The objectives of this study were to evaluate the use of potato (Solanum tuberosum L.) late blight (Phytophthora infestans (Mont.) de Bary) resistant parents in cultivar development and identify superior clones possessing moderate to high late blight resistance combined with acceptable maturity and tuber quality. Ninety-five crosses were made between eight unadapted parents with reported late blight resistance (B0718-3, Bertita, Bzura, Greta, Libertas, Stobrawa, Tollocan and Zarevo) and susceptible parents (cultivars or advanced breeding clones) adapted to North American growing conditions. A total of 408 field selected clones were assessed for late blight resistance in the greenhouse and in the field using a mixture of US8 P. infestans isolates (A2 mating type, metalaxyl resistant) that overcame all known R-genes except R8 and R9. Clones with ≤ 10% infected foliar area in the greenhouse test or ≤ 0.30 RAUDPC (relative area under the disease progress curve) value in the field in 1998 were re-tested in 1999. A total of 118 (29% of 408) putative late blight resistant clones were selected. The eight late blight resistant parents differed in both the ability to transmit late blight resistance and in the level of resistance transmitted to the progeny. The Tollocan and B0718-3 families (half-sib progeny) had the greatest degree of resistance and frequency of resistant clones. Scott-Knott cluster analysis ranked 79 clones (67% of 118) in the high and moderate late blight resistant groups. Among these 79 clones, 19 clones had vine maturity equal to or earlier than mid-season combined with acceptable tuber quality. Further selection in 2000 resulted in eight advanced selected clones (six from Tollocan and two from B0718-3 families) with the same level of resistance as the parent combined with vine maturity and tuber quality equivalent to Atlantic, a standard cultivar for chip processing in North America. The results indicate that this breeding approach can be used to select parents for late blight resistance breeding and to identify superior clones with high levels of late blight resistance and marketable vine maturity and tuber quality. In an effort to select and characterize apple rootstock resistant to apple proliferation (AP), progenies from seven apomictic rootstock selections and their parental apomictic species, Malus sieboldii and M. sargentii, were compared to standard stocks M 9 and M 11. Seedlings derived from open pollinated mother plants were grafted with cv. Golden Delicious and grown under natural infection conditions. The progenies differed greatly in resistance to the AP agent 'Candidatus Phytoplasma mali'. Progenies of M. sieboldii and its descendent rootstock selections D2212, 4608, 4551, and D1131 showed a high level of resistance, whereas progenies of M. sargentii and its descendent selections D1111 and C1828 proved susceptible. M 9 and M 11 showed an intermediate level of resistance. Phytoplasma titer in roots of the M. sieboldii and M. sargentii progeny groups was similarly low, whereas the concentration in the standard stocks was 100 to 5,000 times higher. In trees on most of the resistant stocks, only a minority was colonized in the scion, while in trees on susceptible and standard stocks, infection rate was often higher. Also, the titer in the top of trees on resistant stocks was usually lower than in trees on susceptible and standard stocks. Four progenies derived from open pollinated M. sieboldii and M. sieboldii descendents were subjected to DNA typing using simple sequence repeat (SSR) markers. This study revealed that the selected groups consisted mainly of mother-like plants (apomicts) and type I hybrids (unreduced mother genotype plus one male allele at each locus). Type II hybrids (full recombinants) and autopollinated offspring were rare. In the 4608 progeny, trees grown on type I hybrid rootstocks were significantly less affected than trees on mother-like stocks. In other progenies with fewer or no type I hybrids, trees on type II hybrids and autopollinated offspring suffered considerably more from disease than trees on mother-like stocks.  相似文献

Tuber dormancy and sprouting are important traits in commercial potato varieties. Since wild Solanum species can contribute new alleles in breeding, we combined Solanum chacoense and S. berthaultii species in a mapping population to develop a linkage map with genome wide markers and identify quantitative trait loci (QTL) for tuber dormancy and sprouting. There was genetic variation for the evaluated traits and high correlation (0.80) between dormancy and apical dominance release. QTL for tuber dormancy and sprouting traits were mapped to the same or similar positions on chromosomes 2, 3, 5, and 7, explaining from 9.5% to 16.3% of the phenotypic variance. We identified candidate genes associated with all major phytohormone regulation in close linkage with the QTL. The QTL on chromosomes 2, 3 and 7 are good candidates for marker assisted breeding, since they were associated with dormancy and apical dominance release and should be useful to delay or accelerate tuber sprouting.  相似文献

Greenhouse assays were carried out to characterizeSolanum accessions previously reported to be late blight resistant and to identify individuals within the accessions with high levels of resistance to late blight. Foliage of wildSolanum species accessions or hybrid cultivated potato x wild species (diploid or tetraploid) from Mexico, Russia, or South America was inoculated withPhytophthora infestans (US-8) and was scored for severity of foliar symptoms at 7 and 14 days after inoculation (DAI). Mexican accessions (S. brachycarpum,S. pinnatisectum,S. guerreroense, andS.fendleri) were significantly more resistant (1.1% infection at 14 DAI) than either Russian (59.1% infection at 14 DAI) or South American (53.4% infection at 14 DAI) accessions. Moreover, the genotypes within the Mexican accessions tended to be more uniformly resistant whereas genotypes within the Russian and South American accessions tended to segregate for resistance. The more resistant genotypes of Russian and South American accessions (176/546) were retested, and 56 genotypes were identified as having potential for use in potato breeding programs for resistance to late blight. Fifty percent of the selected genotypes were fromS. microdontum accessions PI498124, PI595510, and PI595511. The potential of these species for incorporation into breeding programs for late blight resistance is discussed.  相似文献

A screening system for apple proliferation resistance was developed, based on in vitro graft‐inoculation with the causal agent ‘Candidatus Phytoplasma mali’. For this, in vitro cultures of the field‐resistant apomictic genotypes Malus sieboldii, H0909, D2212 and the susceptible Malus × domestica genotypes Golden Delicious and rootstock M9 were established, as well as in vitro cultures of Rubinette and Golden Delicious infected with ‘Ca. P. mali’ strains PM4 and PM6, respectively. Healthy in vitro shoots were inoculated by micrografting with infected shoots used as graft tip. After 6 weeks graft contact no significant differences for graft quality were observed between healthy and infected grafts. Mortality of grafts and transmission rates were not significantly different among the different genotypes. The phytoplasma concentration in inoculated shoots was determined at different times post‐inoculation (p.i.) by quantitative real‐time PCR. Infected M. sieboldii and D2212 had lower phytoplasma concentration than the susceptible controls and showed no symptoms. H0909 showed an intermediate behaviour exhibiting lower phytoplasma concentrations with strain PM4 but growth was affected. The dynamics of phytoplasma concentration reached a maximum at 6–8 months p.i. for all genotypes but the values for 3–5 and 10–12 months p.i. were similar. The resistance of M. sieboldii and D2212 was confirmed in vitro. A significant difference in phytoplasma concentration was observed between strains PM4 and PM6.  相似文献

To obtain apple rootstocks resistant to apple proliferation and suitable to modern fruit growing, 24 cross-combinations were performed over a 5-year period using Malus sieboldii and its hybrids as donors of the resistance trait and standard apple rootstock Malus  ×  domestica genotypes as donors of agronomic value. Breeding with these genotypes was achieved despite different degrees of apomixis and polyploidy. Sets of five to six locus-specific microsatellite markers were identified for characterizing each progeny. Supported by flow cytometry these markers were applied to infer mode of reproduction, genomic constitution and ploidy level. Microsatellites allele composition identical to the maternal parent was revealed in 1668 of 3032 seedlings indicating seed formation through apomixis. Complete genetic recombination was found in 398 seedlings. The remaining hybrids displayed a higher ploidy than that of the parental plants which was consistent with the fertilization of unreduced egg cells. Thus, for each cross-combination, microsatellite loci were identified which enabled a reliable prediction of the ploidy level. They can now be applied in routine screening to distinguish sexual from apomictic progeny.  相似文献

Developing disease resistant cultivars is one of the major objectives for a potato (Solanum tuberosum L.) breeding program, but many resistant clones have not achieved commercial acceptance because of late maturity and non-marketable tuber characteristics. Selection for tuber quality should have greater emphasis inbreeding disease resistant cultivars. The objectives of this study were 1) to evaluate the ability of late blight (Phytophthora infestans (Mont.) de Bary) resistant parents to transmit chip-processing (tuber appearance, specificgravity, and chip-color) or tablestock (tuber appearance) quality to the offspring and 2) to compare selecting for tuber quality in single-hill versus eight-hill generations. We made crosses among eight unadapted potato cultivars (B0718-3,Bertita, Bzura, Greta, Libertas, Stobrawa, Tollocan, and Zarevo) with reported late blight resistance with adapted susceptible cultivars/breeding clones to generate 95populations (4,750 seedlings). Approximately 10% of the progeny from each cross were selected from single-hill plots based on tuber appearance, number, shape, and internal defects. These selected clones (408) were evaluated for tuber appearance, specific gravity, and chip-color. The same evaluations in the following year were made on tuber samples from eight-hill plots. Libertas and Tollocan were the best parents for transmitting chip-color; B0718-3, Zarevo, and Tollocan for transmitting tuber appearance; and Bzura, Libertas, and Zarevo for transmitting high specific gravity to the highest percentage of the offspring. Overall, 50% and 56% of the clones based on single- and eight-hill generation, respectively, were considered to possess chip-processing quality; over 90% of the clones had acceptable tablestock quality. A total of 71% of the clones possessing acceptable chip-processing and 95% of the clones possessing acceptable tablestock quality selected in both generations were identified in single-hill plots. The evaluation of tuber quality characteristics in single-hill generation not only permitted the identification of clones with acceptable chip-processing and tablestock, but also increased the amount of clonal information for the following generation of selection. In crosses between late blight resistant and susceptible clones, selection for tuber quality traits can be initiated in single-hill generation using a moderate selection intensity and precede late blight testing.
