The use of phytotherapy to treat human diseases has its roots in pre-historical times. Despite the modern advances achieved in the field of synthetic chemistry, the most efficient drugs available have their genesis directly or indirectly related with the vegetal kingdom. Indigenous communities have long used plant extracts to treat illnesses. Many of these extracts have shown effective action, with new bioactive compounds being extracted and screened every year. These extracts have also proven to be good sources of therapeutic agents to the treatment of Leishmaniasis. This work highlights some of these agents, while trying to emphasize the importance of plants as a source of new and powerful drugs against this widespread disease.  相似文献

Oceans are a rich source of structurally unique bioactive compounds from the perspective of potential therapeutic agents. Marine peptides are a particularly interesting group of secondary metabolites because of their chemistry and wide range of biological activities. Among them, cyclic peptides exhibit a broad spectrum of antimicrobial activities, including against bacteria, protozoa, fungi, and viruses. Moreover, there are several examples of marine cyclic peptides revealing interesting antimicrobial activities against numerous drug-resistant bacteria and fungi, making these compounds a very promising resource in the search for novel antimicrobial agents to revert multidrug-resistance. This review summarizes 174 marine cyclic peptides with antibacterial, antifungal, antiparasitic, or antiviral properties. These natural products were categorized according to their sources—sponges, mollusks, crustaceans, crabs, marine bacteria, and fungi—and chemical structure—cyclic peptides and depsipeptides. The antimicrobial activities, including against drug-resistant microorganisms, unusual structural characteristics, and hits more advanced in (pre)clinical studies, are highlighted. Nocathiacins I–III (91–93), unnarmicins A (114) and C (115), sclerotides A (160) and B (161), and plitidepsin (174) can be highlighted considering not only their high antimicrobial potency in vitro, but also for their promising in vivo results. Marine cyclic peptides are also interesting models for molecular modifications and/or total synthesis to obtain more potent compounds, with improved properties and in higher quantity. Solid-phase Fmoc- and Boc-protection chemistry is the major synthetic strategy to obtain marine cyclic peptides with antimicrobial properties, and key examples are presented guiding microbiologist and medicinal chemists to the discovery of new antimicrobial drug candidates from marine sources.  相似文献

The aim of this study was to identify differentially expressed genes (DEG) in the Longissimus thoracis muscle of Nelore cattle related to fatty acid (FA) profile through RNA sequencing and principal component analysis (PCA). Two groups of 10 animals each were selected containing PC1 and PC2 extreme DEG values (HIGH × LOW) for each FA group. The intramuscular fat (IMF) was compared between cluster groups by ANOVA, and only the sum of monounsaturated FA (MUFA) and ω3 showed significant differences (p < .05). Interestingly, the highest percentage (95%) of phenotypic variation explained by the sum of the first two PC was observed for ω3, which also displayed the lowest number of DEG (n = 1). The lowest percentage (59%) was observed for MUFA, which also revealed the largest number of DEG (n = 66). Since only MUFA and ω3 exhibited significant differences between cluster groups, we can conclude that the differences observed for the remaining groups are not due to the percentage of IMF. Several genes that have been previously associated with meat quality and FA traits were identified as DEG in this study. The functional analysis revealed one KEGG pathway and eight GO terms as significant (p < .05), in which we highlighted the purine metabolism, glycolytic process, adenosine triphosphate binding and bone development. These results strongly contribute to the knowledge of the biological mechanisms involved in meat FA profile of Nelore cattle.  相似文献

In this study, sex and age influenced the hematological profiles of Jundiá (Silver Catfish) Rhamdia quelen. Females showed lower levels of hemoglobin, young fish increased lymphocyte counts, and older fish increased hematocrit values. These results indicate that, depending on age and sex, the fish have disparate hematological profiles. For this reason, it is important to consider the sex and age of an R. quelen when examining the impact of environmental and management factors on this species in terms of their hematological profiles.

The incidence of canine rabies has been widely reported in Brazil, and new rabies virus (RV) variants, genetically similar to canine RV, have recently been isolated from foxes. In order to derive the epidemiological characteristics of Brazilian Carnivora RV, Brazilian RVs isolated from dogs, cats, and foxes were genetically analyzed. Brazilian Carnivora RV isolates were divided into 2 main lineages. The predominant lineage was found in dogs and cats, which included the Argentinean and Bolivian Carnivora RV isolates, and was extensively distributed throughout Brazil and surrounding countries. The other lineage consisted of three sublineages containing Brazilian dog and fox RV isolates, with the dog sublineages located on an internal branch of 2 fox sublineages, suggesting that RV transmission events might have occurred between foxes and dogs in the past. These results suggest that contact between dogs and wildlife has the potential to generate new rabies variants and that it is important to control RV infection cycles in both dogs and wildlife to prevent spread of rabies infection.  相似文献

Evolution of flooding tolerance in weedy rice has occurred in several rice-growing regions, but the genes related to this process and the environmental effects are unknown. The objective of this study was to analyse the expression of genes related to flooding tolerance in response to temperature and flooding during the initial establishment of weedy rice. The experiments were carried out with rice cultivars IRGA 417 and Nipponbare, which are sensitive to flooding, and weedy rice ITJ03 and AV04 genotypes that have high and intermediate tolerance to flooding, respectively. The expression of genes related to reserve mobilisation, anaerobic respiration, escape and quiescence strategies was analysed at periods up to 24 days after sowing. The flooding tolerance of weedy rice genotype ITJ03 was associated with the expression of RAmy3D and OsTPP7 , which are involved in the mobilisation of carbohydrate reserves, ADH1 and ADH2, which participate in anaerobic respiration, and SNRKL1 that triggers rapid elongation of the coleoptile and emergence. Although the genes PDC1, SUS3 and SUB1 are important for flooding tolerance in cultivated rice, their expression was not directly related to flooding tolerance in weedy rice. A temperature of 20°C reduced levels of expression of the RAmy3D, ADH2 and SNRKL1 genes and low temperature had a negative effect on the establishment of weedy rice. Breeding of rice genotypes with tolerance of low temperatures and anaerobic conditions may be a viable strategy to improve the control of weedy rice in paddy fields.  相似文献

The use of saline waters is an option for the irrigation of salt tolerant ornamentals as competition for high quality water increases. However, despite the importance of ornamental shrubs in Mediterranean areas, salt tolerance of such species has received little attention. The aims of our investigation were to quantify the growth response and any injury symptom of 12 widely cultivated ornamental shrubs to irrigation with saline water and to investigate any possible relation with the concentration of Na+ and Cl in the plants. Species were irrigated with different salinities (10, 40, and 70 mM NaCl) for a 120-day period. At the end of salt treatment, plants were sampled and dry biomass recorded; the relative growth rate (RGR) was also calculated. Root and leaf samples from each species were used to evaluate Na+, K+ and Cl concentrations. Growth rates were significantly reduced in Cotoneaster lacteus, Grevillea juniperina and Pyracantha ‘Harlequin’, which also showed the highest percentage of necrotic leaves. The increasing external NaCl lead to an increase of Na+ and Cl in roots and leaves of the different species, although less Na+ was accumulated than Cl: growth reduction well correlated with the concentration of Cl and/or Na+ in the leaves. The most sensitive species (i.e. C. lacteus, G. juniperina and Pyracantha ‘Harlequin’) had high concentrations of Na+ and/or Cl in their leaves and also showed a decrease in their leaf K+/Na+ ratios. Even though other species (i.e. Bougainvillea glabra, Ceanothus thyrsiflorus, Leptospermum scoparium, Leucophyllum frutescens and Ruttya fruticosa) demonstrated a high ion concentration in their leaves, they could be considered relatively salt tolerant as there was little growth reduction and few symptoms of injury in the leaves. In some other cases (i.e. Cestrum fasciculatum, Escallonia rubra and Viburnum lucidum) the observed tolerance was related to higher ion concentration in the roots compared to the leaves, probably indicative of a limited transport to the shoots. Only in Eugenia myrtifolia was the absence of symptoms associated with a limited Na+ and Cl uptake from the rhizosphere.  相似文献

The West-African euryhaline tilapia, Sarotherodon melanotheron heudelotii shift from visually feeding on zooplankton when juveniles to mostly filter feeding on phytoplankton when adults. When reared using an appropriate ration in intensive aquaculture systems, S. m. heudelotii also consume algal-based detritus, and contribute to sediment mineralization, clean up their environment, and ultimately stimulate and sustain algal growth. We analysed such practical advantages for phytoplankton-based recirculating systems, using S. m. heudelotii and Chlorella sp. as biological material originating from the prototype of such a system operated in Senegal. We performed a 24-h factorial design experiment in 36 tubs, cross-classifying three levels of S. m. heudelotii (fishless control, unfed fish, and fed fish) with four levels of Chlorella initial density.Chlorella overall mean density increased significantly from fishless, to unfed fish, and fed fish treatments, and with Chlorella initial density. S. m. heudelotii did not alter nitrogen nor phosphorus concentrations, only affected by algal initial densities. Most ammonia excreted by fish was probably uptaken by Chlorella. Bacteria-mediated diurnal nitrification was possibly an alternative ammonium loss mechanism at highest oxygen concentrations. Algae were not limited by nitrogen or phosphorus but most likely by low dissolved organic carbon availability. Chlorella differential responses with fed vs. unfed Sarotherodon suggest that CO2 supplied by heterotrophic S. m. heudelotii respiration played a key role. Observed Chlorella growth rates were similar to the highest rates obtained in algal mass cultures, enriched with CO2, nitrate and phosphate, under artificial lighting.Our results suggest the existence of a Sarotherodon-Chlorella mutualism in our systems, where S. m. heudelotii provide CO2, the major limiting factor of Chlorella growth, whereas Chlorella oxygenate and detoxify the water media from ammonia, promoting S. m. heudelotii production. This mutualism could be used to optimize photosynthetic suspended-growth aquaculture systems, particularly in the Tropics where light is abundant and temperature is continuously high.  相似文献
