Top dieback of Norway spruce (Picea abies), triggered by drought in 2004–2006, has been observed in Southeast Norway and trees died within four years after appearance of the first symptoms. The aim of our study was to use sap flux measurements as a diagnostic method for assessment of tree vitality. We used the heat field deformation method to monitor the sap flux density (SFD) in four pairs of healthy and declining trees in situ. To provide retrospective information on hydraulic performance of the trees we took samples for wood anatomical analysis. After felling the trees we used the modified differential translucence method (MDT) as a proxy for the SFD measurements. Healthy trees had three times higher SFD values as declining trees. In some healthy trees we detected decreasing SFD with time. The MDT method agreed with the SFD measurements. In conclusion, we detected sap flux dysfunction in declining trees and showed that the SFD reduction may occur during a short period, prior to occurrence of any visual symptoms. We suggest incorporating the SFD measurements into the repertoire of diagnostic tools in forest pathology.  相似文献

Birch (Betula pendula Roth) growth within the Western Siberia forest-steppe was analyzed based on long-term (1897–2006) inventory data (height, diameter at breast height [dbh], and stand volume). Analysis of biometry parameters showed increased growth at the beginning of twenty-first century compared to similar stands (stands age = 40–60 years) at the end of nineteenth century. Mean height, dbh, and stem volume increased from 14 to 20 m, from 16 to 22 cm, and from ~63 to ~220 m3/ha, respectively. Significant correlations were found between the stands mean height, dbh, and volume on the one hand, and vegetation period length (rs = 0.71 to 0.74), atmospheric CO2 concentration (rs = 0.71 to 0.76), and drought index (Standardized Precipitation-Evapotranspiration Index, rs = ?0.33 to ?0.51) on the other hand. The results obtained have revealed apparent climate-induced impacts (e.g. increase of vegetation period length and birch habitat drying due to drought increase) on the stands growth. Along with this, a high correlation of birch biometric parameters and [CO2] in ambient air indicated an effect of CO2 fertilization. Meanwhile, further drought increase may switch birch stand growth into decline and greater mortality as has already been observed within the Trans-Baikal forest-steppe ecotone.  相似文献

Chitosan, the product of chitin deacetylation, is an excellent candidate for enzyme immobilization purposes. Here we demonstrate that papain, an endolytic cysteine protease (EC: 3.4.22.2) from Carica papaya latex immobilized on the matrixes of medium molecular (200 kDa) and high molecular (350 kDa) weight chitosans exhibits anti-biofilm activity and increases the antimicrobials efficiency against biofilm-embedded bacteria. Immobilization in glycine buffer (pH 9.0) allowed adsorption up to 30% of the total protein (mg g chitosan−1) and specific activity (U mg protein−1), leading to the preservation of more than 90% of the initial total activity (U mL−1). While optimal pH and temperature of the immobilized papain did not change, the immobilized enzyme exhibited elevated thermal stability and 6–7-fold longer half-life time in comparison with the soluble papain. While one-half of the total enzyme dissociates from both carriers in 24 h, this property could be used for wound-dressing materials design with dosed release of the enzyme to overcome the relatively high cytotoxicity of soluble papain. Our results indicate that both soluble and immobilized papain efficiently destroy biofilms formed by Staphylococcus aureus and Staphylococcus epidermidis. As a consequence, papain, both soluble and immobilized on medium molecular weight chitosan, is capable of potentiating the efficacy of antimicrobials against biofilm-embedded Staphylococci. Thus, papain immobilized on medium molecular weight chitosan appears a presumably beneficial agent for outer wound treatment for biofilms destruction, increasing antimicrobial treatment effectiveness.  相似文献

Roots under the load of heavy machinery in spruce trees   总被引:2,自引:0,他引:2  
The effects of the passage of forwarders on soil and damage to spruce root systems along an experimental trail were studied. The site was characterized by medium-textured soil of the pseudogley type under favorable moisture conditions. Due to the passages, the soil was compacted down to a depth of 20 cm, soil porosity was decreased by 5% (volume) and soil aeration was decreased by more than 5%. Substantially higher values of mechanical soil resistance occurred (estimated by penetrometric measurements) in a soil pit situated in a rut after passages. Pressure measured by sensors placed at a depth of 10 cm below the soil surface reached values ranging from 0.09 to 0.11 MPa in plots uncovered with slash and 0.03–0.07 MPa in plots covered with slash after two to four passages, and 0.06–0.07 after six to ten passages. Soil surface deformations occurred in the upper soil layers through tire impression. This resulted in the origin of ruts, whose depth and width was dependent on the type of tires, their load, surface conditions, type and texture of soil, soil moisture and number of passages. Pressure in the soil layers imposed by the tire of a given type, inflation and load changed in relation to depth, ground cover, soil properties and reinforcing components on the soil surface. Sap flow in coarse roots actually treated by a moving heavy load clearly and immediately responded with a sharp increase followed by a similar decrease (peak flow) after several minutes. On average, the flow rate decreased by about 8% after the first treatment compared to the untreated state, and remained the same after passing the peak during the second pass when the maximum load was applied. However, this decrease amounted to about 40%, when compared to the “relative zero flow” after root severing. This indicates serious local damage to the conducting system. Even when loading directly damaged rather small fractions of the total root systems, it opened tree tissues to subsequent fungal infection, whose impact could be very serious in future years. Flow in stem sections oriented in the opposite direction to the trail and the flow in stem sections and root buttresses oriented toward the trail (but where roots were not actually growing below the trail or grew deeper in the soil), neither responded to the treatment or responded insignificantly. Sap flow responded only in surface roots below trails, occurring down to a depth of about 10 cm below the original soil (litter) surface. This occurred only when a significant part of the roots (with the total projected area of treated root branches more then 500 cm2) were under the tires. This indicates the protective ability of soils and also, a possible method of artificial root protection.  相似文献

Hydraulic redistribution (HR) by roots of large Norway spruce (Picea abies (L.) Karst.) trees was investigated by means of sap flow measurements made with the heat field deformation method. Irrigation was applied to a limited portion of the root system to steepen gradients of water potential in the soil and thus enhance rates of HR. On completion of the sap flow measurements, and to aid in their interpretation, the structure of the root system of seven of the investigated trees was exposed to a depth of 30 cm with a supersonic air-stream (air-spade). Before irrigation, vertical redistribution of water was observed in large coarse roots and some adjacent small lateral roots. Immediately after localized irrigation, horizontal redistribution of water from watered roots to dry roots via the stem base was demonstrated. The amount of horizontal distribution depended on the position of the receiving roots relative to the watered roots and the absorbing area of the watered root. No redistribution from watered roots via dry soil to roots of neighboring trees was detected. Responses of sap flow to localized irrigation were more pronounced in small lateral roots than in large branching roots where release and uptake of water are integrated. Sap flow measurements with multi-point sensors along radii in large lateral roots demonstrated water extraction from different soil horizons. We conclude that synchronous measurements of sap flow in both small and large lateral roots are needed to study water absorption and transport in tree root systems.  相似文献

Knowledge of sap flow variability in tree trunks is important for up-scaling transpiration from the measuring point to the whole-tree and stand levels. Natural variability in sap flow, both radial and circumferential, was studied in the trunks and branches of mature olive trees (Olea europea L., cv Coratina) by the heat field deformation method using multi-point sensors. Sapwood depth ranged from 22 to 55 mm with greater variability in trunks than in branches. Two asymmetric types of sap flow radial patterns were observed: Type 1, rising to a maximum near the mid-point of the sapwood; and Type 2, falling continuously from a maximum just below cambium to zero at the inner boundary of the sapwood. The Type 1 pattern was recorded more often in branches and smaller trees. Both types of sap flow radial patterns were observed in trunks of the sample trees. Sap flow radial patterns were rather stable during the day, but varied with soil water changes. A decrease in sap flow in the outermost xylem was related to water depletion in the topsoil. We hypothesized that the variations in sap flow radial pattern in a tree trunk reflects a vertical distribution of water uptake that varies with water availability in different soil layers.  相似文献
