BACKGROUND: The cotton bollworm, Helicoverpa armigera (Hübner), is one of the most serious insect pests of cotton. It has developed resistance to almost all groups of chemical insecticides because of their intensive use. The failure of insecticides to control H. armigera has been a strong incentive for the adoption of transgenic cotton (Bt cotton). However, the value of Bt could be diminished by widespread resistance development to Bt toxins in insect populations. Therefore, understanding the genetic basis of resistance is essential for developing and implementing strategies to delay and monitor resistance. RESULTS: A resistant strain designated as BM‐R was obtained from the cross of adults from Bathinda () and Muktsar (), Punjab, India, which showed the highest survival (60.68%) and LC50 value (1.396 µg mL?1 diet). Similarly, a laboratory‐maintained strain from Hoshiarpur, Punjab, showed maximum susceptibility to Cry1Ac toxin with the lowest LC50 value (0.087 µg mL?1), and was designated as HP‐S. The genetic purity of both strains was confirmed by RAPD profile analysis at each generation, and genetic similarity reached more than 90% after the third generation. Continuous maintenance of the resistant BM‐R strain on Cry1Ac resulted in an increase in LC50 from 0.531 µg mL?1 in F0 to 4.28 µg mL?1 in F14 and 7.493 µg mL?1 in F19, while the LC50 values for HP‐S larvae on diet without Cry1Ac increased to 0.106 and 0.104 µg mL?1, which lay within the fiducial limits of the baseline LC50 value. The mode of inheritance of resistance was elucidated through bioassay response of resistant, susceptible heterozygotes and backcross progeny to Cry1Ac incorporated in semi‐synthetic diet. CONCLUSION: Based on dominance, degree of dominance and backcross values, resistance was inferred to be polygenic, autosomal and inherited as a recessive trait. QTL analysis for grain weight in common wheat   总被引:6,自引:0,他引:6  
Quantitative trait loci (QTL) analysis for grain weight (GW = 1000 grain weight) in common wheat was conducted using a set of 100 recombinant inbred lines (RILs) derived from a cross ‘Rye Selection 111 (high GW) × Chinese Spring (low GW)’. The RILs and their two parental genotypes were evaluated for GW in six different environments (three locations × two years). Genotyping of RILs was carried out using 449 (30 SSRs, 299 AFLP and 120 SAMPL) polymorphic markers. Using the genotyping data of RILs, framework linkage maps were prepared for three chromosomes (1A, 2B, 7A), which were earlier identified by us to carry important/major genes for GW following monosomic analysis. QTL analysis for GW was conducted following genome-wide single marker regression analysis (SMA) and composite interval mapping (CIM) using molecular maps for the three chromosomes. Following SMA, 12 markers showed associations with GW, individual markers explaining 6.57% to 10.76% PV (phenotypic variation) for GW in individual environments. The high grain weight parent, Rye Selection111, which is an agronomically superior genotype, contributed favourable alleles for GW at six of the 12 marker loci identified through SMA. The CIM identified two stable and definitive QTLs, one each on chromosome arms 2BS and 7AS, which were also identified through SMA, and a third suggestive QTL on 1AS. These QTLs explained 9.06% to 19.85% PV for GW in different environments. The QTL for GW on 7AS is co-located with a QTL for heading date suggesting the occurrence of a QTL having a positive pleiotropic effect on the two traits. Some of the markers identified during the present study may prove useful for marker-assisted selection, while breeding for high GW in common wheat.  相似文献

The blood pressure lowering effect of a pea protein hydrolysate (PPH) that contained <3 kDa peptides, isolated by membrane ultrafiltration from the thermolysin digest of pea protein isolate (PPI), was examined using different rat models of hypertension as well as hypertensive human subjects. The PPH showed weak in vitro activities against renin and angiotensin converting enzyme (ACE) with inhibitory activities of 17 and 19%, respectively, at 1 mg/mL test concentration. Oral administration of the PPH to spontaneously hypertensive rats (SHR) at doses of 100 and 200 mg/kg body weight led to a lowering of hourly systolic blood pressure (SBP), with a maximum reduction of 19 mmHg at 4 h. In contrast, orally administered unhydrolyzed PPI had no blood pressure reducing effect in SHR, suggesting that thermolysin hydrolysis may have been responsible for releasing bioactive peptides from the native protein. Oral administration of the PPH to the Han:SPRD-cy rat (a model of chronic kidney disease) over an 8-week period led to 29 and 25 mmHg reductions in SBP and diastolic blood pressure, respectively. The PPH-fed rats had lower plasma levels of angiotensin II, the major vasopressor involved in development of hypertension, but there was no effect on plasma activity or renal mRNA levels of ACE. However, renal expression of renin mRNA levels was reduced by approximately 50% in the PPH-fed rats, suggesting that reduced renin may be responsible for the reduced levels of angiotensin II. In a 3-week randomized double blind placebo-controlled crossover human intervention trial (7 volunteers), significant (p<0.05) reductions (over placebo) in SBP of 5 and 6 mmHg were obtained in the second and third weeks, respectively, for the PPH group. Therefore, thermolysin derived bioactive peptides from PPH reduced blood pressure in hypertensive rats and human subjects, likely via effects on the renal angiotensin system.  相似文献

Twelve synthesized ainphiplonds involving Karnal bunt (Neovossia indica)-resisiant accessions of Triticum monococum, T. boeoticum and Aegilops sqiwrrosa and susceptible but otherwise well adapted and high yielding T. Durum cultiviars were evaluated for Karnal bunt resisiance under artificial inoculation conditions. All ihe synthetic amphiploids, except DWI. 5031 x T. monocoirtum aniphlploid, were free from Karnal bunt disease indicating that the Karnal bunt resistance or T. motsococcum, T. boeoticMrn and Ae, squarrosa is expressec in the presence of the dnrum complement. The importance and utilization of the amphiploids fox breeding wheat varieties resistant to karnal bunt are discussed.  相似文献

An attempt was made to assess the effect of dietary pyridoxine supplementation on growth performance and cellular metabolic enzymes of Chanos chanos fingerlings under endosulfan induced stress. Two‐hundred and twenty‐five fish were randomly distributed into five treatments, each with three replicates and fed four isocaloric and isonitrogenous diets with graded levels of pyridoxine for 45 days. Endosulfan concentration in the water was maintained at sublethal level of 1/40th of 96 h LC50 (0.52 ppb). Dietary pyridoxine supplementation significantly enhanced growth performance viz. weight gain (%), specific growth rate, feed conversion ratio and protein efficiency ratio of fish compared to control diet fed group. The results of this experiment revealed that the activities of digestive and metabolic enzymes were significantly improved after feeding with dietary pyridoxine supplementation in C. chanos. Significantly increased muscle and brain ascorbic acid content was observed in fish fed diets containing pyridoxine. Histological studies on liver of fish fed pyridoxine‐supplemented diet showed the normal architecture of hepatocytes compared to non‐pyridoxine fed group. The results obtained in this study revealed that the inclusion of pyridoxine in diet has a protective effect against endosulfan induced stress in C. chanos fingerlings through metabolic enzyme activities and vitamin C content. This study has a potential in ameliorating the toxic effect of endosulfan induced stress using dietary supplementation of pyridoxine.  相似文献

This paper describes atmospheric pressure ionization (API) LC-MS-MS determination of urushiols, 3-n-alkenyl- and -alkyl-substituted catechols responsible for poison oak dermatitis. Urushiol was isolated from Western poison oak according to the method of Elsohly et al. (1) (J. Nat. Prod. 1982, 45, 532-538)-the purified preparation contained C(17)- and C(15)-substituted urushiols with zero, one, two, and three double bonds as determined from GC-MS analysis of trimethylsilyl derivatives. Urushiol mixtures were separated on a C(18) reversed phase HPLC column with a methanol-water gradient with urushiols eluting in 100% methanol. Atmospheric pressure chemical ionization (APCI) produced primarily [M - H](-) and MH(+) molecule ions. Electrospray ionization (ESI) yielded [M - H](-) and adduct ions including [M + Cl](-). Daughter ions of [M - H](-) included quinoid radical anions ([M - H - H(2)](-) and m/z 122(-)) and a benzofuran phenate (m/z 135(-)). A suite of hydrocarbon fragments were produced by collision-induced dissociation of MH(+) directly or via an intermediate [MH - H(2)O](+) daughter ion. Six urushiol congeners, one not previously reported in poison oak, were determined by negative ion API-LC-MS-MS with detection limits of approximately 8 pg/microL (ESI) and approximately 800 pg/microL (APCI). API-LC-MS-MS was used to determine urushiol in surface wipes, air samples, and plant materials.  相似文献

Arbuscular mycorrhizal (AM) fungi alleviating the adverse salt effects on growth was tested in bajra (Pennisetum glaucum). Towards this objective we analyzed the photosynthetic activity, proline, phosphatase activity and various antioxidant enzymes such as peroxidase, catalase and superoxide dismutase at 0, 100, 200 and 300 mM salinity levels in AM inoculated and non inoculated bajra plants. Total chlorophyll content was significantly higher in moderate salinity condition in AM fungus (Glomus fasciculatum) inoculated plants, which lead to increase in growth and nutrient uptake capacity of mycorrhizal plants. Antioxidant activity was either increased or decreased due to responses to different salinity stress conditions. Proline accumulation was induced by salt and it was more in shoot of non-AM inoculated plants, but in roots proline accumulation was higher in AM plants at all levels of salinity. This work suggests that the AM fungus helps bajra plants to perform better under moderate salinity levels by enhancing the antioxidant activity and proline accumulation as compared to non-mycorrhizal plants.  相似文献

The present paper reviews the available literature on the allelopathy of gymnosperms which constitute the famous boreal forests known for their high quality timber and other products of great commercial importance. Nineteen genera belonging to six of the seven taxonomic families of conifers comprising one of the most dominating and largest group of present day gymnosperms, are reported to show the phenomena of allelopathy. The available information signifies the role these mighty trees play in determining the understorey vegetation, ecological succession and forests dynamics besides causing regeneration problems to the donor trees. The allelochemicals, identified from these plants, responsible for such interactions have also been summarised. In most of the cases the leaching of phenolics from the litter predominantly of tree needles, which remain on the ground for a longer time due to slow decomposition rate, are held responsible for these interactions.  相似文献
