The present paper reports the prevalence of coccidiosis in industrial broiler chickens in Faisalabad, Punjab, Pakistan to determine the occurrence of different species of Eimeria in the area and to assess their correlation with the environmental conditions including temperature, humidity and rainfall. The study was conducted from January 2009 to December 2010. Examination of chicken guts revealed 43.89% (n = 3,283/7,480) prevalence of coccidiosis. The highest prevalence (27.04%) was recorded of Eimeria tenella followed by Eimeria maxima (22.42%), Eimeria acervulina (19.89%) and Eimeria necatrix (4.02%). The prevalence of disease was significantly higher (P < 0.05) in autumn (60.02 ± 4.38) followed by summer (47.42 ± 2.92), spring (36.92 ± 2.27) and winter (29.89 ± 3.43). Likewise, prevalence of different species of Eimeria also varied (P < 0.05) in different seasons of the year except that of E. necatrix. A strong correlation (P < 0.05) of environmental conditions with the overall and species-wise prevalence of coccidiosis was recorded except E. necatrix.  相似文献

Bluetongue virus (BTV), a member of Orbivirus genus (family Reoviridae), is a non-contagious infection of domestic and wild ruminants. The current study was designed to detect various serotypes of BTV in small ruminants of Khyber Pakhtunkhwa (KPK) province of Pakistan, along with their effects on hemato-biochemical parameters. A total of 408 serum samples in four districts (Mansehra, Abbottabad, Swabi, and Kohat) of KPK from small ruminants were screened based on competitive ELISA (cELISA). A total of 204 (50%) samples were found positive for BTV group–specific antibodies. The seropositive samples were processed for the detection of BTV serotypes through real-time polymerase chain reaction (qPCR). Out of 204 cELISA-positive samples, 60 (29.41%) were found positive through qPCR. Three serotypes [6, 8, 9] were detected from Mansehra District and two from Kohat [2, 8] and Abbottabad [6, 8], while only one from Swabi [8]. The serotype “8” was found consistently in all the four study districts. A significant (p?<?0.05) increase in the level of blood urea nitrogen (BUN) and alkaline phosphatase (ALP) was recorded in goats, whereas aspartate aminotransferase (AST) in sheep infected with BTV, compared to healthy animals. The hematological parameters showed significantly (p?<?0.05) raised total leucocyte count (TLC) in both sheep and goats, whereas only hematocrit (HCT) value was increased significantly (p?<?0.05) in infected sheep. This is the first report on serotyping of BTV among small ruminants in Pakistan.

Heat stress at reproductive stages of cotton crop is the main cause for instability in yield than other abiotic stresses of crops. An experiment was conducted to observe the impacts of heat stress at squaring and flowering stages of cotton and potential of exogenously applied ascorbic acid to alleviate this stress. The experiment was planted for two consecutive years, that is 2013 and 2014, in randomized complete block design with split plot arrangement having three replications. Treatments were comprised of heat stress in main plots, viz. H0 = no heat imposition; H1 = heat imposition at square initiation; H2 = heat imposition at flower initiation and foliar‐applied ascorbic acid in subplots, viz. A0 = control or water spray; A1 = 20 mg/L; A2 = 40 mg/L; and A3 = 60 mg/L. Results showed that heat stress badly affected plant water relations (relative water contents and osmotic potential), total chlorophyll contents and activities of antioxidant enzymes that led to a corresponding decrease in seed cotton yield. Foliar application of ascorbic acid (40–60 mg/L) ameliorated adverse effects of heat and thus helped the crop to improve its water relations, stay green and antioxidant enzymes resulting into better crop yield.  相似文献

The objective of this study was to identify allelic variations at Glu-1 loci of wheat (Triticum aestivum L.) advanced lines derived from hybridization of bread wheat and synthetic hexaploid wheats (2n = 6x = 42; AABBDD). Locally adapted wheat genotypes were crossed with synthetic hexaploid wheats. From the 134 different cross combinations made, 202 F8 advanced lines were selected and their HMW-GS composition was studied using SDS-PAGE. In total, 24 allelic variants and 68 HMW-GS combinations were observed at Glu-A1, Glu-B1, and Glu-D1 loci. In bread wheat, the Glu-D1 locus is usually characterized by subunits 1Dx2+1Dy12 and 1Dx5+1Dy10 with the latter having a stronger effect on bread-making quality. The subunit 1Dx5+1Dy10 was predominantly observed in these advanced lines. The inferior subunit 1Dx2+1Dy12, predominant in adapted wheat germplasm showed a comparative low frequency in the derived advanced breeding lines. Its successful replacement is due to the other better allelic variants at the Glu-D1 locus inherited in these synthetic hexaploid wheats from Aegilops tauschii (2n = 2x = 14; DD).  相似文献

Heat stress at reproductive stages of cotton is a leading encumbrance to accomplish yield potential. An experiment was conducted to alleviate adversities of terminal heat stress on cotton. Experiment was conducted at University of Agriculture Faisalabad, Pakistan during 2013 and repeated in 2014 using Randomized Complete Block Design (RCBD) with split arrangement. Treatments were comprised of heat stress in main plots, viz., H0 = no heat imposition, H1 = heat imposition at squaring, and H2 = heat imposition at flowering. Selenium was foliar applied in split plot, viz., Se0 = 0, Se50 = 50, Se100 = 100, and Se150 = 150 mg L?1. Cell membrane thermostability, soluble proteins, and total phenolics were reduced significantly under heat stress leading to reduced cotton yield and related components. Maximum membrane thermostability, total phenolics, soluble proteins, opened bolls, boll weight, sympodial branches, and seed cotton yield were recorded with the application 150 mg Se L?1.  相似文献

Mushrooms (Lentinus edodes) were processed for hot water (HWE), methanolic (ME), and polysaccharide (PSE) extracts. Polysaccharides were isolated through ion exchange (DEAE cellulose) and size exclusion (Sephadex G-100) chromatography. Monosaccharides including maltose (0.282%), glucose (0.113%), and mannose (0.451%) were identified, qualitatively and quantitatively, from the isolated polysaccharides through high-performance liquid chromatography. The whole study was divided into two experiments. Experiment 1 was meant for the evaluation of HWE and ME; whereas, experiment 2 was meant for the evaluation of PSE for immunostimulatory and immunotherapeutic activities. The cellular and humoral immune responses were demonstrated through lymphoproliferative response to phytohemagglutinin-P (PHA-P) and anti-body response to sheep red blood cells (SRBCs), respectively. The immunotherapeutic effects of these extracts were demonstrated against eimeriasis in terms of lesion scoring, oocysts per gram of droppings, and percent protection. Cell-mediated immune responses observed at 24, 48, and 72 h post-PHA-P injection were significantly higher (P < 0.05) in chickens administered with any of the three extracts (PSE, ME, and HWE), when compared with the controls. Humoral immune response in terms of anti-SRBCs anti-body titers was also observed higher in chickens administered with mushroom extracts. In the challenge experiment, significantly higher (P < 0.05) OPG and lesion scores were observed in controls as compared to the groups administered with mushroom extracts (HWE, ME, and PSE). Significantly higher (P < 0.05) percent protection against eimeriasis was observed in all groups administered with different extracts of L. edodes as compared to controls. In conclusion, L. edodes extracts showed immunostimulatory potential which persisted against eimeriasis in chicken.  相似文献

High molecular weight glutenin subunit composition and variation in 95 Elite-1 synthetic hexaploid (SH) wheats (Triticum turgidum/Aegilops tauschii; 2n = 6× = 42; AABBDD) were determined by sodium dodecyl sulphate polyacrylamide gel electrophoresis method (SDS-PAGE). Twenty two different alleles at Glu-1 loci in SHs were observed. Forty four different patterns of HMW-GS in synthetics were found. This higher HMW glutenin composition was due to higher proportion of D-genome encoded subunits in these SHs. 8% urea/SDS-PAGE better discriminated subunit 2* than 12% gels. However 12% urea/SDS-PAGE allowed differentiated mobility of Glu-Dt1 subunits. Genetic variability at Glu-Dt1 locus was greater than Glu-A1 and Glu-B1 loci. The relative high frequency of superior alleles, Glu-B1b and Glu-Dt1d indicated the superior bread making quality attributes embedded in these synthetic hexaploid wheats. Of the 95 Elite-1 SHs 27.1% possessed superior alleles at Glu-A1 and 51% had superior alleles at Glu-B1 locus. At Glu-Dt1 frequency of inferior allele 1Dx2 + 1Dy12 was very low (5.26%) and nine different rare alleles along with the higher frequency (22.1%) of D-genome encoded subunit, 1Dx5 + 1Dy10, were observed. These superior alleles shall form the priority selective sieve for their usage in wheat improvement efforts.  相似文献
