New biodegradable cellulose acetate/layered silicate grafted poly(ε-caprolactone) [(CA/layered silicate)-g-PCL] nanocomposites were prepared by in situ polymerization of ε-caprolactone in the presence of cellulose acetate (CA) and organically modified layered silicate (OMLS). The structures of the resulting composites were investigated. X-ray diffractometry was carried out to survey general structural features of (CA/OMLS)-g-PCL nanocomposites, and revealed that OMLSs having hydroxyl groups in the organic modifiers greatly altered the layered silicate structure by monomer intercalation and successive exfoliation through its polymerization. Two of the representative cases were characterized by wide-angle and small-angle X-ray scattering analyses with a synchrotron source. The morphology of these nanocomposites was further examined by transmission electron microscopy. When SPN, one of OMLSs having one hydroxyl group in its modifier, was used, the silicate layers could not be dispersed thoroughly, but existed as aggregates consisting of several silicate layers. Among them, the crystal growth of PCL developed by transcrystallization, where the crystal growth was restricted in the confined space. When Cloisite 30B, having two hydroxyl groups within the modifier, was used, the silicate layers forming the clay were dispersed completely in the composite and random orientation of the OMLS was observed.  相似文献

Lignin is an integral component of the cell wall of vascular plants. The mechanism of supply of lignin precursors from the cytosol into the cell wall of differentiating xylem has not yet been elucidated. The present study showed that a certain amount of coniferyl alcohol glucoside (coniferin) occurred in the differentiating xylem of Japanese cypress (Chamaecyparis obtusa), as previously reported in gymnosperms. Coniferin content peaked in the early stages of secondary wall formation and decreased during lignification. In contrast to gymnosperms, coniferin content was limited in the differentiating xylem of poplar (Populus sieboldii × Populus grandidentata). Moreover, coniferyl alcohol was not detected in all specimens. In the differentiating xylem of poplar, a higher amount of sinapyl alcohol occurred than glucoside (syringin). However, the phloem contained syringin and not sinapyl alcohol. The sinapyl alcohol content in the xylem peaked in the cells with ceasing cell wall formation, and decreased gradually towards the boundary of the annual ring, where the lignin content kept increasing. Sinapyl alcohol in the differentiating xylem of poplar may be used for the lignification of the xylem.  相似文献

The lignification process in poplar tension wood lignified cell wall layers, specifically the S(1) and S(2) layers and the compound middle lamella (CML), was analysed using ultraviolet (UV) and transmission electron microscopy (TEM). Variations in the thickness of the gelatinous layer (G-layer) were also measured to clarify whether the lignified cell wall layers had completed their lignification before the deposition of G-layers, or, on the contrary, if lignification of these layers was still active during G-layer formation. Observations using UV microscopy and TEM indicated that both UV absorbance and the degree of potassium permanganate staining increased in the CML and S(1) and S(2) layers during G-layer formation, suggesting that the lignification of these lignified layers is still in progress during G-layer formation. In the context of the cell-autonomous monolignol synthesis hypothesis, our observations suggest that monolignols must go through the developing G-layer during the lignification of CML and the S(1) and S(2) layers. The alternative hypothesis of external synthesis (in the rays) does not require that monolignols go through the G-layer before being deposited in the CML, or the S(1) and S(2) layers. Interestingly, the previous observation of lignin in the poplar G-layer was not confirmed with the microscopy techniques used in the present study.  相似文献

Peroxidases are the major candidate enzymes involved in dehydrogenative polymerization of monolignols. Peroxidases have the signal sequence at their N-terminus and this suggests that they are transported to extracellular spaces or developing cell walls. In this study, we focused on an anionic peroxidase isozyme encoded by prxA3a, which seems to be related to lignification. To investigate the localization of peroxidase in differentiating xylem cells of poplar (Populus sieboldii × Populus grandidentata), anti-PRX3 antibody was raised against the anionic peroxidase. Western blotting and peroxidase activity inhibition assay showed specificity of the antibody. Labeling by anti-PRX3 antibody was localized in vessels and fibers during the secondary wall formation and was observed along the plasma membrane beside the microtubules. The labeling was not seen in the cell wall, where localization of peroxidases was expected during lignification. The peroxidase isozyme, which is suggested to be involved in monolignol polymerization, is localized on the plasma membrane and its localization might be regulated by microtubules.  相似文献

Summary Barley plants are able to accumulate glycinebetaine (betaine) at high levels in their leaves in response not only to water and salt stress but also to cold stress. Such accumulation of betaine during acclimation to cold is associated to some extent with freezing tolerance in leaves of barley plants, as previously demonstrated with near-isogenic lines that differed only in a single gene for the spring type of growth habit (Plant, Cell and Enyironment 17: 89–95, 1994). We now present evidence that the levels of betaine accumulated during cold acclimation might be associated with the earliness or lateness of the maturity of cultivars, namely, that late cultivars accumulate more betaine than early cultivars. Moreover, the grade of the vermalization requirement of the cultivars seemed unlikely to be associated with the level of betaine acumulated during cold acclimation. However, the trait that controlled accumulation of betaine during cold acclimation was not linked with the earliness or lateness of the maturity of cultivars. The higher levels of betaine in the late cultivars might have resulted from co-selection for lateness of maturity and freezing tolerance, which is generally a requirement in the areas of Japan where such late cultivars were originally cultivated.  相似文献

Oil production from oil palm is adversely affected by drought and salt. Under drought and salt stress, proline content increases in oil palm; the mechanism for this is unknown. Here, an 8319-nucleotide sequence including cDNA, genomic DNA and the promoter region of proline transporter gene from oil palm Elaeis guineensis was determined. The transporter gene exhibited high similarity to Bet/ProT genes from several plants, but the highest homology was found with rice ProT1. The exon-intron structure of genomic DNA was unique, and numerous stress-response cis-elements were found in the promoter region. Expression of cDNA EgProT1 in Escherichia coli mutant exhibited uptake activities for glycinebetaine and choline as well as proline. Under salt-stressed conditions, exogenously applied glycinebetaine was taken up into the root more rapidly than the control. These data indicate that oil palm has a unique Pro/T1 gene. Nucleotide sequence data for the cDNA and genomic DNA of proline transporter gene from Elaeis guineensis are available in the DDJB database under accession numbers AB597035 and AB597036, respectively.  相似文献

Arrangement of the cambium and marginal zone of xylem were illustrated by drawing the outlines of fusi-form cells and tissue on serial tangential sections inZelkova serrata andRobinia pseudoacacia with storied cambium and inAlbizia julibrissin with non-storied cambium. The comparison of cambium drawings with parenchyma drawings revealed that the lengths and arrangement of parenchyma strands correspond closely with those of corresponding fusiform initials in all three species with storied or non-storied cambium. Based on this result, we deduced the correlation between length and variation of length and arrangement of the fusiform initials in 20 species of Japanese hardwoods using parenchyma drawings instead of cambium drawings. The species with a smaller variation of length tend to exhibit a storied arrangement regardless of the length of the fusiform initial. Thus, it is suggested that the variation of the length of the fusiform initial has more influence than the length itself on the development of the storied arrangement.  相似文献
