A new instrument called the tunneling microscope has recently been developed that is capable of generating real-space images of surfaces showing atomic structure. These images offer a new view of matter on an atomic scale. The current capabilities and limitations and the physics involved in the technique are discussed along with specific results from a study of silicon crystal surfaces.  相似文献

Structural transformations on elemental semiconductor surfaces typically occur above several hundred degrees Celsius, and the atomic motions involved are extremely rapid and difficult to observe. However, on the (111) surface of germanium, a few lead atoms catalyze atomic motions so that they can be observed with a tunneling microscope at temperatures below 80 degrees C. Mass transport and structural changes are caused by the creation and propagation of both vacancy-like and interstitial-like point defects within the crystal surface. The availability of dangling bonds on the surface is critical. A detailed atomic model for the observed motions has been developed and is used to explain the structural phase transition Ge(111)-c(2x8) <--> 1x1, which occurs near 300 degrees C.  相似文献

The microscopic mechanism of the matching effect in a superconductor, which manifested itself as the production of peaks or cusps in the critical current at specific values of the applied magnetic field, was investigated with Lorentz microscopy to allow direct observation of the behavior of vortices in a niobium thin film having a regular array of artificial defects. Vortices were observed to form regular and consequently rigid lattices at the matching magnetic field, at its multiples, and at its fractions. The dynamic observation furthermore revealed that vortices were most difficult to move at the matching field, whereas excess vortices moved easily.  相似文献

A total of 350 nymphs of the common tick Ixodes ricinus (Linnaeus, 1758) were collected in an endemic focus of Lyme borreliosis (South Moravia, Czech Republic) and examined for the presence of the protozoan Babesia microti (Fran?a, 1909) by polymerase chain reaction (PCR), using primers specific for the B. microti gene encoding small subunit rRNA. The assay revealed five positive pools (out of 70 pools examined); the corresponding prevalence rate was about 1.5%. Sequence analysis of the PCR products confirmed their 100% homology with that of B. microti. The study represents the first evidence of B. microti in ixodid ticks in the Czech Republic.  相似文献

Bacterial diversity in peat bogs of the karst origin was studied with the use of traditional inoculation methods and modern molecular techniques. Representatives of phylogenetic groups of Proteobacteria and Acidibacteria predominated among the domain of Bacteria. Aquaspirillum and Bacillus predominated in the saprotrophic bacterial complex. The amount of actinobacteria in the studied peat samples was higher than that in typical oligotrophic peat bogs.  相似文献

This review presents an analysis of literature data and original studies by the authors aimed at revealing the factors inhibiting the destruction of high-moor (oligotrophic) peat. Each of the ecological factors that prevent the decomposition of the high-moor peat by different groups of microorganisms is considered. The acid reaction, low temperatures, and lack of nutrients were found not to be the primary factors inhibiting the destruction of the peat. The limited content of oxygen in the peatbogs leads to a drastic decrease in the number of mycelial microorganisms and a reduction of the activity of hydrolytic and oxidizing enzymes. The main factor inhibiting the decomposition of sphagnum is its mechanical and chemical stability, since animals crushing sphagnum are absent in the soil, and this moss has polysaccharides of special composition. The toxicity of phenol compounds, which is manifested under the aerobic conditions, prevents the activity of all the hydrolytic enzymes. This is the main reason for the slow decomposition of sphagnum peat and the long-term preservation of the residues of bodies and food in high-moor peatlands.  相似文献

Lyme borreliosis (LB) is a serious infectious disease of humans and some domestic animals in temperate regions of the Northern Hemisphere. It is caused by certain spirochetes in the Borrelia burgdorferi sensu lato (s.l.) species complex. The complex consists of 11 species (genospecies). Borrelia burgdorferi sensu stricto (s.s.), Borrelia garinii and Borrelia afzelii are the major agents of human disease. Borrelia burgdorferi s.l. species are transmitted mainly by ticks belonging to the Ixodes ricinus species complex plus a few additional species not currently assigned to the complex. B. burgdorferi infections may produce an acute or chronic disease with a wide array of clinical symptoms such as erythema migrans (EM), carditis, arthritis, neuroborreliosis, and acrodermatitis chronica atrophicans (ACA). Differences in LB spirochetes 'genospecies' and strains/isolates determine the occurrence and severity of this multi-system disease. Accurate and reliable identification of the LB spirochetes in ticks as well as knowledge of their prevalence are essential for prevention against the disease and development of an effective vaccine. An overview of the knowledge of molecular factors with emphasis on potential protein-carbohydrate interactions in the tick-borrelia system is the main focus of this review.  相似文献

This study aimed to evaluate various concentrations of egg yolk (5, 10, or 20%) in combination with different concentrations of glycerol (3% or 6%) added to a Tris‐based extender on the post‐thaw characteristics of sperm obtained from Tayassu tajacu. For this purpose, semen from 10 sexually male mature collared peccaries was collected by electroejaculation and evaluated for sperm motility, vigour, viability, morphology and functional membrane integrity. The ejaculates were initially extended in Tris‐fructose plus egg yolk (5%, 10% or 20%). After cooling, the semen was added to Tris‐egg yolk plus glycerol (6% or 12%), resulting in a final concentration of 3% or 6% glycerol of the extender. Straws were frozen using liquid nitrogen and thawed in a water bath at 37°C for 30 s. The frozen–thawed semen was evaluated as reported for fresh semen. After thawing, a significant decrease was verified for sperm motility and vigour, for all the samples in comparison with fresh semen. However, no differences were evidenced among treatments for any sperm characteristics evaluated (p > 0.05), except for the combination between 10% egg yolk and 6% glycerol, which provided the worst preservation of functional membrane integrity (p < 0.05). The interactions between higher concentrations of egg yolk (20%) and glycerol (6%) and also between lower concentrations of the same substances (5% egg yolk and 3% glycerol) added to the Tris‐based extender negatively affected the preservation of the normal sperm morphology after thawing (p < 0.05). In conclusion, the use of Tris‐based extender added to 10% or 20% egg yolk plus 3% glycerol is recommended for effective sperm cryopreservation in collared peccaries.  相似文献
