Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease (JD) in ruminants. MAP is known to enter a dormant phase outside the host, typically on soil. In vitro experiments have reported regulation of certain MAP genes when exposed to stressors similar to what is thought to produce dormancy. It is believed that in vivo regulation of dormancy genes and associated proteins by MAP may play a role in evading the host defence mechanisms and induce the host immune response against these dormancy-related proteins. Five proteins encoded by dormancy-related genes that were previously found to be upregulated under stress conditions and predicted through in silico analysis to possess immune epitopes (three hypothetical proteins and two proteins involved in fatty acid metabolism) were selected. Recombinant proteins were produced, purified and evaluated by indirect enzyme-linked immunosorbent assay (ELISA) for immunogenicity using a panel of sera obtained from sheep unexposed and exposed to MAP. The antibody levels of the exposed group were significantly higher than the unexposed group (P<0.001). Individually, the five proteins were found to discriminate between sera from sheep exposed to MAP compared to unexposed sheep. At 91% diagnostic specificity, the diagnostic sensitivity of the recombinant antigen ELISA ranged from 24% to 42% and AUC(ROC) from 0.7015 to 0.8405.  相似文献

Background:Hyperuricemia induces nephropathy through the mediation of oxidative stress, tubular injury, inflammation, and fibrosis. The high uric acid level is associated with the reduction of vitamin D levels. However, the reno-protective effects of this vitamin in hyperuricemia condition remain unknown. This study aimed to elucidate calcitriol treatment in a uric acid-induced hyperuricemia mice model. Methods:Uric acid (125 mg/kg BW) was administered intraperitoneally for 7 (UA7) and 14 (UA14) days. Calcitriol (0.5 g/kg BW) was intraperitoneally injected for the following seven days, after 14 days of uric acid induction (UA14VD7 group). The control group received NaCl 0.9%, by the same route. Serum creatinine was measured using calorimetric method, and uric acid levels were assessed using enzymatic calorimetric assay. Tubular injury and fibrosis were assessed using PAS and Sirius red staining. RT-PCR and qRT-PCR were carried out for the analyses of SOD-1, Collagen-1, and TGF-1 mRNA expression in the kidney. Immunostaining of SOD-1 was performed to detect its expression in the kidney. Results:Uric acid and creatinine levels markedly increased in UA14 groups, followed by an exacerbation of tubular injury. RT-PCR revealed the upregulation of Collagen-1 and TGF-1, along with the downregulation of SOD-1. Calcitriol treatment attenuated the injury with reducing uric acid and creatinine levels, as well as tubular injury. This was associated with lower Collagen-1 and TGF-1 mRNA expression compared to the UA7 and UA14 groups. SOD-1 was upregulated in epithelial cells in the UA14VD7 group. Conclusion:Calcitriol treatment after uric acid induction may attenuate kidney injury through upregulation of SOD-1 and downregulation of Collagen-1 and TGF-1 gene expression. Among the Dayak people in East Kalimantan, simpukng (“forest gardens”) are an important component of their traditional farming systems. Simpukng is managed secondary forests in which selected species of fruits, rattan, bamboo, timber and other plants are planted. While most are owned by families and passed down from one generation to the next, some are managed on a communal basis. Complex customary Dayak rules exist that control the use and inheritance of these forests that help to avoid over-exploitation of resources. There is clear gender division of labour among Dayak in the management of simpukng that provide a range of products – for household consumption and sale and for customary rituals – fruits, vegetables, medicines, fire wood, honey, rattan, bamboos, and timber. Local knowledge about the more highly valued species are discussed. These indigenous forest garden systems are currently under threat from large-scale mining and logging activities; conflicts between local and external agencies are unfortunately frequent. This paper examines the development and management of simpukng in four Dayak villages in East Kalimantan and their implications on sustainable management of natural resources, with particular emphasis on the role of local knowledge of some of the more highly valued species and the current challenges faced by these communities in maintaining their traditional agroforest management practices.  相似文献

Chawali and Lekali are two common farmer’s barley varieties or landraces in Jumla, Nepal (2,240–3,000 m) with small to bold grains and wide adaptation from irrigated low lands to high hills. This study was undertaken to test whether features of the traditional seed system can significantly influence the diversity of a crop and its conservation on-farm. In Jumla (high-hill), the barley seed system is completely informal and is mainly from farmer to farmer. In the present study, the seed flows and the pattern of genetic diversity in barley were investigated to detect differences between the two varieties and test the divergence among populations of each variety These data suggested that Chawali, the more common variety, was less subject to homogenising gene flow between farms than was Lekali. A total of 128 farming households were surveyed for seed supply information and 128 populations for each landrace from two villages: Kartikswami and Talium were collected for SSR diversity analysis. Some 92 SSRs were screened in an initial sample of 20 barley populations of both landraces and 2 improved varieties (LG-51 and Soluwa). Of the 81 SSRs that consistently amplified, only 15 SSRs (19%) were polymorphic with gene diversity values ranging from 0.09 to 0.71. A medium to low diversity was detected among the landrace populations of barley varieties. Chawali populations were less polymorphic within ecological groups, and more divergent between than were Lekail populations. This result accords with Chawali having a more conservative local seed system.  相似文献

Jhinuwa is an aromatic rice (Oryza sativa L.) landrace from the Pokhara Valley of Nepal. A total of 210 accessions comprising seven types of Jhinuwa rice landraces were randomly collected from the rice fields to evaluate inter- and intra-population variability based on agro-morphological traits. The experiment was conducted in factorial randomized complete block design with three replicates in 2005. The first six principal components (PCs) accounted for 76.6 % variation for agro-morphological traits. Major traits that accounted for the variation by six PCs includes days to heading, days to maturity, total grain panicle?1, fertile grain panicle?1, culm length, panicle length, milling recovery, head rice recovery, aroma, 1,000 grain weight, sterile grain panicle?1, grain sterility %, and leaf characteristics. Both principal coordinate analysis and cluster analyses revealed four phenotypic groups, two of which represent Bayarni, Jhinuwa, and Biramphul while the other two account for Tunde and Pakho Tunde. Tunde, Pakho Tunde, Kalo Bayarni, and Seto Bayarni showed higher intra- as well as inter-population variation compared to other populations. The phenotypic and genotypic coefficients of variation, broad sense heritability (h 2B) and genetic advance (GA) as a percent of the mean assessed for 210 accessions revealed high h 2B and GA estimates for leaf width, leaf length breadth ratio, ligule length, sterile grain panicle?1, grain sterility % and 1,000 grain weight. The current study demonstrates that improvement in Jhinuwa rice landrace is possible by selecting superior accessions from existing natural populations while selection should be focussed to market traits with higher h 2B and GA estimates.  相似文献

Protein quality, as determined by the PDCAAS method, is a measure of a protein's ability to provide adequate levels of essential amino acids for human needs. PDCAAS is calculated using an amino acid profile and true digestibility of a food protein. Soy protein is recognized as a high quality plant protein, but published PDCAAS values may vary based on the soy protein ingredient as well as the reproducibility and accuracy of the testing methods. Comparison of PDCAAS values for four differently processed soy ingredients, including three isolated soy proteins (ISP) and one soy protein concentrate (SPC), was made using two different laboratories with evaluation of the impact of the reproducibility and accuracy of amino acid profiles. PDCAAS calculations, using amino acid values from one laboratory, yielded a truncated PDCAAS of 1.00 for all four ingredients, while a second laboratory provided statistically significantly lower scores (0.95-1.00). We conclude that analytical method error can be a significant contributor to PDCAAS differences and can be mitigated by the application of amino acid nitrogen recovery correction factors.  相似文献

Continuous use of herbicides has resulted in the evolution of resistance to all major herbicide modes of action worldwide. Besides the well‐documented cases of newly acquired resistance through genetic changes, epigenetic regulation may also contribute to herbicide resistance in weeds. Epigenetics involves processes that modify the expression of specific genetic elements without changes in the DNA sequence, and play an important role in re‐programming gene expression. Epigenetic modifications can be induced spontaneously, genetically or environmentally. Stress‐induced epigenetic changes are normally reverted soon after stress exposure, although in specific cases they can also be carried over multiple generations, thereby having a selective benefit. Here, we provide an overview of the basis of epigenetic regulation in plants and discuss the possible effect of epigenetic changes on herbicide resistance. The understanding of these epigenetic changes would add a new perspective to our knowledge of environmental and management stresses and their effects on the evolution of herbicide resistance in weeds. © 2017 Society of Chemical Industry  相似文献

A questionnaire survey of 408 households explored the role of socio-economic and cultural factors in rice (Oryza sativa L.) varietal diversity management on-farm in two contrasting eco-sites in Nepal. Multiple regression outputs suggest that number of parcels of land, livestock number, number of rice ecosystems, agro-ecology (altitude), and use of chemical fertilizer have a significant positive influence on landrace diversity on-farm, while membership in farmers’ groups linked to extension services has significant but negative influence on landrace diversity. Factors with significant positive influence on diversity of modern varieties on-farm were number of parcels of land and of rice ecosystems, access to irrigation, membership in farmers’ groups, and use of insecticide. Within communities, resource-endowed households maintain significantly higher varietal diversity on-farm than resource-poor households and play a significant role in conserving landraces that are vulnerable to genetic erosion and those with socio-cultural and market-preferred traits. Resource-poor households also contribute to local diversity conservation but at lower richness and area coverage levels than resource-endowed households. Households where a female had assumed the role of head of household due to death or migrant work of her husband had less diversity due to lower labor availability. Landraces with socio-cultural and market-preferred traits are few in number but have potential to be conserved on-farm.

A genomic DNA fragment (pAA7-2) amplified by a random amplified polymorphic DNA primer from the rice cultivar IR68 was used to assess the genetic variation and genetic relationships among the species of genus Oryza. A Southern hybridization experiment of diverse upland rice accessions using a single restriction enzyme HindIII generated unique DNA fingerprint for each accession. The differential hybridization pattern reflecting the copy number variation of pAA7-2 in a collection of wild species and cultivated species of rice provided insight about the genetic relationships among them. All AA genome species exhibited clear banding pattern suggesting presence of fewer copies of this sequence. Strongest hybridization signal was obtained in species belonging to BB, CC, GG, BBCC, CCDD genomes, whereas weakest hybridization signal was visible in EE, FF, and HHJJ genome species. Oryza brachyantha was the most divergent species. Clear difference in banding pattern was evident between Oryza schlechteri and Oryza coarctata belonging to HHKK genome. Although pAA7-2 had no repetitive sequences often associated with hypervariable loci, homology to a putative unclassified expressed retrotransposon distributed over several rice chromosomes was responsible for the complex banding patterns. There were more sites homologous to pAA7-2 sequence in corn and sorghum genome compared with the rice genome. The study demonstrates the potential of pAA7-2 as a powerful molecular tool for DNA fingerprinting, genetic diversity, phylogenetic, and evolutionary studies in Oryza sativa and its wild relatives and other grasses.  相似文献
