Thirteen sexually mature captive male lesser Malay chevrotains (Tragulus javanicus) were each anesthetized twice with tiletamine-zolazepam for electroejaculation. Viable spermatozoa were collected from all animals. The semen was creamy, milky, pale yellowish, or watery. The mean values for ejaculate volume, sperm concentration, and percentages of sperm motility, normality and viability were 23.7 +/- 2.5 microl, 366.9 +/- 127.8 x 10(6) spermatozoa/ml, 40.0% +/- 3.1%, 71.4% +/- 1.6%, and 59.6% +/- 2.1%, respectively. Semen pH was 7-8. No adverse effects of electroejaculation were noted. These are the first reported values for semen of lesser Malay chevrotain. Electroejaculation should be usable for routine semen collection in this species.  相似文献

In contrast to conventional inbreeding that takes up to seven generations to develop inbred lines, the doubled haploid (DH) technology allows production of inbred lines in two generations. The objectives of the present study were to: (a) evaluate testcross performance of 45 doubled haploid lines under drought stress and non-stress conditions (b) estimate heritabilities for grain yield and other traits and (c) to assess the genetic distance and relationship among the DH lines using 163,080 SNPs generated using genotyping-by-sequencing (GBS). The 45 hybrid and five checks were evaluated using a 10 × 5 alpha lattice in six drought stress and nine well-watered environments in Kenya, Uganda, and Tanzania. Differences in trait means between the drought stress and well-watered conditions were significant for all measured traits except for anthesis date. Genetic variances for grain yield, grain moisture, plant height and ear height were high under well-watered environments while genetic variance for anthesis date, root lodging and stalk lodging were high under drought stress environments. Combined analyses across drought stress and well-watered environments showed that ten top hybrids produced 1.6–2.2 t/ha grain yield under well-watered condition and 1–1.4 t/ha under drought stress condition higher than the mean of the commercial checks. Genetic distance between pairwise comparisons of the 38 of the 45 DH lines ranged from 0.07 to 0.48, and the overall average distance was 0.36. Both cluster and principal coordinate analysis using the genetic distance matrix calculated from 163,080 SNPs showed two major groups and the patterns of group was in agreement with their pedigree. Thirteen (13) of the best hybrids are currently in National Performance Trials testing, an important step towards commercialization in Kenya, Tanzania and Uganda.  相似文献

The effects of different estrus synchronization techniques on follicular development and estrus response were studied in 81 nulliparous Boer does. The does were divided into nine groups. Eight of the nine groups were synchronized with prostaglandin F2-alpha (PGF(2α)) or flugestone acetate (FGA) or their combinations, and the ninth group was a control group. In addition to the above combinations, four of the eight synchronized groups were given 5?mg follicle-stimulating hormone (FSH) and the remaining four groups were administered 300?IU equine chorionic gonadotrophin (eCG). Posttreatment follicular development was monitored until ovulation occurred using a real-time B-mode ultrasound scanner (Aloka, 500 SSD, Japan), with a 7.5-MHz transrectal linear probe. All the does from the synchronized groups that were given eCG exhibited oestrus while only 88.9% of the does synchronized with FSH showed estrus. The estrus response was observed to be the least among the does synchronized with PGF(2α) + FSH (33.3%) combination followed closely by the FGA + FSH (42.9%) combinations. It was observed that the combinations of FGA + PGF(2α) + FSH resulted in increased percentage of estrus response, duration of estrus, and ovulation. The number of follicles was higher (P?相似文献

Striga is an obligate parasitic weed that attacks cereal crops in sub-Saharan Africa. In Western Kenya, it has been identified by farmers as their major pest problem in maize. A new technology, consisting of coating seed of imidazolinone resistant (IR) maize varieties with the imidazolinone herbicide, imazapyr, has proven to be very effective in controlling Striga on farmer fields. To bring this technology to the farmer, a sustainable delivery system needs to be developed, preferably with substantial participation of the private sector. To help extension agents and seed companies to develop appropriate strategies, the potential for this technology was analyzed by combining different data sources into a Geographic Information System (GIS). Superimposing secondary data, field surveys, agricultural statistics and farmer surveys made it possible to clearly identify the Striga-prone areas in western Kenya. Results found that Striga affected a maize area of 246,000 ha annually, with a population of 6.4 million people and maize production of 580,000 tons, or 81 kg/person. Population density in this area is high at 359 people/km2. A survey of 123 farmers revealed that 70% of them have Striga in their fields. A contingent valuation (CV) survey indicated that farmers would, on average, be willing to buy 3.67 kg of the IR-maize seed each at current seed prices, sufficient to sow 44% of their maize area. By extrapolation over the maize area in the zone, total potential demand for IR-maize seed is estimated at 2000–2700 tons annually. Similar calculations, but based on much less precise data and expert opinion rather than farmer surveys or trials, gives an estimate of the potential demand for IR-maize seed in Africa of 153,000 tons.  相似文献

Imidazolinone resistant maize seed dressed with imazapyr has been successfully used to control the parasitic weed Striga in many locations, and has begun to be commercially successful in Africa. Despite this, occasionally poor effectiveness of control had been documented in some sites that required explanation. Analysis of the data against rainfall patterns suggested that: 1. poor maize emergence occurred when there was limited rainfall at germination, possibly due to inhibition by a very high herbicide concentration too near the maize seed; 2. there was poor Striga control in seasons of very high rainfall, possibly due to herbicide washout away from the maize root zone where Striga germinates and attaches. These field assessments were matched by experiments suggesting that slow release formulations might alleviate the problems. A series of slow release formulations were synthesized based on binding imazapyr to high capacity anion exchangers and using them to coat maize seed. The best seems to be a polyethyleneimine gel. Epidemiological field data from a multitude of sites support the conclusion that the slow release formulations increased stand establishment across sites and seasons compared to the control where there was low rainfall.  相似文献

In the present study, various explants of Murraya paniculata (Jack) Linn., such as cotyledons, shoots and young stems were cultured on MS medium supplemented with various concentrations of Benzyl Amino Purine (BAP) under 25 +/- 1 degree C with 16 h light and 8 h dark and also 8 h light and 16 h dark to obtain complete plant regeneration. In vitro flowering was observed from shoot explants cultured on MS supplemented with 0.5-2.0 mg L(-1) Naphthalene Acetic Acid (NAA) and also on MS basal medium under similar conditions. The leaves and flowers obtained from both in vivo and in vitro conditions were examined and compared. Morphological studies such as leaf clearing, epidermal peeling were studied using light and scanning electron microscope. Macromorphological studies of the flowers produced from in vivo and in vitro conditions were also examined. Morphologically, there were no differences between in vivo and in vitro flowers except the flowers produced from tissue culture systems were smaller in size with protruding stigmas. Differences were also found in the number of layers of palisade cells and the presence or absence of epicuticle layer of the leaves. Leaves produced from tissue culture system were smaller in size with membranous texture. Stomata were present only on the abaxial surfaces of both in vivo and in vitro leaves but the stomata were raised above the epidermis in the latter.  相似文献
