In a previous experiment it has been found that maximum uptake rate (Imax), Michaelis constant (Km), and minimum nutrient concentration (Cmin) as plant physiological characteristics may be important for phosphorus (P) uptake in peat-substrate. Thus, variation of P uptake parameters was evaluated with a series of depletion studies for poinsettia (Euphorbia pulcherrima) and marigold (Tagetes patula) under fluctuating climatic conditions and different developmental stages.  相似文献

The progressive morphohistopathologic changes, distribution pattern of lesions and ultrastructural characteristics in Eimeria arloingi infection were investigated in experimentally infected kids. The 18 newborn animals allocated to 3 equal groups. Two of groups, A, B were inoculated with a single dose of 1 × 103 and1 × 105 sporulated oocysts of E. arloingi, respectively. At 7, 14, 21, 28, 35, and 42 days postinoculation (DPI), 1 kid from each group was necropsied for pathologic and ultrastructural studies. Progressive lesions were present at 21, 28, 35 and 42 DPI in the jejunum, ileum, cecum with fewer in the duodenum and proximal colon. The oocysts shedding begin between 16 to 18 DPI. Grossly, minimal changes were observed at 21 DPI as few whitish plaques or nodules and advanced lesions at 42 DPI as pseudoadenomatous pattern in the mucosa and a cerebriform pattern on the serosal surface of jejunum and ileum. Early histopathologic lesions due to schizogony phase were including presence of intracytoplasmic developmental stages of the parasite such as trophozoites, immature to mature schizonts and mild infiltration of inflammatory cells. In late lesions due to various stages of gametogony, the histological pattern was mainly remarkable hyperplasia of the villi and crypts epithelial cells, eventually developed into papillary projections of reactive epithelium. The mesenteric lymph nodes showed a few numbers of large schizonts in the cortical lacteals. This study showed E. arloingi as a highly pathogenic species for kids, the incubation period was 16–18 days and the main target organ was jejunum with characteristic morphohistopathologic lesions.  相似文献

Streptococci are frequently isolated from bovine mastitis in dairy cows with only limited information available on the antimicrobial susceptibility of these organisms. A total of 42 Streptococci isolated from 148 milk samples of normal, sub acute and acute bovine mastitis cases. Overall, 35% of the strains tested were Streptococcus dysgalactiae, Streptococcus agalactiae 26%, Streptococcus uberis 18 and 4% were Enterococcus sp. Differences between the number of isolations in acute and sub acute groups were statistically significant, (p<0.5). The antimicrobial susceptibility for these organisms was determined for the following antimicrobial agents: cephalexine, penicillin, clindamycin, cloxaciline, gentamicin, streptomycin, amoxicillin, tetracycline, kanamycin, oxytetracycline, ampicillin, chloramphenicol and erythromycin. S. agalactiae, S. dysgalactiae, S. uberis and Enterococci demonstrated high level of resistance against streptomycin, penicillin and cloxaciline. Low level of sensitivity to other tested antimicrobials was demonstrated.  相似文献

The diagnostic characteristics of immunohistochemistry (IHC) and polymerase chain reaction (PCR) methods were studied in the tissues of broiler chicken embryos experimentally infected by Neospora caninum. An infection with N. caninum NC-1 isolate was conducted in 70 broiler chicken embryonated eggs randomly divided into seven equal groups. After 8 days of incubation, six groups were inoculated with 10, 10(2), 10(3), 10(4), 10(5), and 10(6) doses of tachyzoites/embryonated egg. The 7th group was considered as control. The mortality rate and pathological changes of the dead embryos and hatched chickens up to 60 days old were noticed. Consecutive sections to those used for histopathological examination including the liver, heart, brain, and chorioalantoic (CA) membrane were subjected to IHC. The intensity and distribution of the immunostaining was graded as highly to mildly positive. For PCR procedure, DNA was extracted from 50mg of the tissues and primer pair Np21/Np6 was used for amplification of the Nc-5 gene. The results of the immunosignaling ranged from variable degrees of mild to moderate staining as dark-brown to brown and coarsely to finely granular, mostly within the cytoplasm of infected cells such as the endothelial cells of blood vessels. The parasite aggregation was more predominant in the heart than other tissues. Immunoreactivity for N. caninum antigen was multifocally moderate positive in the heart, liver and CA of the 10(3) dose, and also heart, liver, brain and CA of the 10(4) dose. IHC showed mildly positive in the liver and heart of the chicken embryos infected with 10 and 10(2) tachyzoites, as well. The results of the PCR confirmed the existence of the parasite in all of the examined tissues from the 10(3) and 10(4) doses. In conclusion, the results indicate a good agreement between IHC and PCR in diagnosis of neospora antigen in the infected tissues.  相似文献

Banana is an important crop in the Kagera region of Tanzania. Banana xanthomonas wilt (BXW) was first reported in Kagera in 2006, and is now an important limiting factor in banana production, because all cultivars are susceptible and infected plants can fail to produce fruit. BXW is caused by Xanthomonas campestris pv. musacearum (Xcm), which is spread by farm tools, infected planting materials, and pollinating insects. Practices that address Xcm dissemination, such as mat removal, debudding and tool sterilization, have not prevented the spread of BXW in the region. Disease surveys were conducted in Kagera from 2007 to 2011 to assess BXW presence, monitor its intensity and evaluate its socioeconomic impacts. Spatiotemporal clusters of BXW were analysed with Arc GIS and sas . The relationship between BXW clusters and environmental variables was examined using bivariate correlations in spss ; two modelling approaches, MaxEnt (maximum entropy) and logistic regression, were used to predict the potential distribution of BXW in Tanzania. Disease progress over time was best described with the Gompertz model. Significant clustering of BXW was observed in all years and hotspots were located in the Muleba, Karagwe, Misenyi and Bukoba rural districts. These findings suggest that BXW spreads rapidly over short distances. BXW clusters were positively correlated with rainfall and negatively with temperature and altitude. According to MaxEnt, precipitation was the main factor associated with BXW development. MaxEnt and logistic regression predicted a wide potential distribution of BXW in Tanzania because the climate in all banana‐growing regions is conducive for its establishment.  相似文献

Physicochemical traits of peat-based pot substrates prepared from mixing of 20 or 40?vol.% of some different mineral fractions (sub. 1 to 7, subjected to decreasing amount of clay content of 66.1, 61.2, 49.7, 38.5, 26.0, 23.7 and 19.4%, respectively) with black peat were evaluated and then phosphorus uptake of poinsettia (Euphorbia pulcherima) simulated mechanistically as a case study pot plant. For preparation of substrate number 8, pure peat was mixed with 40?vol.% of the same mineral component as substrate number 6 (with 23.7% clay). Result revealed that adding different mineral fractions up to 40?vol.% to pure peat, water-holding capacity and total pore volume were reduced only by 11 and 15?vol.%, respectively. It was found that keeping the moisture content of peat-based substrates with 20?vol.% of mineral soils around 60%–70%, the optimum ventilation and enough free space of about 15%–25% will be provided around the root system. In addition, simulated P uptake did not change significantly with addition of 20?vol.% of different mineral components in peat-based substrates. As a result, P uptake mainly was dependent on the amount of plant available P level than different mineral fractions.  相似文献

Phytophthora root and stem rot of soybean is a destructive disease of soybean in Iran. Races 1 and 3 of pathogen have already been reported from two major growing regions of the crop, Lorestan and Golestan provinces. In a survey during 2004-2005, 142 isolates of P. sojae were recovered from infected plants and naturally infested soil samples using selective media and soybean leaf baiting technique. The majority of tested isolates (110 isolates) belonged to race one of P. sojae and 32 isolates belonged to race 3. ITS region of 23 isolates were amplified with specific primers Ps1 and Ps2. Sequences of this regions were similar to other gene banks sequences except two isolates from China. This survey showed low diversity in Iranian population of P. sojae.  相似文献

It is important to develop integrated fertilization strategies for various crops that enhance the competitive ability of the crop, maximize crop production and reduce the risk of nonpoint source pollution from fertilizers. In order to study the effects of mineral nitrogen fertilization and biofertilizer inoculation on yield and some physiological traits of rapeseed (Brassica napus L.) under different levels of sulfur fertilizer, field experiments in factorial scheme based on randomized complete block design were conducted with three replications in 2012 and 2013. Experimental factors were: (1) four levels of chemical nitrogen fertilizer (0, 100, 150 and 200 kg N ha?1), (2) two levels of biofertilizer (with and without inoculation) consisting Azotobacter sp. and Azospirillum sp. and (3) two levels of sulfur application (0 and 50 kg S ha?1). Rapeseed yield, oil content of grains and studied physiological traits had a strong association with the N fertilization, biofertilizer inoculation and sulfur (S) application. Higher rates of N fertilization, biofertilizer inoculation and S application increased the grain yield of rapeseed. In the case of physiological traits, the highest value of relative water content (RWC) was recorded in 100 kg N ha?1 that was statistically in par with 150 kg N ha?1 application, while usage of 150 kg N ha?1 showed the maximum cell membrane stability (CMS). Inoculation with biofertilizer and S fertilization resulted in higher RWC and CMS in rapeseed plants. The chlorophyll content showed its maximum values in the highest level of N fertilization, biofertilizer inoculation and S application. The usage of 200 kg N ha?1 significantly decreased the oil content of rapeseed grains, but the highest grain oil content was obtained from the application of 150 kg N ha?1, Azotobacter sp. and Azospirillum sp. inoculation and S fertilization. It seems that moderate N rate (about 150 kg N ha?1) and S application (about 50 kg S ha?1) can prove to be beneficial in improving growth, development and total yield of inoculated rapeseed plants.  相似文献

A pot experiment was conducted to investigate factors contributing to phosphorous (P) efficiency of ornamental plants. Marigold (Tagetes patula) and poinsettia (Euphorbia pulcherima) were cultivated in a peat substrate (black peat 80% + mineral component 20% on a volume basis), treated with P rates of 0, 10, 35, 100, and 170 mg (L substrate)–1. During the cultivation period, plants were fertigated with a complete nutrient solution (including 18 mg P L–1) every 2 d. Both poinsettia and marigold attained their optimum yield at the rate of 35 mg P (L substrate)–1 and the critical level of P in shoot dry matter of both crops was 5–6 mg g–1. After planting, plant‐available P increased at lower P rates to a higher level for poinsettia than for marigold, but no significant change was observed at higher P rates. Balance sheet calculations indicated that at lower P rates more P was fertigated than was taken up by the plants. Root‐length density, root‐to‐shoot ratio, and root‐hair length of marigold were doubled compared to that of poinsettia. Root‐length density increased with crop growth, and 10 d after planting the mean half distance between roots exceeded the P‐depletion zone around roots by a factor of 3 and 1.5 for poinsettia and marigold, respectively. Thus, at this early stage poinsettia exploited only 10% of the substrate volume whereas marigold utilized 43%. Later in the cultivation period, the depletion zones around roots overlapped for both crops. Taking into account P uptake via root hairs, the simulation revealed that this was more important for marigold compared to poinsettia especially at low P‐supply levels. However, increase of P uptake due to root hairs was only 10%–20% at optimum P supply. For the two lower P levels, the P‐depletion profile around roots calculated for 10 d after planting showed that after 2 d of depletion the concentration at the root surface was below the assumed Km value (5 μM) and the concentration gradient was insufficient to fit the demand. A higher content of plant‐available P in the substrate was observed for poinsettia compared to marigold in the treatment with P application adequate for optimum growth, because more fertigated P was accumulated during early stages of cultivation due to lower root‐length density of poinsettia. The observed difference of root morphological parameters did not contribute significantly to P‐uptake efficiency, since P mobility in the peat substrate was high.  相似文献

Gross and histopathologic lesions of the integumentary system were studied in 103 camels (Camelus dromedarius) slaughtered in Yazd Province of Iran. Dermatophilosis was diagnosed in 14 (13.6%) of the studied camels. Grossly, patchy thickening of the skin with dark brown scabs or crusts were observed on the rump, flanks, abdomen, shoulders, and neck. A heavy infestation with brown hard ticks was present on the lesions in almost all of the affected camels. On parasitology examination, all the ticks were identified as Hyalomma spp. Histopathologic examination of the affected skins revealed prominent superficial thickening of the epidermis as orthokeratotic and parakeratotic hyperkeratosis, degenerating neutrophils, serous fluid, and bacterial filaments. A lot of keratinaceous debris associated with an exudate rich in neutrophils were seen as remarkable features of the lesions on the superficial epidermis. Dermal inflammation was mild with infiltration of mononuclear inflammatory cells, particularly lymphocytes, around superficial vessels. A Gram-positive filamentous organism which was divided into parallel rows of cocci was observed in the crusts by Brown and Brenn Gram stain. This is the first report of the occurrence of dermatophilosis in the camels of Iran. With regard to infestation of the lesions by Hyalomma spp., it seems that this tick may have an important role in the pathogenesis of dermatophilosis in the camels of this region.  相似文献
