A series of N-substituted derivatives of aminomethylenebisphosphonic acid were evaluated as potential inhibitors of delta1-pyrroline-5-carboxylate reductase (EC 1.5.1.2), the enzyme that catalyzes the last step in proline biosynthesis, partially purified from Arabidopsis thaliana suspension cultured cells. At millimolar concentrations, three compounds out of 26 were found to interfere with the catalytic mechanism. One of them, namely, 3,5-dichloropyridyl-aminomethylenebisphosphonic acid, retained such inhibitory activity in the micromolar range. Kinetic analyses ruled out the possibility that the inhibition could simply rely upon the chelating properties of bisphosphonates and showed mechanisms of a noncompetitive type against NADH and an uncompetitive type against delta1-pyrroline-5-carboxylic acid, with KI values of 199 +/- 6 and 10.3 +/- 1.5 microM, respectively. A computer-aided docking analysis, performed on the basis of the crystal structure of the enzyme from Streptococcus pyogenes, suggested that this phosphonate may interact with amino acid residues near the binding site of delta1-pyrroline-5-carboxylic acid, thus blocking the substrate in a pocket and preventing its interaction with NADH. Because in higher plants the step catalyzed by delta1-pyrroline-5-carboxylate reductase is shared by all pathways leading to proline synthesis, such a compound may represent a lead structure to be exploited for the design of new substances endowed with herbicidal activity.  相似文献

A series of 12 3-(4-bromobenzyl)-5-(arylmethylene)-5 H-furan-2-one lactones, designed using the naturally occurring toxin nostoclides as a lead structure, were synthesized and screened as potential inhibitors of photosynthetic electron transport. The structures were confirmed by (1)H and (13)C NMR, MS, and IR analyses. Their biological activity was evaluated both in vitro, as the ability to interfere with light-driven reduction of ferricyanide by isolated spinach chloroplasts, and in vivo, as the capability to inhibite the oxygen production by intact Chlorella cells. Some of the compounds exhibited inhibitory properties in the micromolar range against basal and phosphorylating electron flow from water to K 3[Fe(CN) 6], with no effect on uncoupled electron flow. Thus, they seem to behave as energy-transfer inhibitors. Although poor solubility in water may limit their effectiveness, the active derivatives could present structures to be exploited for the design of new substances endowed with herbicidal activity.  相似文献

Purpose

The existence of black horizons (BHs) is often highlighted in European soils, and in the Po River plain of northern Italy. Nevertheless, BH chronological frameworks and genetic models are still debated. The present study investigated the genesis of BHs in the eastern Po Plain where they are buried at various depths.

Materials and methods

Soil sequences were investigated with a multidisciplinary approach integrating geomorphologic, stratigraphic, pedologic, geochemical, isotopic, palynological, and radiometric analyses.

Results and discussion

The formation of the studied BHs was scattered over time from the Last Glacial Maximum to at least the middle Holocene. The new data indicate that BHs developed when the landscape was dominated by coniferous forest during conditions that were totally different from the current pedoclimatic setting. The recurrent presence of black particles indicates that this vegetation cover was systematically affected by fire episodes that induced soil degradation and mineralization processes of the original organic compounds, thus contributing to darkening of the upper soil horizons.

Conclusions

BH formation clearly coincided with cold time lapses. Evidence for repeated fire events (natural or human-induced?) provides insights for the controversial debate on early anthropogenic impacts on the environment.

Some pyrazolo[3,4-d]pyrimidin-4(5H)-thione, pyrazolo[3,4-d][1,3]thiazin-4-one/thione, and pyrazolo[1,5-c][1,3,5]thiadiazine-4-one/thione derivatives were synthesized and screened for antifungal activity against the causal agent of rice blast disease, Magnaporthe grisea. In all cases a remarkable inhibition of fungal growth was found in the range from 10 to 200 microg x mL(-1). Several compounds were able to control mycelium growth at a rate of 10 microg x mL(-1), a concentration at which the reference compound tricyclazole was completely ineffective. At least in the case of the most active substance, at the same dose the growth of seedlings or cultured cells of rice was substantially unaffected. Results allowed definition of structural requirements either to maintain or to enhance mycotoxic activity.  相似文献

The [4 + 3] cycloaddition of the proper furans with the oxyallyl cation, generated in situ from 2,4-dibromopentan-3-one, produced a series of 8-oxabicyclo [3.2.1]oct-6-en-3-ones. Exposure of the oxabicycles to ozone afforded the corresponding 8,9,10,11-tetraoxatricyclo[5.2.1.1 (2,6)]undecan-4-ones in variable yields (7-100%). The phytotoxic properties of these ozonides (or 1,2,4-trioxolanes) and their oxabicycle precursors were evaluated as the ability to interfere with the growth of Sorghum bicolor and Cucumis sativus seedlings. Among oxabicycles, the highest inhibitory activity was shown by compounds possessing a alpha,beta-unsaturated carbonyl moiety. A differential sensitivity of the two crops was evident with ozonides. The most active compounds were also tested against the weed species Ipomoea grandifolia and Brachiaria decumbens. To the best of our knowledge, this is the first article describing ozonides as potential herbicides.  相似文献

The effects of chemical (acid-heating treatment) and enzymatic (microbial transglutaminase, TGase) modification (deamidation) of gluten proteins on their physicochemical and celiac disease-related properties were studied. Ammonia release, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and sample solubility analyses were employed to check the extent of gluten modification. Among different treatments achieved, the acid-heating treatment performed at 90 degrees C for 3 h induced gluten deamidation, paralleling an increase of gluten solubility without relevant proteolysis. Changes in the immunoreactivity of celiac IgA anti-gliadin antibodies (AGAs) to modified gluten proteins were detected by using a competitive indirect enzyme-linked immunosorbent assay method. Chemical deamidation by acid-heating treatment of gluten lowered IgA-AGA immunoreactivity. IgA-AGA immunoreactivity to gliadins was increased when they were submitted to TGase-catalyzed deamidation. The acid-heating treatment of gluten reduced its cytotoxic activity on human colon adenocarcinoma LoVo cell line. These results showed that chemical deamidation of gluten may be envisaged as a way to lower the potential risk for celiac people due to widespread use of gluten as a food additive.  相似文献

Yield losses from ozone pollution can be estimated by two methods: one involves the use of sensitive (S) and resistant (R) biotypes of white clover (Trifolium repens L., cv. Regal) exposed in ambient air, the other is based on the use of open-top chambers (OTC) supplied either with charcoal-filtered (CF) or non-filtered (NF) air. In southern Italy the two methods have been compared using the clover biotypes. The aim was (1) to compare the extent of ozone-induced yield reductions estimated by the two methods, (2) to evaluate the effect of the chamber enclosure on the growth of both biotypes, and (3) to compare plant water consumption in the different environments. On the average, the yield reduction was 23% when derived from the S/R yield ratio in ambient air, and 18% obtained by the CF/NF yield ratio of the S-type, without a significant difference between the two values. The slightly lower value for the OTC-based system may be due to the lower ozone levels in NF chambers due to losses in the ventilation system. Thus, both methods yielded equivalent yield reductions of about 20% due to ozone at this Mediterranean site. However, the higher air temperature inside OTCs influenced the plant growth, and this effect was stronger in the case of the R type. Therefore, R/S yield ratios in NF chambers differed from ambient air. Also, plant water consumption was higher in OTCs than in ambient air. The results suggest that the OTC-based method enables yield loss estimates at this Mediterranean site, in spite of chamber effects on plant growth and water use.  相似文献
