This study was conducted over a two year period in the north west of Madeira Island (Macaronesian Atlantic Islands, Portugal). It investigates the relationship between fruiting patterns in the indigenous forest and the use by the Madeira laurel pigeon of forest habitats and cultivated areas. Habitat use observations were carried out using line-transects in forest sites where the availability of fruit (the most important element of the diet) was assessed. The use of agricultural fields was measured indirectly by the assessment of the damage caused to crops. The findings provided strong evidence that bird movements and shifts in abundance are related to fruit usage and availability. The use of fruits is mostly opportunistic but when many species are available, selection was observed. From an ecological perspective, our findings indirectly confirm a high concordance between pigeon diet, studied by microhistological techniques, and habitat use. The use of crops also seems to be mostly opportunistic and is governed by the birds’ movements within the adjacent forest. It is hypothesised that fruit phenology will influence the use of agricultural areas only to the extent that it governs such movements; there is no strong evidence that crops are attacked only when the availability of natural foods is low. Our results suggest that the current management strategy for the forest is adequate for the conservation of this pigeon but that the conservation value of forest edges should be redefined. It is also proposed that crop damage may be reduced by identifying the proximate factors, which lead to the use of specific fields.  相似文献

We describe a new molecular class of genetic-pairing system that has a native DNA backbone but has all four base pairs replaced by new, larger pairs. The base pairs include size-expanded analogs of thymine and of adenine, both extended by the width of a benzene ring (2.4 A). The expanded-diameter double helices are more thermodynamically stable than the Watson-Crick helix, likely because of enhanced base stacking. Structural data confirm a right-handed, double-stranded, and base-paired helical form. Because of the larger base size, all the pairs of this helical system are fluorescent, which suggests practical applications in detection of natural DNA and RNA. Our findings establish that there is no apparent structural or thermodynamic prohibition against genetic systems having sizes different from the natural one.  相似文献

Summary

The dependence between flowering cycles and woody stem parts of a rose crop was studied, with respect to plant architecture and carbohydrate content. Two harvesting methods (“control” versus “flush”) and two plant types (“control” versus “one stemmed”) were compared. Crop growth and partitioning of dry matter were studied as influenced by crop management during 18 months of culture. For a full-productive year, flush harvesting generally promoted bud break as compared with continuous harvesting but at the same time, also due to lower light interception, blind-shoot formation was enhanced and the individual flower weight reduced. Numbers of basal shoots were hardly related to flower production over a full cropping year. Flower production was much more sensitive to the number and diameter of branches at the height of cutting the flowers. The treatments did not affect carbohydrate allocation in the plant. Total carbohydrate storage was much too low to argue a clear role for the possible use of movement of carbohydrate reserves towards new growth. Maximum starch concentration was found at the beginning of summer and gradually dropped to a minimum in December and then increased again to a spring maximum. A cold treatment did increase the total non-structural carbohydrate (TNC) concentration but no positive influence on new basal-shoot formation was observed. No starch gradient was found in basal stem parts.  相似文献

Combined oxygen (O) and nitrogen (N) stable isotope analyses are commonly used in the source determination of nitrate . The source and fate of are studied based on distinct O and N isotopic signatures (δ18O and δ15N) of various sources and isotopic effects during transformation processes, which differ between sources like fertilizer, atmospheric deposition, and microbial production (nitrification). Isotopic fractionation during production and consumption of further affects the δ18O and δ15N signal. Regarding the δ18O in particular, biochemical O exchange between O from and H2O is implicitly assumed not to affect the δ18O signature of . This study aimed to test this assumption in soil-based systems. In a short (24 h) incubation experiment, soils were treated with artificially 18O and 15N enriched . Production of from nitrification during the incubation would affect both the 18O and the 15N enrichment. Oxygen exchange could therefore be studied by examining the change in 18O relative to the 15N. In two out of the three soils, we found that the imposed 18O enrichment of the declined relatively more than the imposed enrichment. This implies that O exchange indeed affected the O isotopic signature of , which has important implications for source determination studies. We suggest that O exchange between and H2O should be taken into consideration when interpreting the O isotopic signature to study the origin and fate of in ecosystems.  相似文献

The Portuguese island of Selvagem Grande (Great Salvage) in Macaronesia is an important seabird breeding station in the eastern Atlantic. Significant populations of Cory's shearwater Calonectris diomedea (Scopoli, 1769), Bulwer's petrel Bulweria bulweria (Jardine & Selby, 1828) and little shearwater Puffinus assimilis baroli (Bonaparte, 1857) are present, and white-faced storm-petrel Pelagodroma marina (Latham, 1790) and Madeiran storm-petrel Oceanodroma castro (Harcourt, 1851) populations are of global significance. Selvagem Grande also provides diverse habitats for an extensive flora, including 11 endemic species. The 270-ha island was also inhabited by two alien invasive mammals: the European rabbit Oryctolagus cuniculus (Linnaeus, 1758) and the house mouse Mus musculus (Linnaeus, 1758). Both are known to have had adverse impacts on breeding seabirds and island vegetation. In 2002, the Natural Park of Madeira conducted a program using brodifacoum bait formulations aimed at rabbit and mouse eradication. Approximately 17 000 individual baiting points were established on a 12.5 × 12.5 m grid. Baits were also applied by hand “seeding” on steep slopes and cliffs where bait stations could not be placed. Rabbits were removed after a month. However, mice persisted for considerably longer and strategic bait applications against them continued for a further six months. Subsequent assessments by trapping, bait takes and systematic observation of signs over three years, has confirmed the removal of both alien invasive species. This paper presents information on these operations, on measures adopted to mitigate adverse impacts of the eradication program on important vertebrate non-target species, including Berthelot's pipit Anthus berthelotii Bolle, 1862 and a species of gecko Tarentola bischoffi Joger, 1984 and on the initial response of the island's ecosystem to the eradication of rabbits and mice.  相似文献

Drainage-weighing lysimeters allowed monitoring of water balance components of non-bearing olive (Olea europaea cv Barnea) trees over a 3-month period including short-term events of controlled but severe water stress. The objective of the study was to evaluate a variety of soil and plant-based water status and drought stress monitoring methods on the basis of tree-scale evapotranspiration (ET). As the trees entered into and recovered from water stress, meteorological data, actual ET (ETa), soil water content and changes in leaf turgor pressure were continuously monitored. Additionally, midday measurements of stem water potential, stomatal conductance, canopy temperature, and quantum yield of PSII photochemistry were conducted. Diurnal (dawn to dusk) measurements of all the above were made hourly on days of maximum stress. Shoot elongation rate was measured for periods of stress and recovery. Quantum yield of PSII photochemistry, stomatal conductance, and stem water potential all successfully indicated reductions in whole-tree water consumption beginning at moderate stress levels. These measured parameters fully recovered to the levels of non-stressed trees soon after water application was renewed. Shoot elongation was reduced 25-30% for the 10-day period during and following drought and recovered thereafter to levels of non-stressed trees. Whole-tree ETa was reduced by as much as 20% even following full recovery of the leaf level parameters, suggesting reduced canopy size and growth due to the stress period. Non-destructive, continuous (turgor pressure) and remotely sensed (canopy temperature) methods showed promising potential for monitoring effects of water stress, in spite of technological and data interpretation challenges requiring further attention.  相似文献

Background

Information on the genetic events leading to thyroid cancer in dogs is lacking.

Hypothesis/Objectives

Upregulation of the PI3K/Akt pathway has an important role in the tumorigenesis of thyroid carcinoma in dogs.

Animals

Fifty‐nine dogs with thyroid carcinoma and 10 healthy controls.

Methods

Quantitative RTPCR was performed for VEGFR‐1, VEGFR‐2, EGFR, PIK3CA, PIK3CB, PDPK1, PTEN, AKT1, AKT2, COX‐2, and CALCA. Mutation analysis was performed for known hotspots of RAS (N, K, H), PIK3CA, BRAF, RET, and for the entire coding region of PTEN.

Results

Forty‐three dogs (73%) had follicular cell thyroid carcinoma (FTC) and 16 dogs (27%) had medullary thyroid carcinoma (MTC). The relative mRNA expressions of VEGFR‐1 (P < .001), VEGFR‐2 (= .002), PDPK1 (< .001), AKT1 (= .009), and AKT2 (< .001) were increased in FTC, and those of EGFR (< .001), VEGFR‐1 (= .036), and PIK3CA (= .019) were increased in MTC when compared to normal thyroid glands. Mutation analysis of K‐RAS identified 2 activating missense mutations, which also have been described in thyroid cancer of humans. A G12R substitution was present in 1 FTC and an E63K substitution was present in 1 MTC. No functional mutations were found in the sequenced regions of H‐RAS, N‐RAS, PIK3CA, BRAF, RET, and PTEN.

Conclusions and Clinical Importance

The increased expression of several genes associated with PI3K/Akt signaling suggests the involvement of this pathway in the pathogenesis of thyroid carcinoma in dogs, warranting further research on pathway activation and gene amplification. The mutations most frequently associated with thyroid cancer in humans are rare in dogs.  相似文献

Summary Flower colour is the major contributor to the total ornamental value of a flower. The combination of biochemical knowledge and genetic engineering technology has resulted in the addition of a new colour to the existing colour range ofPetunia hybrida. This has been achieved by expression of the maize dihydroflavonol-4-reductase (DFR) gene in a suitable petunia acceptor which leads to the accumulation of pelargonidin-derived pigments in flowers. The resulting flower colour, however, was a pale brick-red, which is commercially unattractive in petunia. Our objective was to produce a product suitable for commercialisation by introducing the DFR gene into our breeding material via normal sexual recombination. Although the initial transformant exhibited many negative characteristics, first analyses indicated that it was feasible to obtain material for creating commercial hybrids. Experimental hybrids based on F4 lines were obtained with improved phenotypical expression of the orange flower colour in combination with a good general performance. In order to assess consumer-related characteristics, selected experimental hybrids were tested under field conditions. All transgenic plants had a normal appearance when compared to non-transgenic control plants. No linkage was observed between the transgenic trait and any negative characteristic. From these studies it can be concluded that through a combination of biochemistry, breeding and genetic engineering it is possible to generate unique flower colours in a cultivar with commercial potential.  相似文献
