Fifteen and eight mature beagles, without (normal group) and with experimental mitral regurgitation (MR group), respectively, were given 0.02 mg/kg/day digoxin powder for 10 days orally. The optimum time for sample collection after administration of digoxin was observed to be 8-18 hr and 10-22 hr in the normal and MR groups, respectively. In both groups, a stable concentration was reached after 3-5 days of treatment. No differences in plasma level were observed between sexes. The optimum concentration of digoxin was attained at an earlier stage than has been previously reported for both dogs and humans.  相似文献

In order to elucidate the relationship between cellular injury and lipid peroxidation induced by hexavalent chromium (CrVI), isolated rat hepatocytes treated with any one of scavengers of active oxygen species, antioxidants or antichromium agent were incubated with K2Cr2O7 as CrVI (1 mM Cr). After the incubation, the development of lipid peroxidation was determined as thiobarbituric acid (TBA)-reacting materials in total lipid extracts from the incubated hepatocytes. Cellular injury was observed as a leakage of lactate dehydrogenase (LDH) from hepatocytes into incubation medium. The contents of reduced glutathione (GSH) in hepatocytes were also assessed. Results obtained were as follows: (1) CrVI facilitated lipid peroxidation in isolated hepatocytes after 20 min of incubation. On the other hand, the cellular injury induced by CrVI was barely observed even after 60 min of incubation. (2) The CrVI-induced lipid peroxidation was inhibited by catalase and mannitol as scavengers of active oxygen species, or N,N'-diphenyl-p-phenylenediamine and alpha-tocopherol as antioxidants. However the cytotoxicity of CrVI could not be prevented by these chemicals. (3) CrVI depleted the contents of intracellular GSH and diminished the activities of glutathione reductase (GR) and glutathione-S-transferase (GST) except glutathione peroxidase. (4) The scavengers of active oxygen species and the antioxidants could not prevent the depletion of intracellular GSH induced by CrVI. (6) Ascorbic acid, antichromium agent, prevented all of the lipid peroxidation, the cellular injury and intracellular GSH depletion induced by CrVI.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The DNDC (DeNitrification-DeComposition)-Rice model, one of the most advanced process-based models for the estimation of greenhouse gas emissions from paddy fields, has been discussed mostly in terms of the reproducibility of observed methane (CH4) emissions from Japanese rice paddies, but the model has not yet been validated for tropical rice paddies under alternate wetting and drying (AWD) irrigation management, a water-saving technique. We validated the model by using CH4 and nitrous oxide (N2O) flux data from rice in pots cultivated under AWD irrigation management in a screen-house at the International Rice Research Institute (Los Baños, the Philippines). After minor modification and adjustment of the model to the experimental irrigation conditions, we calculated grain yield and straw production. The observed mean daily CH4 fluxes from the continuous flooding (CF) and AWD pots were 4.49 and 1.22?kg?C?ha?1?day?1, respectively, and the observed mean daily N2O fluxes from the pots were 0.105 and 34.1?g?N?ha?1?day?1, respectively. The root-mean-square errors, indicators of simulation error, of daily CH4 fluxes from CF and AWD pots were calculated as 1.76 and 1.86?kg?C?ha?1?day?1, respectively, and those of daily N2O fluxes were 2.23 and 124?g?N?ha?1?day?1, respectively. The simulated gross CH4 emissions for CF and AWD from the puddling stage (2 days before transplanting) to harvest (97 days after transplanting) were 417 and 126?kg?C?ha?1, respectively; these values were 9.8% lower and 0.76% higher, respectively, than the observed values. The simulated gross N2O emissions during the same period were 0.0279 and 1.45?kg?N?ha?1 for CF and AWD, respectively; these values were respectively 87% and 29% lower than the observed values. The observed total global warming potential (GWP) of AWD resulting from the CH4 and N2O emissions was approximately one-third of that in the CF treatment. The simulated GWPs of both CF and AWD were close to the observed values despite the discrepancy in N2O emissions, because N2O emissions contributed much less than CH4 emissions to the total GWP. These results suggest that the DNDC-Rice model can be used to estimate CH4 emission and total GWP from tropical paddy fields under both CF and AWD conditions.  相似文献

ABSTRACT:   The ability of salmon growth hormone (sGH) to accelerate the somatic growth of juvenile abalone Haliotis discus hannai by immersion was examined. After immersion for one hour into a sGH-rich solution at a concentration of 30 mg/L, the sGH immunoreactivity in the body fluid of abalone was maximal after one day, and levels were still detectable at two days. No immunoreactivity was observed in the control group. Following immersion for one hour at one or two-week intervals for 120 weeks into the sGH-rich solution at 30 mg/L, the sGH-immersed abalone exhibited a significant increase in shell length and body weight. On the other hand, abalone immersed into a salmon prolactin (sPRL)-rich solution at 30 mg/L for one hour showed a lower increase in both shell length and body weight than the sGH-immersed abalone. No increases in shell length and body weight were observed in either the bovine serum albumin-immersed abalone or untreated controls. These results provide evidence that sGH can be transported from ambient water into the circulatory system of abalone, and can subsequently improve the somatic growth of juveniles.  相似文献

We investigated the morphological effects of testosterone on placental development in a rat model of polycystic ovarian syndrome (PCOS). Testosterone propionate (TP), which was subcutaneously administered to pregnant rats with 5 mg/animal from gestation day (GD) 14 to GD 18, induced a maternal weight reduction without mortality or clinical signs from GD 19 onwards. A decrease in fetal and placental weight, an increase in intrauterine growth retardation (IUGR) rates, and histological changes in the placenta were observed on GD 21 but not on GD15 or 17. Histopathologically, on GD 21, the trophoblast septa thickened, and the maternal sinusoids were narrowed in the labyrinth zone, resulting in a small placenta. Additionally, the placental weight, thickness, and histological morphology in the labyrinth zone on GD 21 in the TP-treated group were nearly identical to those on GD 17 in the control and TP-treated groups. Therefore, it was assumed that the testosterone-induced small placenta was induced in association with the developmental inhibition of the fetal part of the placentas from GD 17 onwards.  相似文献

We have previously established the method for isolation of ceramide aminoethylphosphonate (CAEP) from jumbo flying squid Dosidicus gigas. In this study, we performed a MTT assay to evaluate the safety of CAEP to the cell lines for the application to health food and supplements. The CAEP did not show any cytotoxicity to various HEK293-transfectant cells. Next, we elucidated the positive function of CAEP to the somatic cells. Recently, we have reported that hepatotoxin microcystin-LR was taken up into the hepatocytes mediated by hepatocellular uptake transporters OATP1B1 and OATP1B3, and the cells were induced cytotoxicity subsequently. Cytotoxicity of microcystin-LR to permanently OATP1B3-expressing HEK293-OATP1B3 cells rather than to HEK293-OATP1B1 cells was preferentially attenuated by CAEP in a concentration-dependent manner. In addition, the enzyme activity of serine/threonine phosphatase, which was inhibited by microcystin-LR, was recuperated by co-exposure to CAEP. Furthermore, microcystin-LR-induced cellular protein phosphorylation were disrupted by CAEP exposure. These results suggested that CAEP is a promising remedy and/or preventive medicine for liver damage with microcystin-LR.  相似文献

A detailed list of dinoflagellate species for the Mergui Archipelago, off southern Myanmar, was compiled for the first time. This was based on surveys conducted in two seasons in pre- and post-monsoons, and on observations for both plankton and cyst assemblages. In the plankton samples, 57 and 26 dinoflagellate species were recorded, respectively, in the pre- and post-monsoons. Among the list from plankton and cyst samples, harmful species were found: red-tide species including Prorocentrum spp. and Alexandrium affine, and causative species of paralytic shellfish poisoning, e.g., Alexandrium tamiyavanichii and Gymnodinium catenatum, and of diarrhetic shellfish poisoning, e.g., Dinophysis spp. Therefore, although no incidences have been reported yet, attention must be paid to these harmful algal bloom (HAB) events in Myanmar where exploitation of fishery resources is drastically increasing. Other dinoflagellate compositions were also unique and may relate to the oceanographic system in this region. In the pre-monsoon, rather rare oceanic species of Ornithocercus spp. were found concurrently with neritic species, probably due to the extensive southwesterly current from the Indian Ocean. Throughout the seasons, diverse species of Protoperidinium were predominately recorded in both planktonic and sediment samples, indicating richness of prey organisms probably sustained by upwelling systems.  相似文献

In the present study, the daily excretion of potassium (K) in urine (urinary K(UK)) was estimated from a 6 h urine sample using urinary creatinine (UC) as the index substance. All urine was collected from six pregnant Holstein cows at 6 h intervals for 24 h on 3 days of the 4th, 2nd and final week before the expected date of parturition. In total, 72 6 h urine samples were obtained. Daily UC excretion (mg/day per kg bodyweight (BW)) was almost the same for the three sampling days. Daily UC excretion varied among cows from 22.1 to 24.3 mg/day per kg BW with a mean of 22.8 mg/day per kg BW with no significant difference. Thus, daily UC excretion was confirmed to be constant throughout the prepartum period with no differences among individuals. The concentration ratios of K to creatinine ((UK mg/dL)/(UC mg/dL) (UK/UC)) correlated strongly to the hourly K excretions (mg/h per kg BW) (r = 0.952, P < 0.01) in the 6 h urine samples. The differences in the UK/UC ratio between sampling periods were not significant within each cow. Therefore, daily UK excretion (mg/day) can be estimated using the equation: daily UK excretion (mg/day) = daily UC excretion (mg/day per kg BW) × BW (kg) × 6 h urine sample UK/UC, where daily UC excretion can be a given value.  相似文献

A pinealoma (benign) was found in a 61-week-old male Crj:CD (SD) IGS rat. The neoplasm was located between the cerebral hemispheres and the cerebellum. Histologically, the tumor cells consisted of two cell types: large, pale-staining cells and small dark-staining cells. A fibrovascular stroma divided the tumor cells into incomplete lobules or nest structures. Relatively numerous mitoses were noted in the tumor cells. Ultrastructurally, the tumor cells contained dense-cored vesicles, approximately 120 nm in diameter.  相似文献
