Leptin may play a role in the endocrine-metabolic processes that guarantee the physiological course of lactation in dairy cattle. This study was aimed at determining the changes in plasma concentrations of leptin and some of the main hormones and metabolites involved in the lactogenetic process in high-yielding dairy cows throughout lactation; we also wanted to assess whether leptin secretion is subjected to seasonal influences. Blood samples were collected from 23 Italian Friesian dairy cows from the end of a lactation to the ninth month of the subsequent one; in addition, blood was sampled from 47 dairy cows in different phases of lactation during February and July. Plasma concentrations of leptin, growth hormone (GH), insulin, prolactin (PRL), glucose, non-esterified fatty acids (NEFA) and urea were quantified by either validated radioimmunoassay (RIA) or enzymatic colorimetric methods. At the beginning of lactation, GH concentrations significantly increased, while a significant reduction occurred in leptin and insulin. This endocrine condition, such as the significant increase in NEFA plasma concentrations, is indicative of a marked lipid mobilization. In the more advanced stages of lactation, when both energy and protein balances become positive, leptin plasma concentrations increased, whereas GH and NEFA concentrations declined. During the summer months, a significant increase in leptin plasma concentrations, irrespective of the phase of lactation, was observed. Collectively, our findings suggest that, in dairy cows, leptin may represent a 'metabolic signal' of animal's status of fattening and nutritional level; in addition, leptin seems to be influenced by photoperiod and environmental temperature.  相似文献

Purpose

The existence of black horizons (BHs) is often highlighted in European soils, and in the Po River plain of northern Italy. Nevertheless, BH chronological frameworks and genetic models are still debated. The present study investigated the genesis of BHs in the eastern Po Plain where they are buried at various depths.

Materials and methods

Soil sequences were investigated with a multidisciplinary approach integrating geomorphologic, stratigraphic, pedologic, geochemical, isotopic, palynological, and radiometric analyses.

Results and discussion

The formation of the studied BHs was scattered over time from the Last Glacial Maximum to at least the middle Holocene. The new data indicate that BHs developed when the landscape was dominated by coniferous forest during conditions that were totally different from the current pedoclimatic setting. The recurrent presence of black particles indicates that this vegetation cover was systematically affected by fire episodes that induced soil degradation and mineralization processes of the original organic compounds, thus contributing to darkening of the upper soil horizons.

Conclusions

BH formation clearly coincided with cold time lapses. Evidence for repeated fire events (natural or human-induced?) provides insights for the controversial debate on early anthropogenic impacts on the environment.

In this study we investigated some biological properties of flavonoids recovered in the aqueous (AqE) and ether (EtE) extracts from four Italian multifloral honeys. In particular, a cell-free assay was employed to detect direct reduction of ferricyanide, whereas an assay using intact human erythrocytes was used to measure the ability to donate electrons to a trans-plasma membrane oxidoreductase. It was found that the AqE displays greater "in vitro" ferricyanide-reducing activity than the EtE but, unlike the latter, is virtually ineffective in the cell-based assay. Uptake studies employing high-performance liquid chromatography/mass spectrometry (HPLC/MS) showed that the different results were explained by the inability of AqE components to cross the erythrocyte plasma membrane and by the excellent uptake of EtE flavonoids, which, once within the cell, donate electrons to the membrane oxidoreductase to efficiently reduce extracellular oxidants. The latter property appears to depend on the content of ether-soluble flavonoids in the starting honeys.  相似文献

We have demonstrated that insulin-like growth factor binding protein-5 (IGFBP-5) production by mammary epithelial cells increases dramatically during forced involution of the mammary gland in rats, mice and pigs. We proposed that growth hormone (GH) increases the survival factor IGF-I, whilst prolactin (PRL) enhances the effects of GH by decreasing the concentration of IGFBP-5, which would otherwise inhibit the actions of IGFs. To demonstrate a causal relationship between IGFBP-5 and cell death, we created transgenic mice expressing IGFBP-5, specifically, in the mammary gland. DNA content in the mammary glands of transgenic mice was decreased as early as day 10 of pregnancy. Mammary cell number and milk synthesis were both decreased by approximately 50% during the first 10 days of lactation. The concentrations of the pro-apoptotic molecule caspase-3 was increased in transgenic animals whilst the concentrations of two pro-survival molecules Bcl-2 and Bcl-x were both decreased. In order to examine whether IGFBP-5 acts by inhibiting the survival effect of IGF-I, we examined IGF receptor- and Akt-phoshorylation and showed that both were inhibited. These studies also indicated that the effects of IGFBP-5 could be mediated in part by IGF-independent effects involving potential interactions with components of the extracellular matrix involved in tissue remodeling, such as components of the plasminogen system, and the matrix metallo-proteinases (MMPs). Mammary development was normalised in transgenic mice by R3-IGF-I, an analogue of IGF-I which binds weakly to IGFBPs, although milk production was only partially restored. In contrast, treatment with prolactin was able to inhibit early involutionary processes in normal mice but was unable to prevent this in mice over-expressing IGFBP-5, although it was able to inhibit activation of MMPs. Thus, IGFBP-5 can simultaneously inhibit IGF action and activate the plasminogen system thereby coordinating cell death and tissue remodeling processes. The ability to separate these properties, using mutant IGFBPs, is currently under investigation.  相似文献

Soil is one of the most important factors for agricultural production. In tropical regions, soil variability is considerable, with the most diverse combinations of physical and chemical characteristics, an influence factor in crop growth and productivity. In this research, the main objective was to identify how soil characteristics and parent material can influence sugarcane development over time using remote sensing. An area located in Sao Paulo, Brazil, of 182 ha (one point per ha with soil analysis), with high variability in the parent material and soil types, was selected. Images from the Sentinel2-MSI satellite were used to describe the spectral behavior of sugarcane over a period of one year. The NDRE (normalized difference red-edge index) was calculated for each image and then the leaf area index (LAI) was obtained from it. Maps of soil classes, soil properties at two depths (0–0.20 and 0.80–1.0 m), and parent material classes were related to sugarcane LAI variability over time. Production environment zones, which is a classification based on soil characteristics to support sugarcane development, were also obtained and related to LAI variability. Spectral signatures of the crop presented different behaviors through the season, soil types and soil attributes provided useful responses for this variability. At the beginning of the season, the surface and subsurface soil properties (texture and fertility) impacted differently on crop development. On the other hand, soil classes and parent material influenced LAI in all production environments studied. The results indicated that the soil types and their properties at different depths have a significant impact on sugarcane development. Furthermore, RS was able to monitor the plant evolution and be related to soil types which may assist in plant management. The results can bring light on how better sugarcane management can be conducted using remote sensing data and soils variability.
