AIMS: To determine the pharmacokinetics, and anaesthetic and sedative effects of alfaxalone after I/V and I/M administration to cats.

METHODS: Six European shorthair cats, three males and three females, with a mean weight of 4.21 (SD 0.53) kg and aged 3.8 (SD 0.9) years were enrolled in this crossover, two–treatment, two-period study. Alfaxalone at a dose of 5?mg/kg was administered either I/V or I/M. Blood samples were collected between 2–480 minutes after drug administration and analysed for concentrations of alfaxalone by HPLC. The plasma concentration-time curves were analysed by non-compartmental analysis. Sedation scores were evaluated between 5–120 minutes after drug administration using a numerical rating scale (from 0–18). Intervals from drug administration to sit, sternal and lateral recumbency during the induction phase, and to head-lift, sternal recumbency and standing position during recovery were recorded.

RESULTS: The mean half-life and mean residence time of alfaxalone were longer after I/M (1.28 (SD 0.21) and 2.09 (SD 0.36) hours, respectively) than after I/V (0.49 (SD 0.07) and 0.66 (SD 0.16) hours, respectively) administration (p<0.05). Bioavailability after I/M injection of alfaxalone was 94.7 (SD 19.8)%. The mean intervals to sternal and lateral recumbency were longer in the I/M (3.73 (SD 1.99) and 6.12 (SD 0.90) minutes, respectively) compared to I/V (0 minutes for all animals) treated cats (p<0.01). Sedation scores indicative of general anaesthesia (scores >15) were recorded from 5–15 minutes after I/V administration and deep sedation (scores 11–15) at 20 and 30 minutes. Deep sedation was observed from 10–45 minutes after I/M administration. One cat from each group showed hyperkinesia during recovery, and the remainder had an uneventful recovery.

CONCLUSIONS AND CLINICAL RELEVANCE: Alfaxalone administered I/V in cats provides rapid and smooth induction of anaesthesia. After I/M administration, a longer exposure to the drug and an extended half life were obtained compared to I/V administration. Therefore I/M administration of alfaxalone could be a reliable, suitable and easy route in cats, taking into account that alfaxalone has a slower onset of sedation than when given I/V and achieves deep sedation rather than general anaesthesia.  相似文献

The interaction of alkyl sulfonate ligands (AL) with bovine beta-lactoglobulin AB was measured using Trp fluorescence enhancement. One binding site per protein molecule was observed. The location of this site was related with the dimer formation and could be coincident with the fatty acids and SDS binding site. The apparent binding constants for AL were in the range of 10(-)(6) M, at pH 6.8. At pH 3.0 no binding was observed by this fluorescence method. The strength of the interaction was decreasing in the following way: AL16 > AL12 > AL14 > AL10. Other sites on the monomer were evidenced by the protective action of the AL toward the urea unfolding of the protein.  相似文献

Testing of new rootstocks for drought tolerance targets traditionally rain-fed districts where supplemental irrigation is more frequently needed due to the pressures of global warming. A seasonal evaluation of gas exchange and water-use efficiency (WUE) of cv. Sangiovese grafted to the new drought-tolerant genotype M4 in a dry-down trial against the commercial SO4 stock is reported. The experiment was conducted in 2014 on twelve 2-year-old, non-fruiting potted Sangiovese grapevines grafted on M4 and SO4 stocks and assigned to SO4-WW (well-watered), SO4-WS (water-stressed), M4-WW and M4-WS treatments. Progressive water deficit was imposed by reducing water supply to 70, 50 and 30 % of whole-canopy demand derived from concurrent measurements of transpiration in WW. Unlike SO4, M4 showed slower stress progression, as highlighted by pre-dawn leaf water potential, and retained higher whole-canopy net CO2 exchange rates (NCER) and transpiration rates per unit of leaf area at all replenishment levels as well as exhibiting higher canopy WUE at both 50 and 30 % WW. Although single-leaf assessment was in partial disagreement with data recorded on the whole-canopy basis, robust data were acquired to confirm that the M4 stock performs better than SO4 at any water replenishment level in terms of higher NCER/leaf area and/or canopy WUE. Findings feed expectations that grafting Sangiovese to the M4 rootstock should result in a problem-solving tool for rain-fed areas subject to temporary summer drought.  相似文献

Paspalum dilatatum Poir., is a perennial C4 grass widely distributed in the Argentinean Pampas. The response to water availability for materials developed with forage‐production purposes is unknown. We hypothesized that genetic differences between commercial varieties are reflected in their regrowth capacity under water stress. The effect of five levels of constant water supply on three plant varieties (two derived from apomictic materials: ‘Relincho’ and ‘Alonso’ and one from sexually‐derived material: ‘Primo’) were examined in the greenhouse. Leaf‐ and plant‐response traits were followed during 38 d after a single defoliation event. Seven response variables were measured: three of them were morphogenetic (leaf elongation rate, leaf appearance rate and leaf elongation duration) and four were structural (number of live leaves, lamina length, tiller biomass and tiller production). The sexual material showed higher values for growth variables than the apomictic varieties (leaf elongation rate, leaf length and tiller biomass) across the environmental range. Apomictic varieties showed a proportionally similar drought response to the sexual material for the seven variables. No intra‐specific trade‐off (statistical interaction) was found between growth under high water availability conditions and drought tolerance.  相似文献

This study examined how leaf and stem functional traits related to gas exchange and water balance scale with two potential proxies for tree hydraulic architecture: the leaf area:sapwood area ratio (A(L):A(S)) and wood density (rho(w)). We studied the upper crowns of individuals of 15 tropical forest tree species at two sites in Panama with contrasting moisture regimes and forest types. Transpiration and maximum photosynthetic electron transport rate (ETR(max)) per unit leaf area declined sharply with increasing A(L):A(S), as did the ratio of ETR(max) to leaf N content, an index of photosynthetic nitrogen-use efficiency. Midday leaf water potential, bulk leaf osmotic potential at zero turgor, branch xylem specific conductivity, leaf-specific conductivity and stem and leaf capacitance all declined with increasing rho(w). At the branch scale, A(L):A(S) and total leaf N content per unit sapwood area increased with rho(w), resulting in a 30% increase in ETR(max) per unit sapwood area with a doubling of rho(w). These compensatory adjustments in A(L):A(S), N allocation and potential photosynthetic capacity at the branch level were insufficient to completely offset the increased carbon costs of producing denser wood, and exacerbated the negative impact of increasing rho(w) on branch hydraulics and leaf water status. The suite of tree functional and architectural traits studied appeared to be constrained by the hydraulic and mechanical consequences of variation in rho(w).  相似文献

Fourteen calves were inoculated intranasally (i.n.) with the viral isolates as follows: 5 with 85/BH 16TV, 1 with 85/BH 17TV, 1 with 85/BH 18TV, 2 with 85/BH 231TN and 5 with 85/BH 232TN. Strain 85/BH 16TV was the only one which caused overt respiratory-like disease in all inoculated calves. Onset of the disease was observed after 7-8 days of incubation and was characterized by fever, depression, nasal discharge and coughing. Virus was isolated from the nasal swabbings of calves obtained from post-infection day (PID) 2-10. The other viral strains did not cause any sign of disease although virus was isolated regularly from the nasal swabbings of the inoculated calves. Virus was recovered from central nervous system tissues of calves that were infected with 85/BH 16TV or 85/BH 232TN strains and were killed on PID 4 or 8. Virus was also isolated from other tissues, such as lymph node, nasal mucosa (PID 8), or lung (PID 4). It was speculated that the nervous system could be one of the target areas of the virus of the naturally occurring infection by BHV-4. This might indicate a possible role of the nervous system (site of latency?) in the pathogenesis of BHV-4 as is the case in certain herpesviral infections of man and the lower animals.  相似文献

The culture supernatant of Escherichia coli, isolated from ostriches with diarrhea in Brazil, caused elongation in Vero cell, rounding in Chinese hamster ovary (CHO) cells and a cytoplasmic vacuolation in ostrich embryo fibroblasts (OEF), but it was not cytotoxic for chicken embryo fibroblasts (CEF). These effects were not neutralized by antiserum to cholera toxin. Polymerase chain reaction assays showed that the ostrich E. coli contained the gene encoding (eltII-A), but not those for type 1 heat-labile enterotoxin (eltA), heat-stable enterotoxins (estA, estB), verocytotoxins (stx-I, stx-II), or cytotoxic necrotizing factors (cnf 1, cnf 2). All isolates belonged to serotype O15:H8. The enteropathogenic relevance of LT-II in ostrich diarrhea remains undetermined.  相似文献

A serological survey of respiratory virus infections was carried out from 1998 to 2001 in Lecco province, Italy, as part of a health monitoring programme in a population of alpine chamois, many of which died of pneumonia in autumn and winter 2000 to 2001; 194 carcases of all age classes were found over a short period and in a small area. Eighteen of them, which were examined postmortem, consistently showed signs of severe fibrinous lobar pneumonia or catarrhal bronchopneumonia. Samples of serum from 145 chamois collected from hunted animals and carcases were tested by a virus neutralisation test against bovine respiratory syncytial virus (BRSV), bovine viral diarrhoea virus, bovine herpesvirus type 1 and parainfluenzavirus type 3. Positive results were detected only for BRSV. The area was divided into two subunits on the basis of the distribution of deaths; in the areas where fatalities were observed there was a significant increase of BRSV titres at the beginning of the outbreak. Furthermore, during the 2000 and 2001 hunting seasons antibody titres to BRSV were significantly higher in the areas where mortality occurred. The roe deer living in the same area were not affected by pneumonia and had a low prevalence of titres to BRSV which did not vary during the period of the study.  相似文献
