Yellow mosaic disease (YMD) of legumes endemic to South Asia are caused by begomoviruses transmitted by whiteflies. Based on molecular characterization, two distinct viruses – Mungbean yellow mosaic India virus (MYMIV) and Mungbean yellow mosaic virus (MYMV) – were found previously to be the etiological agents of YMD in legumes. Here, host range studies with a soybean isolate of MYMIV (MYMIV-[Sb]) were carried out by both whitefly transmission and agroinoculation. MYMIV-[Sb] was similar to a cowpea isolate of MYMIV (MYMIV-[Cp]) in its ability to infect cowpea, thus differing from blackgram (MYMIV) and mungbean (MYMIV-[Mg]) isolates, which do not infect cowpea. Genomic analysis of DNA A and DNA B components of these MYMIV isolates show characteristic differences in complete DNA B nucleotide sequence correlating with host range differences.  相似文献

The giant fresh water prawn Macrobrachium rosenbergii is known to be highly tolerant to white spot syndrome virus (WSSV) infections when compared to the widely cultured marine tiger shrimp Penaeus monodon. At present, the exact mechanism of tolerance by M. rosenbergii to WSSV is not known. In this study, we attempt to study the effect of WSSV injections on the hemagglutination activity of the hemolymph serum of both P. monodon and M. rosenbergii and look for changes if any, on their hemolymph serum protein electrophoretic patterns. Our results show that M. rosenbergii had significantly (p < 0.05) higher hemagglutinating activity against mouse erythrocytes when compared to P. monodon. As the infection progressed to 48 h there was a further increase (p < 0.05) in the hemagglutination activity in M. rosenbergii, while it decreased in P. monodon. 12% SDS-PAGE analysis of the hemolymph serum of M. rosenbergii infected with WSSV did not show any new protein bands, whereas few bands with decreased intensity was observed in moribund P. monodon where the hemagglutinating activity was also observed to be decreased. The results indicate that hemolymph hemagglutinin levels are modulated in crustaceans as a response to viral infections.  相似文献

Sixteen Pallic (Aqualfs, Ustalfs, Aquepts, Ochrepts) and related soils collected from the North and South Islands of New Zealand were studied to determine if variations in nonexchangeable K supply could be explained by mineralogical composition. A strong relationship was found which indicates that mica is weathering to vermiculite. Proportions of vermiculite and kandite are generally higher in the North Island than in the South Island. Dominance of either micas or vermiculite or kandite is determined by the stage of weathering and/or the nature of parent materials. Soils that supplied more nonexchangeable K (Knex) to ryegrass plants contained more mica in the clay fraction than soils that supplied less Knex. The results confirmed that use of a soil test that includes a measure of Knex (i.e. acid-extractable K) may be a vital part of identifying variations in plant available K status of the soils. It is concluded that the K supplying power of the Pallic and related soils is related directly to the amounts of mica present in clay fraction and that good K supplying soils will be transformed to K depleted soils as a result of increased weathering and leaching (pedogenic factors) and K exploitation in intensive farming systems.  相似文献

Using dry dog food label information, the hypothesis was tested that the risk of gastric dilatation-volvulus (GDV) increases with an increasing number of soy and cereal ingredients and a decreasing number of animal-protein ingredients among the first four ingredients. A nested case-control study was conducted with 85 GDV cases and 194 controls consuming a single brand and variety of dry food. Neither an increasing number of animal-protein ingredients (P=0.79) nor an increasing number of soy and cereal ingredients (P=0.83) among the first four ingredients significantly influenced GDV risk. An unexpected finding was that dry foods containing an oil or fat ingredient (e.g., sunflower oil, animal fat) among the first four ingredients were associated with a significant (P=0.01), 2.4-fold increased risk of GDV. These findings suggest that the feeding of dry dog foods that list oils or fats among the first four label ingredients predispose a high-risk dog to GDV.  相似文献

Summary Semilooper resistant transgenic castor plants were produced through Agrobacterium-mediated genetic transformation method. Two castor cultivars, Jyothi and VP1 were transformed using the super-binary vector pTOK233 carrying gus A and hpt genes. Putative transformants were regenerated following selection on the hygromycin containing medium. GUS positive primary transformants, when subjected to Southern analysis, revealed stable integration of gus A into their genomes. In the T1 generation, a monogenic segregation ratio of 3 GUS positive: 1 GUS negative plants was observed. Furthermore, transformation experiments were carried out with the Agrobacterium pSB111 super-binary vector carrying a synthetic delta endotoxin gene cryIAb and the herbicide resistance gene bar both driven by cauliflower mosaic virus 35S promoter. Putative transformants were regenerated through selection on the phosphinothricin containing medium and Basta tolerant transformants were subjected to molecular analysis. PCR analysis revealed the presence of both bar and cryIAb genes in the Basta tolerant primary transformants. Southern analysis of PCR positive plants with cryIAb probe showed a 3 Kb band upon HindIII digestion and a > 6 Kb band with BamHI digestion, thus suggesting stable integration of cryIAb intact expression cassette and independent nature of the transformants. The primary transformants subjected to ELISA disclosed varied levels of Cry protein. These transgenics expressing cryIAb – when bioassayed against freshly hatched semilooper larvae – induced substantial (> 88%) insect mortality. Southern analysis of 2T1 plants revealed the presence of cryIAb gene, indicating stable inheritance of the transgene into the next generation. In T1, all the Southern-positive plants for cryIAb invariably exhibited tolerance to Basta, denoting co-segregation of both bar and cryIAb genes. Introgression lines (ILs) derived from interspecific crosses are a source of new genetic variability. A total of 55 ILs derived from two crosses Swarna × O. nivara IRGC81848 (population A) and Swarna × O. nivara IRGC81832 (population B) were characterized for yield and yield-related traits/QTLs. Segregation of 103 simple sequence repeat (SSR) markers associated with yield-related QTLs was studied. Population A showed an average of 12.6% homozygous O. nivara alleles and population B showed 10.6%. Interestingly, three SSR markers, RM223, RM128 and RM517, showed conspicuous pattern of segregation. The distribution of parental alleles at three loci RM223, RM128 and RM517 linked to yield-related traits was unique. These markers flanked to several yield-related QTLs. RM223, flanking to qyld8.3, was heterozygous in almost all the 55 ILs except in IL10-3S and IL131S. RM128 on chromosome 1 and RM517 on chromosome 3 were mutually exclusive in 46 out of 55 ILs. These 46 ILs showed either of the marker allele RM128 or RM517 from O. nivara but not both. IL166S had both RM128 and RM517 from O. nivara and the other ILs showed homozygous Swarna allele at RM517 except IL65S. Population structure assigned the 55 ILs to three sub-populations based on their genomic diversity. IL65S, IL166S, IL248S, IL7K and IL250K showed high yields in multi-location trials, and IL248S was released for cultivation as DRRDhan 40.  相似文献

Bemisia tabaci is a cryptic species complex, causing significant loss on many agriculturally important crops worldwide. Knowledge on species composition and diversity within B. tabaci complex is critical for evolving sustainable pest management strategies. Here we investigate the whitefly species complex in soybean in major soybean growing states of India. The mitochondrial cytochrome oxidase gene subunit-1(mt COI) based phylogenetic relationships established using Bayesian methods indicated the existence of three cryptic species namely AsiaⅠ, AsiaⅡ1, and AsiaⅡ7. All the haplotypes detected in the study could be assigned to these three cryptic species following the species demarcation criteria of 3.5% divergence threshold. Of these, AsiaⅡ1 was found to be predominant with wide spread distribution across the surveyed regions from cool temperate zones to hot and humid tropical plains. On the contrary, cryptic species AsiaⅡ7 showed localized distribution. The AsiaⅡ1 exhibited the highest haplotype diversity and AsiaⅠ showed high level of nucleotide diversity. There was a significantly high genetic differentiation among these three cryptic species. The MEAM 1, a dreadful invasive species was not detected in the specimens tested in the current study. The diversity and distribution of three cryptic species is discussed in the light of current knowledge on distribution of whitefly species in India and yellow mosaic disease observed during sampling survey.  相似文献

A nested case-control study was conducted among 1634 dogs with complete diet information in a 5-year prospective study to determine diet-related risk factors for gastric dilatation-volvulus (GDV). Cases included 106 dogs that developed GDV; controls included 212 dogs without GDV that were frequency matched to cases by year of GDV onset. Proportionate energy consumed from major food types and from carbohydrates was determined. Dogs were categorized as consuming either a low volume or high volume of food based on the median number of cups of food fed per kg of body weight per meal. Dogs fed a larger volume of food per meal were at a significantly (P<0.05) increased risk of GDV, regardless of the number of meals fed daily. For both large- and giant-breed dogs, the risk of GDV was highest for dogs fed a larger volume of food once daily.  相似文献
