Euplotin C is a sesquiterpene of marine origin endowed with significant anti-microbial and anti-tumor properties. Despite the promising functional profile, its progress as a novel drug candidate has failed so far, due to its scarce solubility and poor stability in aqueous media, such as biological fluids. Therefore, overcoming these limits is an intriguing challenge for the scientific community. In this work, we synthesized β-cyclodextrin-based nanosponges and investigated their use as colloidal carriers for stably complex euplotin C. Results obtained proved the ability of the carrier to include the natural compound, showing remarkable values of both loading efficiency and capacity. Moreover, it also allowed us to preserve the chemical structure of the loaded compound, which was recovered unaltered once extracted from the complex. Therefore, the use of β-cyclodextrin-based nanosponges represents a viable option to vehiculate euplotin C, thus opening up its possible use as pharmacologically active compound.  相似文献

As with many urban environments, a number of sources of airborne elemental mercury (Hg°) exist in New York City, yet little research has been conducted to examine the flux and sources of mercury in New York. In this study, we conducted ambient monitoring of Hg° at six locations in New York City. Airborne Hg° averaged 3.84±0.10 ng m-3 and 3.70±0.08 ng m-3 in the boroughs of Manhattan and Brooklyn respectively, yet only 2.69±0.03 ng m-3 in a more residential neighborhood in Queens. Both precipitation and ambient temperature were significantly correlated with ambient Hg° levels in New York City, suggesting that the surface emission of mercury from urban surfaces plays a role in urban Hg° concentrations. Local sources were also seen to contribute to urban Hg° levels by leading to `spikes' of Hg° in which elevated concentrations were recorded for short periods of time.  相似文献

Anchovy, Engraulis encrasicolus, forms the basis of Italian small pelagic fisheries in the Adriatic Sea. The strong dependence of this stock on environmental factors and the consequent high variability makes the dynamics of this species particularly complicated to model. Weekly geo‐referenced catch data of anchovy obtained by means of a Fishery Observing System (FOS) from 2005 to 2011 were referred to a 0.2 × 0.2 degree grid (about 20 km2) and associated with the environmental parameters calculated by a Regional Ocean Modelling System, AdriaROMS. Generalized Additive Mixed Models (GAMM) with and without random effects were used to identify a relationship between abundance in the catch and oceanographic conditions. The outcomes of models with no random effects, with random vessel effects and with the random vessel and random week‐of‐the‐year effects were examined. The GAMM incorporating a random vessel and week‐of‐the‐year effect were selected as the best model on the basis of the Akaike information criteria (AIC). This model indicated that catches (abundance) of anchovy in the Adriatic Sea correlate well with low temperatures, salinity fronts and sea surface height, and allowed the identification of areas where high concentrations of this species are most likely to occur. The results of this study demonstrate that GAMM are a useful tool to combine geo‐referenced catch data with oceanographic variables and that the use of a mixed‐model approach with spatial and temporal random effects is an effective way to depict the dynamics of marine species.  相似文献

Different species of mercury have different physical/chemical properties and thus behave quite differently in air pollution control equipment and in the atmosphere. In general, emissions of mercury from coal combustion sources are approximately 20–50% elemental mercury (Hg°) and 50–80% divalent mercury (Hg(II)), which may be predominantly HgCl2. Emissions of mercury from waste incinerators are approximately 10–20% Hg° and 75–85% Hg(II). The partitioning of mercury in flue gas between the elemental and divalent forms may be dependent on the concentration of particulate carbon, HCl and other pollutants in the stack emissions. The emission of mercury from combustion facilities depends on the species in the exhaust stream and the type of air pollution control equipment used at the source. Air pollution control equipment for mercury removal at combustion facilities includes activated carbon injection, sodium sulfide infection and wet lime/limestone flue gas desulfurization. While Hg(II) is water-soluble and may be removed from the atmosphere by wet and dry deposition close to combustion sources, the combination of a high vapor pressure and low water-solubility facilitate the long-range transport of Hg° in the atmosphere. Background mercury in the atmosphere is predominantly Hg°. Elemental mercury is eventually removed from the atmosphere by dry deposition onto surfaces and by wet deposition after oxidation to water-soluble, divalent mercury.  相似文献

The natural environment represents an important source of drugs that originates from the terrestrial and, in minority, marine organisms. Indeed, the marine environment represents a largely untapped source in the process of drug discovery. Among all marine organisms, sponges with algae represent the richest source of compounds showing anticancer activity. In this study, the two secondary metabolites pelorol (PEL) and 5-epi-ilimaquinone (EPI), purified from Dactylospongia elegans were investigated for their anti-melanoma activity. PEL and EPI induced cell growth repression of 501Mel melanoma cells in a concentration- and time-dependent manner. A cell cycle block in the G1 phase by PEL and EPI was also observed. Furthermore, PEL and EPI induced significant accumulation of DNA histone fragments in the cytoplasmic fraction, indicating a pro-apoptotic effect of both compounds. At the molecular level, PEL and EPI induced apoptosis through the increase in pro-apoptotic BAX expression, confirmed by the decrease in its silencing miR-214-3p and the decrease in the anti-apoptotic BCL-2, MCL1, and BIRC-5 mRNA expression, attested by the increase in their silencing miRNAs, i.e., miR-193a-3p and miR-16-5p. In conclusion, our data indicate that PEL and EPI exert cytotoxicity activity against 501Mel melanoma cells promoting apoptotic signaling and inducing changes in miRNA expression and their downstream effectors. For these reasons could represent promising lead compounds in the anti-melanoma drug research.  相似文献
