Fusarium spp. cause severe damage in many agricultural crops, including sugar beet, with Fusarium oxysporum historically being considered as the most damaging of all species. Sugar beet needs to be protected from this class of soil-borne pathogens in order to ensure an optimal sugar yield in the field. Genetic control of the disease is crucial in managing these pathogens. Identification of single nucleotide polymorphism (SNP) markers linked to resistance can be a powerful tool for the introgression of valuable genes needed to develop Fusarium-resistant varieties. A candidate gene approach was carried out to identify SNP markers linked to putative Fusarium resistance sources in sugar beet. Five resistant analogue genes (RGAs) were screened by means of high resolution melting (HRM) analysis in a set of sugar beet lines, considered as resistant and susceptible to Fusarium oxysporum. HRM polymorphisms were observed in 80% of amplicons. Two HRM polymorphisms were significantly associated with Fusarium resistance (P < 0.05). The amplicons that showed association were sequenced and two SNPs were identified. The association was further validated on 96 susceptible and 96 resistant plants using competitive allele-specific PCR (KASPar) technology. The selected SNPs could be used for marker-assisted breeding of Fusarium resistance in sugar beet.  相似文献

Tillage erosion is increasingly recognised as an important soil erosion process on agricultural land. In view of its potential significance, there is a clear need to broaden the experimental database for the magnitude of tillage erosion to include a range of tillage implements and agricultural environments. The study discussed in this paper sought to address the need for such data by examining tillage erosion by a duckfoot chisel plough in stony soils on steep slopes in a semi-arid environment. Results of the investigation of coarse fraction (rock fragment) translocation by tillage in this environment have been presented elsewhere and the paper focuses on tillage translocation and erosion of the fine earth. Tillage translocation was measured at 10 sites, representing both upslope and downslope tillage by a duckfoot chisel plough on five different slopes, with tangents ranging from 0.02 to 0.41. A fine-earth tracer, comprising fine earth labelled with 134Cs, was introduced into the plough layer before tillage. After a single pass of the plough, incremental samples of plough soil were excavated and sieved to separate the fine earth from the rock fragments. Translocation of the fine-earth tracer was established by analysing the 134Cs content of the samples of fine earth. These data were used to establish translocation distances for each combination of slope and tillage direction. Translocation distances of the fine earth were not significantly different from translocation distances of the coarse fraction. For all sites, except uphill on the 0.41 slope, translocation distances were found to be linearly related to slope tangent. The soil flux due to tillage for each site was calculated using the translocation distance and the mass per unit area of the plough layer. For slopes with tangents <0.25, the relationship between soil flux and tangent was linear and the soil flux coefficient derived was 520–660 kg m−1 per pass. This is much larger than the coefficients found in other studies and this high magnitude is attributed to the non-cohesive nature and high rock fragment content of the soil in this investigation. A second contrast with previous studies was found in non-linearity in the relationship between soil flux and tangent when steeper slopes were included. This was a product of variation in plough depth between the steepest slopes and the remainder of the study area. On the basis of the study it is suggested that an improved understanding of tillage erosion may be obtained by considering the dual processes of tillage detachment (mass per unit area of soil subject to tillage) and tillage displacement (equivalent to translocation distance per pass) in assessing, comparing and modelling tillage translocation. An improved model is proposed that recognises the complexity of soil redistribution by tillage, provides a framework for process-based investigation of the controls on tillage fluxes, and allows identification of potential self-limiting conditions for tillage erosion.  相似文献

Rainfall variability within a storm can have a significant impact on the amount of chemical transported by surface runoff and preferential flow. Previously, studies have evaluated only a few slowly varying rainfall patterns and related sorption capacities. We use a bounded random cascade approach to generate 50 000 realizations of realistic rainfall intensity patterns within a single storm event (96 minutes duration, mean intensity of 18.75 mm hour?1) to explore the effects on the partitioning of rainfall and linearly sorbing solutes between fast preferential flow (loading) and slow flow in the soil matrix for a silt loam and a sandy clay. Loading and infiltration are modelled by a near‐surface mixing model and Green–Ampt infiltration. The statistical properties of loading were evaluated from these simulations. For this storm the mean total of resident solute mixing from the soil to preferential flow reached a maximum for a retardation factor R ~ 5. Much smaller loadings occurred for more weakly sorbing and more strongly sorbing solutes. The variability of loading tended to decrease with increasing R. Ensemble averaged rainfall patterns were derived which related to the magnitude of loading. The patterns of rainfall generating large preferential flows did not necessarily lead to large solute loading. Early peaking, mid‐storm peaking and late peaking rainfall contributed to large solute loadings, depending upon soil and chemical properties. These patterns result from a balance between the amount of preferential flow generated and the amount of solute available when preferential flow is triggered. The results suggest that the use of R as a measure of the mobility of resident solutes depends on the flow pathway considered. In addition, characterization of flux distributions in soil with weakly sorbing, resident tracers, may underestimate the potential for rapid transport of strongly sorbing solutes subject to natural variations in rainfall.  相似文献

Two trials using broiler chickens were undertaken to determine if variations in commercial slaughter techniques affected either the appearance or the residual blood content of the carcases produced. In the first trial groups of broilers were either stunned (55, 80 or 105 volts), electrocuted (240 volts) or anaesthetised and the subsequent bleed-out volumes after one, two, three and four minutes exsanguination were measured. The method of slaughter made no difference to the amount of blood lost after venesection. Haemoglobin content of the pectoral muscle was used to measure residual blood and was uniformly low in all groups (0.7 to 0.16 mg haemoglobin/g of muscle). In the second trial groups of chickens were either stunned and bled out, electrocuted, electrocuted and bled out, decapitated or halal killed before processing. The edible portions of the carcases were minced and the average haemoglobin content for each group was 0.19, 0.36, 0.22, 0.17 and 0.17 mg/g of soft tissue. The appearance of chickens that were stunned and bled out was the same as that of chickens that were electrocuted and bled out. The authors conclude that killing chickens by electrocution before exsanguination does not adversely affect the carcase appearance. Furthermore, electrocution ensures that broiler chickens are humanely slaughtered.  相似文献

In a long‐term study of the effects on soil fertility and microbial activity of heavy metals contained in sewage sludges, metal‐amended liquid sludges each with elevated Zn, Cu or Cd concentrations were applied over a 3‐year period (1995–1997) to three sites in England. The experiments were sited adjacent to experimental plots receiving metal‐rich sludge cakes enabling comparisons to be made between the effects of heavy metal additions in metal‐amended liquid sludges and sludge cakes. The liquid sludge additions were regarded as ‘worst case’ treatments in terms of likely metal availability, akin to a long‐term situation following sewage sludge additions where organic matter levels had declined and stabilised. The aim was to establish individual Zn (50–425 mg kg?1), Cu (15–195 mg kg?1) and Cd (0.3–4.0 mg kg?1) metal dose–response treatments at each site, but with significantly smaller levels of organic matter addition than the corresponding sludge cake experiments. There were no differences (P > 0.05) in soil respiration rates, biomass carbon concentrations or most probable numbers of clover Rhizobium between the treatments at any of the sites at the end of the liquid sludge application programme. Soil heavy metal extractability differed between the metal‐amended liquid sludge and metal‐rich sludge cake treatments; Zn and Cd extractabilities were higher from the liquid sludge additions, whereas Cu extractability was higher from the sludge cake application. These differences in metal extractability in the treated soil samples reflected the contrasting NH4NO3 extractable metal contents of the metal‐amended liquid sludges and sludge cakes that were originally applied.  相似文献

Bacterial populations in constructed and fumigated golf course putting greens is a topic that has not been widely explored. The objective of this project was to evaluate the root mass and rhizosphere bacterial populations of creeping bentgrass (Agrostris palustris Huds. Crenshaw) greens over time as affected by root-zone mix and nitrogen rate. Individual miniature putting greens (1x0.5 m) were built at ground level. Two root-zone mixes (80%/20% sand-peat and 100% sand) and two N rates (1.0 and 0.5 g N m–2 week–1 all year) were evaluated, with four replications of each root-zone x N rate treatment. Quarterly (February, May, August, November) root samples (10 cm deep) were removed from each green for 3 years. Standard dilution plating techniques were used to enumerate fluorescent pseudomonads, gram-positive bacteria, gram-negative bacteria, Stenotrophomonas maltophilia-like bacteria, actinomycetes, heat-tolerant bacteria, and total aerobic bacteria. There was rarely a significant N rate x root-zone mix interaction, although main effects of N rate and root-zone mix did affect microbial populations at some samplings. If differences did exist, bacterial populations were higher in the sand-peat root-zone mix or in the treatments receiving the higher N rate. While a significant decrease in all bacterial populations enumerated was observed between November 1998 and February 1999, no similar decrease was observed for the same time period in 1997–1998 or 1999–2000. In general, there were no obvious population trends in any microbial population over the 3-year sampling period. Populations of total bacteria ranged from a low of 5.4 to a high of 8.3 log10 CFU per gram of root and rhizosphere soil throughout the 3-year sampling period.  相似文献

The Mars Exploration Rover Spirit has traversed a fairly flat, rock-strewn terrain whose surface is shaped primarily by impact events, although some of the landscape has been altered by eolian processes. Impacts ejected basaltic rocks that probably were part of locally formed lava flows from at least 10 meters depth. Some rocks have been textured and/or partially buried by windblown sediments less than 2 millimeters in diameter that concentrate within shallow, partially filled, circular impact depressions referred to as hollows. The terrain traversed during the 90-sol (martian solar day) nominal mission shows no evidence for an ancient lake in Gusev crater.  相似文献
