensetzung des Gesamtlipids und einige besondere Lipidbestandteile und ihr Nichts?ttigungsverh?ltnis (UR) wurden nur für Verfahren C berechnet. Wenn dieses Verfahren weggelassen wurde, wie dargestellt mit Hilfe der St?rkelipid-Zusammensetzung, ergab sich nicht nur ein Verlust von Membran-PL und Galactolipiden. Fast die H?lfte der zurückgebliebenen neutralen Lipidfraktion war in Form von FFA in verschiedenen Graden oxydiert (Tabelle 9). Das UR der Gesamtlipide (TL) schwankte zwischen 0,4 und 2,9. Es war niedrig in der Zellwand, h?her im Plasmalemma und am h?chsten innerhalb der Zytoplasmamembranen. Das UR nahm dann bei den Vacuolen mit St?rkek?rnern ab. Die Bedeutung der Ergebnisse für die Bestimmung der Funktion von Lipiden in einem handelsüblichen Verfahren zur Herstellung von Püreepulver wirdt diskutiert.

Résumé La dégradation enzymatique des lipides par la lipide acyl hydrolase (LAH) et par la lipoxygénase (LOX) génait l’analyse biochimique des tubercules de pomme de terre. L’activité de la LOX était importante dans les pommes de terre cultivées pour la commercialisation (tableau 1) et n’était pas affectée par les inhibiteurs courants (tableau 2). L’activité de la LAH, qui était forte avec les phospholipides (PL) et les galactolipides, était inhibée sur différents substrats par la nuperca?ne (tableau 3); l’absence de cette dernière durant l’analyse de la membrane et des organites intracellulaires du tubercule causait de fortes pertes en lipides (tableau 4). Les résultats obtenus avec les lipides des préparations de microsomes, mitochondries, peroxysomes, plasmalemme et de grains d’ amidon (tableaux 5 à 9) sont exposés; après isolement à pH 7,2 sans inhibition de l’activité de la LOX (procédé A); dans une solution tampon à pH 7,8 sans nuperca?ne alors que l’activité de la LOX est inhibée mais non celle de la LAH (procédé B); et à pH 7,8 avec nuperca?ne et inhibition de l’activité des deux enzymes (procédé C). Dans le cas où l’activité des deux enzymes n’était pas inhibée, la composition en lipides montrait un enrichissement relatif en PL, stérols, acides gras libres (FFA) et en leurs produits de dégradation ainsi qu’une diminution de la teneur en galactolipides. Une petite quantité de lipides était retrouvée sous la forme de particules lipidiques associées aux membranes cellulaires (tableau 10). L’étude de la composition en acides gras des lipides totaux et de quelques constituants lipidiques ainsi que la mesure de leur taux d’insaturation (U.R.) ont été effectuées seulement pour le procédé C. Quand ce procédé n’était pas employé, comme l’illustre la composition en lipides de l’amidon, il y avait non seulement une perte de PL et galactolipides membranaires mais la moitié de la fraction lipidique neutre retenue se trouvait sous la forme de FFA oxydés à différents degrés (tableau 9). L’U.R. des lipides totaux (TL) variait de 0,4 à 2,9; il était bas dans les membranes cellulaires, plus élevé dans le plasmalemme et le plus haut dans les membranes cytoplasmiques. L’U.R. diminuait dans les vacuoles contenant des grains d’amidon. L’intérêt de cette découverte pour la détermination des lipides au cours de la fabrication industrielle de granules de pomme de terre est discuté.

The influence of breed on fattening performance, slaughter and meat quality traits was studied in Awassi and Morkaraman male lambs at approximately 8 months of age. All of the groups were fed a diet consisting of concentrate mixture offered ad libitum, and 300 g of grass hay per lamb per day during 60-day fattening period. Initial live weight, final live weight, daily weight gain and feed conversion efficiency (concentrate and hay consumption for 1 kg of live weight gain) were 39.63 kg, 55.08 kg, 0.258 kg and 6.37 for Awassi; 40.54 kg, 55.58 kg, 0.234 kg and 6.77 for Morkaraman, respectively. The effect of breed on fattening performance and slaughter traits except for LD area was not significant. In addition, meat colour parameters (L = lightness, a = redness, b = yellowness, H = hue angle and C = chroma), pH values, drip loss and sensory attributes were not affected by breed in present study. A significant muscle effect was observed for instrumental measurements of some meat quality characteristics as meat colour parameters (L, a and C), collagen, drip loss, pH, moisture and protein content, WBS, and for some sensory attributes (tenderness, juiciness, acceptability and number of chewing). Results of this study indicate that fattening performance, slaughter and meat quality traits were similar between Awassi and Morkaraman male lambs.  相似文献

Nanocomposites of high-density polyethylene/linear low-density polyethylene (HDPE/LLDPE) filled with untreated and surface treated nano-calcium carbonate (nCC) were prepared. The influence of isopropyl tri-(dioctylpyrophosphato) titanate (JN114) treatment of nCC on the morphology, mechanical, crystallization and flow properties of the nanocomposites were studied. The results of scanning electron microscopy (SEM) showed that JN114 treated nCC was better dispersion in the matrix than the untreated one. A fine dispersion of the treated nanoparticles in the nanocomposites was observed by transmission electron microscopy (TEM). The FTIR spectrum analysis revealed that the JN114 could change the surface properties of nCC, resulting in greater hydrophobicity of the surface and enhanced compatibility with nonpolar matrices. The tensile elastic modulus (E c ) and Izod impact strength (SIC) of nanocomposites increased with the increasing of nCC content while tensile fracture strength (σ b ) decreased. The JN114 treated nanocomposites had superior mechanical properties to those of the untreated ones. The compatibility of these nanocomposites was examined by DSC to estimate melting point (T m ) and crystallization temperature (T c ). Furthermore, the melt flow index (MFI) of the nanocomposite materials were measured. It was found that the MFI decreased with the addition of weight fraction of the nCC particles.  相似文献

Coir fibers were modified with methyl acrylate (MA) mixed with methanol (MeOH) under thermal curing method at different temperatures (40?C80 °C) for different curing times (20?C60 min). A series of solutions of different concentrations of MA in methanol along with 2 % benzoyl peroxide, were prepared. Monomer concentration, curing temperature, and curing time were optimized with the extent of grafting of monomer and mechanical properties of cured fiber and found to be 30 % MA, 60 °C and 40 min curing time registered as better performance (Grafting (Gr) = 5.7 %, tensile strength (TS) = 72 %, elongation at break (Eb) = 88 %) than those of untreated fiber. For further improvement of the properties, untreated coir fibers were pretreated with gamma and UV radiations at different doses and then pretreated fibers were soaked in the optimized monomer and cured under optimum conditions. Coir fiber pretreated with UV radiation and grafted with optimized monomer showed the best properties such as Gr (7.12 %), TS (132 %), and Eb (153 %) over raw fiber. Water uptake and simulated weathering test of untreated and treated coir fibers were studied.  相似文献

Oat stem rust, caused by Puccinia graminis f. sp. avenae (Pga), is one of the most severe diseases of oats worldwide. Population studies are scarce for this pathogen, mainly due to the lack of polymorphic molecular markers suitable for genetic analysis. In this study, an Australian Pga isolate was sequenced, the abundance of simple sequence repeats (SSRs) was determined and PCR‐based polymorphic markers suitable for genetic diversity analysis were developed. The amplification of 194 primer pairs was initially assessed using a set of 12 isolates of different cereal rust species and their formae speciales. A high frequency of cross‐species amplification was observed for most markers; however, 36 SSRs were diagnostic for P. graminis only. A subset of 19 genome‐derived SSRs were deemed useful for genetic diversity analysis of Pga and were assessed on 66 Pga isolates from Australia, Brazil and Sweden. Brazilian and Australian isolates were characterized by one and two predominant clonal lineages, respectively. In contrast, the Swedish isolates, previously shown to undergo sexual recombination, were highly diverse (nine distinct genotypes out of 10 isolates) and divided into two subpopulations. The genome‐derived SSR markers developed in this study were well suited to the population studies undertaken, and have diagnostic capabilities that should aid in the identification of unknown rust pathogen species.  相似文献

Jute fabrics (hessian cloth) reinforced polypropylene (PP) matrix composites (45 wt% fiber) were fabricated by compression molding. Jute fabrics were treated with 2-hydroxyethyl methacrylate (HEMA) using ultraviolet radiation in order to improve the mechanical properties of the composites. Concentration of HEMA, soaking time and radiation dose were optimized. It was found that 15% HEMA in methanol along with photoinitiator Darocur-1173 (2 %), 10 min soaking time and 20th pass of radiation rendered better performance. Urea of different concentrations (0.5–2 %) was incorporated with 15 % HEMA to monitor its effect on the properties and 1 % urea revealed the best results. For the improvement of the properties, jute fabrics were treated with potassium permanganate (KMnO4) solution in acetone of different concentrations (0.02, 0.03, 0.05, and 0.5 %) at different soaking times (1, 2, 3, and 5 min) before the composite fabrication. Optimized jute fabrics (jute fabrics treated with 0.03 % KMnO4) were again treated with HEMA (15 %) solution along with urea (1 %) and promising improvement of mechanical properties of the composites was observed. Scanning electron microscopy, water uptake, soil degradation and thermal aging of the treated and untreated composites were also performed.  相似文献

Jute yarns were cured with acrylic acid (AA) and phosphoric acid (PA) using UV radiation in order to prepare selective ion adsorbent. A series of formulations were prepared in methanol containing varying percentages of (10–70 %) of AA and 2 % photo-initiator (PI) (Darocur-4043). jute yarns were soaked in this formulation for various soaking times (10–30 min) and cured under UV radiation of different intensities (20–50 UV passes). Concentration of AA, soaking time and intensity of UV radiation were optimized based on polymer loading (PL). The maximum PL (21 %) was observed for 50 % AA solution for 20 min soaking time at 40 UV passes. Various formulations were prepared using 5–15 % (w/w) of phosphoric acid, 50 % AA and 2 % PI in methanol. Then jute yarns were soaked in this solution for 20 min and irradiated at 40th UV pass. The concentration of PA was again optimized for maximum PL. It was found that the formulation containing 10 % PA, 50 % AA, 38 % methanol and 2 % PI showed 70.95 % of PL. To investigate the adsorbent behavior, a 10 ppm CuSO4 solution was prepared and then grafted yarns were soaked in the solutions for 30–300 min at different conditions. After withdrawing the yarns, the remaining copper in the solution were measured by atomic absorption spectrophotometer (AAS). It was revealed that copper was successfully removed by using the grafted jute yarns.  相似文献

The main objective of this research was to study the effect of fiber content variation and stearic acid (SA) treatment on the fundamental properties of unidirectional coir fiber (CF) reinforced polypropylene (PP) composites. Several percentages of filler contents were used (10–40 wt %) in order to gain insights into the effect of filler content on the properties of the composites. Coir/PP composites were fabricated by compression molding, and the properties of composites were studied by physico-mechanical and thermal properties. The results from mechanical properties such as tensile strength (TS), tensile modulus (TM) and impact strength (IS) of the CF/PP composites were found to be increased with increasing fiber content, reached an optimum and thereafter decreased with further increase in fiber content. Treatment of the coir with SA as the coupling agent enhanced the mechanical properties, crystallization temperature and crystallinity of virgin PP and water desorption of the resulting composites, resulting from the improved adhesion between the CF and PP matrix. Scanning electron micrographs (SEM) of the tensile fractured samples showed improved adhesion between fiber and matrix upon treatment with SA. Interfacial shear strength (IFSS) of the composites was measured by single fiber fragmentation test (SFFT).  相似文献

Two new races of the wheat (Triticum aestivum L.) stem rust pathogen, representing the fifth and sixth variants described within the Ug99 lineage, were detected in South Africa. Races TTKSP and PTKST (North American notation) were detected in 2007 and 2009, respectively. Except for Sr24 virulence, race TTKSP is phenotypically identical to TTKSF, a commonly detected race of Puccinia graminis f. sp. tritici (Pgt) in South Africa. PTKST is similar to TTKSP except that it produces a lower infection type on the Sr21 differential and has virulence for Sr31. Simple sequence repeat (SSR) analysis confirmed the genetic relationship amongst TTKSF, TTKSP, PTKST and TTKSK (Ug99). TTKSK, PTKST and TTKSF grouped together with 99% similarity, while sharing 88% genetic resemblance with TTKSP. These four races in turn shared only 31% similarity with other South African races. It is proposed that both TTKSP and PTKST represent exotic introductions of Pgt to South Africa.  相似文献
