The confirmatory diagnosis of Mycobacterium bovis (M. bovis) in animal samples is carried out by culture in Stonebrink media. However, culture is very slow because of the extremely long duplication time of the bacillus and difficult because of the scarcity of bacilli in diagnostic samples. This study describes the development of a single-tube touch-down polymerase chain reaction (PCR) protocol for the detection of M. bovis using primers that target the IS6110 element. Spiked water and milk as well as routine diagnostic samples (milk and nasal swabs) from M. bovis-positive cattle were tested. This protocol allows the rapid and sensitive detection of M. bovis in bovine samples by enhancing the sensitivity of standard PCR amplification.  相似文献

Application of a theory of nuclear symmetry-based reaction restrictions to the O2 + O --> O3 reaction provides a potential explanation for the symmetry-induced isotopic enrichment observed for laboratory and atmospherically produced O3. Within this theory, the rate of formation of O3 from collisions of O and isotopically homonuclear O2 depends on whether the O2 molecule is in an f (allowed) or an e (restricted) parity label state. The restriction can be relaxed by various potential energy surface coupling terms, and the assumption that approximately 78 percent of the restricted O2(e) levels produce O3 with the same efficiency as the allowed O2(f) levels can account for laboratory-observed isotopic fractionation. In particular, the theory explains the special enhanced formation of the completely asymmetric isotopomer 16O17O18O.  相似文献

Bovine Group A Rotavirus (RVA) is one of the main causes of neonatal calf diarrhea worldwide. The present study reports the genotyping of bovine RVA strains circulating in Argentinean cattle from 2004 to 2010. Additionally, a new set of typing primers was designed and tested to differentiate between G8 and G6 (lineage III and IV) RVA strains. Bovine RVA was detected in 30% (435/1462) of the tested samples, corresponding to 49% (207/423) of the studied outbreaks with a similar detection rates in beef (53%; 67/127) and dairy herds (52%; 65/126). The RVA strains circulating in Argentinean cattle belonged to the common bovine genotypes G6 (lineages III and IV), G8, G10, P[5] and P[11]. A different RVA G/P-genotype distribution was found between the exploitation types, with the combination G6(IV)P[5] being by fare the most prevalent RVA strain in beef herds (58%), whereas a more even distribution of G6(III)P[11] (15%), G10P[11] (17%), G6(IV)P[5] (14%), and G6(IV)P[11] (6%) RVA strains was detected in dairy herds. G8 RVA strains were found in two dairy farms in calves co-infected with G8+G6(III)P[11]. A high percentage of co-infections and co-circulation of RVA strains with different genotypes during the same outbreak were registered in both exploitation types (20% of the outbreaks from beef herds and 23% from dairy herds), indicating a potential environment for reassortment. This finding is significant because G10P[11] and G6(III)P[11] strains may possess zoonotic potential. Continuous surveillance of the RVA strains circulating in livestock provides valuable information for a better understanding of rotavirus ecology and epidemiology.  相似文献

Foliar fertilization may be a viable strategy to boron supply in irrigated cropping systems with common beans (Phaseolus vulgaris), since it prevent B leaching. The aim of this work was to evaluate the economic viability and physiological parameters of the common beans production in irrigated cropping systems using sources and increasing foliar boron doses. A field experiment was carried out using an experimental block design in a factorial scheme 2?×?5?×?3, with two sources of B (boric acid and borax) and five doses: 0 (control), 2, 4, 6 e 8?kg?ha?1, with three repetitions. Foliar B applications were performed at 40 days after seeds germination, in pre-flowering stage. Physiological process (transpiration, stomatal conductance, CO2 internal concentration, net photosynthesis, and relative chlorophyll index), B level in leaves and grain yield were measured. These data were used to determine the economic viability of B fertilization in common beans. Both boric acid and borax increased B levels in common beans leaves. Borax affected some physiological process reducing stomatal conductance and increasing net photosynthesis. Using borax, the highest net photosynthesis was observed at a rate of 4?kg?ha?1, while the boric acid increased net photosynthesis linearly after increasing B doses application. An enhance of 311?kg?ha?1 in the grain yield was observed using borax related to the control (without B application); however, grain yield decreased linearly after application of increasing B doses, as boric acid. Comparing the economic viability of sources and doses of B, the highest profitability is obtained using borax at a rate of 4?kg?ha?1, which promoting a differential profit of US$534.44 per hectare compared to common beans cropping without B.  相似文献

Summary A reverse of the delaying effect of gibberellic acid (GA3) was observed in in vitro tuberization of potato plantlets when the medium contained 0.8×10−5 M jasmonic acid (JA). The promoting effect of JA seemed to be correlated with the initial absence of growing roots, probably through direct activity of JA in microtuber-producing buds in the stoloniferous shoots. This is based on the fact that JA did not inhibit root growth a posteriori. Endogenous gibberellins (GAs) synthesized by roots did not interfere with the previous activity of JA in tuberization. The absence of a JA promoting-effect on tuberization of previously-rooted plantlets could be related to the capacity of potato roots to synthesize endogenous GAs which might antagonize the JA effect on buds of stoloniferous shoots.  相似文献

Algarrobo (Prosopis alba) is an important nitrogen-fixing tree adapted to the semiarid regions of northwestern Argentina. The tree provides fuelwood and dimensionally stable lumber for flooring and furniture; its pods that have a high sugar content are consumed by livestock and humans. P. alba has been one of the most heavily harvested species. This paper reports on the evaluation of a nine-year-old Prosopis alba progeny trial containing 57 half-sibling families of eight provenances in northwestern Argentina (of the 1,596 trees planted, 1,289 survived in 1999). Considering the multiple uses of P. alba, the selection criteria included: total biomass production (from basal diameter using regression equations), height, rate of pod production, and pod sensory characteristics. The family-narrow-sense-heritability was 0.487 for height. 0.548 for biomass production, and 0.244 for pod production. In 1998, 12 of the 1,289 trees were more than 4 m tall and had more than 1.75 kg pods per tree; and their pods had a sweet or very sweet non-astringent taste. Scions from these 12 trees were successfully grafted onto unselected P. alba rootstock. These are currently being used to produce rooted cuttings. The pod production, biomass, and height of the 12 clones ranged from 6.55 to 14.4, 1.57 to 13.5, and 1.29 to 1.70 times the respective population means. The genetic gains of 13.5%, 77%, and 147% for height, biomass, and pod production respectively are greater than genetic gains for other tree species and are probably due to the great genetic variability in the local population and the lack of any prior genetic improvement program.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献
