Alteration of somatostatin receptor 2 expression in canine mammary gland tumor

Somatostatin receptor 2 (SSTR2) is a negative regulator of cell proliferation in human breast cancer. Since there is little information about SSTR2 in canine mammary gland tumor (MGT), we clarified its distribution and expression level in normal mammary gland, benign MGT and malignant MGT. SSTR2 expression determined by immunohistochemical staining was observed in the cytoplasm of luminal epithelial cells. The intensity was negatively correlated with malignancy: normal tissues and some of the benign tumors had the highest levels, while the malignant tumors had little or no SSTR2 expression. As for the Western blotting, SSTR2 protein level in benign tumors was significantly lower than the normal mammary gland. On the other hand, SSTR2 protein levels in two of three malignant tumors were higher than the other groups. These results suggest that SSTR2 expression alters according to the malignancy of canine MGT.     

Influence of acute exposure to a low dose of systemic insecticide fipronil on locomotor activity and emotional behavior in adult male mice

Fipronil (FPN) is a systemic insecticide that antagonizes the gamma-aminobutyric acid type A (GABA_A) receptors in insects. Recently, adverse effects of FPN on mammals have been reported, but most of those were caused by high doses of FPN and additives in the products. We investigated the effects of low-dose pure FPN on the emotional behavior of mice. Nine-week-old male mice conducted behavioral tests 24 hr after FPN administration by gavage at doses of 0.05 or 5 mg/kg based on the no-observed-effect level (NOEL), showed a significant increase in locomotor activity and dose-dependent responses on the time they spent in the central zone in the open field test. Pure FPN below the NOEL dose may affect the emotional behavior of mice.     

Immunohistochemical study on the delayed progression of epithelial apoptosis in follicle-associated epithelium of rat Peyer's patch

It is well known that some caspases in apoptosis is involved in determinant of terminal differentiation and maturation of various cells. Our previous study ultrastructurally clarified the differentiation into M cells from immature microvillous epithelial cells and the redifferentiation from M cells to microvillous epithelial cells in the follicle-associated epithelium (FAE) of rat Peyer's patch. In this study, the difference of epithelial apoptosis between the FAE of Peyer's patch and intestinal villi was immunohistochemically investigated in rat jejunoileum. As a result, cleaved caspase-3 was limited to several epithelial cells at the tip of FAE, whereas almost all of the epithelial cells were cleaved caspase-3 positive in intestinal villi. Cleaved caspase-9 was detected only in a few exfoliating or exfoliated epithelial cells of both FAE and intestinal villi. Nuclear DNA-fragmentation was detected only in several epithelial cells of the tip of FAE, while it was expressed from the middle regions in the intestinal villi. The DNase I expression of the epithelial cytoplasm was much weaker in FAE than in intestinal villi. Bcl-x expression was restricted in the apical cytoplasms of epithelial cells in the FAE, whereas it was restricted in whole cytoplasms in villous epithelial cells. These findings suggest that the progression of the apoptotic process in the epithelial cells of FAE is later than in the intestinal villi, so that the possibility of epithelial differentiation might be remained in the FAE, unlike in the intestinal villi.     

Evaluation of coral bleaching condition <Emphasis Type="Italic">in situ</Emphasis> using an underwater pulse amplitude modulated fluorometer

ABSTRACT:   Photosynthetic activities of hermatypic corals (28 genera, c . 68 species and c. 92 specimen) were measured in November 2000 at Sekisei lagoon, Okinawa, Japan, using an underwater pulse amplitude modulated fluorometer. Maximum quantum yield (Fv/Fm) ranged from 0.61 to 0.70 and had an average value of 0.664 (standard deviation [SD], 0.0289). Following the measurement of Fv/Fm, quantum yield (Fm'-F)/Fm' at eight-leveled actinic-light intensities were obtained from all specimens. Values of electronic transport rates (ETR) were nearly the same under low photosynthetic active radiation (PAR; µmol quanta/m² per s) condition and varied with increasing PAR. The value of ETR at 100 µmol quanta/m² per s ranged from 21.3 to 30.0 and had an average of 27.8 (SD, 2.09). In August 2001, a coral bleaching event was observed in Sekisei Lagoon. During the event, photosynthetic activities of 21 specimens of eight species (seven genera) were measured. By applying two indicators, Fv/Fm and ETR at 100 µmol quanta/m² per s obtained in August 2000, 18 of 21 specimens were suggested under the bleaching process. In case of Porites lutea , seven colonies were visually evaluated for their condition of bleaching and were found to have two 'normal', three 'slightly bleached' and two 'heavily bleached'. From the density of zooxanthellae, colonies evaluated 'normal' or 'slightly bleached' did not show an apparent difference. However, they were clearly identified by Fm/Fv and ETR at 100 µmol quanta/m² per s.     

Levels of active oxygen species are controlled by ascorbic acid and anthocyanin in Arabidopsis

Stabilization of the levels of active oxygen species (AOS) is important to the survival of organisms. To clarify the system controlling levels of AOS in plants, this study used an electron spin resonance (ESR) method to directly measure superoxide radical (O(2)(.-)) scavenging activities in the wild-type Arabidopsis thaliana (Col and Ler ecotypes), two anthocyanin mutants (tt3 and ttg1), and an ascorbic acid mutant (vtc1). Under ordinary growth conditions, Arabidopsis contained superoxide-scavenging activity (SOSA) of approximately 300-500 SOD units/g of fresh weight. The ESR pattern indicated that most (40-50%) of this activity was due to ascorbic acid. For the analysis of SOSA under conditions of oxidative stress, synthesis of AOS was induced by gamma-irradiation. The radical scavenging activity in irradiated plants increased approximately 10-fold following an associated increase in the accumulation of ascorbic acid and anthocyanin. The accumulation of ascorbic acid and anthocyanin was suppressed by treatment with an antioxidant before irradiation and was induced by treatment with a radical-generating reagent. The contributions of ascorbic acid and anthocyanin to the total superoxide radical scavenging activity differed among ecotypes. In the Ler ecotype, ascorbic acid accumulated at twice the level of that in the Col ecotype, and induction of anthocyanin was half that in Col. To confirm the activity of ascorbic acid and anthocyanin against AOS stress, the viability of the wild type and mutants (tt2, tt3,tt5, ttg1, and vtc1) was examined after gamma-irradiation. Only the plants in which ascorbic acid and anthocyanin were induced had the ability to grow and flower.     

Langerhans cells within the follicular epithelium and the intradermal sweat duct in equine insect hypersensitivity "Kasen"

Histopathologic and electron microscopic observations were given on Langerhans cells (LCs) within the follicular epithelium (FE) and intradermal sweat duct (ISD) of equine "Kasen". By light microscopy, LCs were present in the greatest numbers within the FE and ISD than within the epidermal layer and the normal skin, with an occasional formation of several aggregated foci. By electron microscopy, LCs within the FE and ISD widely extended their dendritic processes between the keratinocytes and contained Birbeck granules (Bgs), mitochondria, rough endoplasmic reticula and ribosomes in the cytoplasm. Numerous Type 2 LCs, with a number of Bgs and endocytosis, and Type 3 LCs, with multivesicular bodies and endosomes of various sizes, were recognized within the FE and ISD, although inactive Type 1 LCs, with a narrow and lucid cytoplasm, were rarely seen. LCs observed within the FE and ISD in the "Kasen" skin lesions might express the particular stage corresponded to recognize, intake and process the antigens which permeate them.     

Extraskeletal myxoid chondrosarcoma with systemic metastasis in a five-month-old irish setter dog

Extraskeletal myxoid chondrosarcoma was found in a five-month-old male Irish setter dog. At necropsy, the largest mass, measuring 15 × 13 × 13 cm, was found in the right caudal lobe of the lung, and metastatic small masses were observed in multiple organs. Microscopically, the tumor comprised mainly atypical mesenchymal cells and myxoid stroma, which stained positively with Alcian blue. Immunohistochemically, the tumor cells stained positively for vimentin, S-100 protein, neuron-specific enolase, calretinin, and chromogranin A. Ultrastructurally, the cytoplasm of the tumor cells was comprised abundant rough endoplasmic reticulum, mitochondria, Golgi complex, free ribosomes and short irregular microvillous processes extending from the cytoplasm. Based on these pathological findings, this tumor was diagnosed as extraskeletal myxoid chondrosarcoma.     

Density estimation of the giant jellyfish Nemopilema nomurai around Japan using an alternative modified detection function for left truncation in a line transect survey

Information on the distribution and bloom scale of the giant jellyfish Nemopilema nomurai is necessary to understand the impact on fisheries. In October 2009, we had an opportunity for visual counting of N. nomurai around Japan from a research/training vessel. To take into account possibly severe missed detection of N. nomurai in the close vicinity of the vessel, detection functions were modified by introducing an additional parameter δ expressing the proportion of detectable N. nomurai within a threshold perpendicular distance. Some covariate effects in the detection process were also investigated. The δ-modified hazard-rate detection function with δ variation parameter of jellyfish body size was chosen as the best-fit one by AIC model selection. The results suggested that a higher proportion (>30 % or more) of N. nomurai were undetected in the vicinity of vessel. The δ-modified model had an advantage of no loss of left-truncated data and provided better estimation of density with the smaller coefficient of variation than the conventional left truncation approach. It is applicable to other line transect surveys, e.g. aerial surveys. High densities of N. nomurai were found in the central part of the Sea of Japan and off Iwate Prefecture.     

Shear strengths of wood measured by various short beam shear test methods

We conducted three types of short beam shear tests of western hemlock (Tsuga heterophylla Sarg.) under various span/depth ratios, and examined whether the maximum shear stress was used as the shear strength. The following results were obtained. (1) In the short beam shear tests under the three-point loading method, it was difficult to have the specimen failing by horizontal shear. We thought that this method should not be recommended for determining the shear strength of wood. (2) In the short beam shear tests under the asymmetric four-point loading of the specimen with a rectangular cross-section, the failure caused by horizontal shear occurred under some span/depth ratio range. Nevertheless, this range was dependent on the specimen geometry and was quite restricted. We therefore think that this method should not be recommended for determining the shear strength of wood. (3) In the short beam shear tests under the asymmetric four-point loading of the I-shaped specimen, failure caused by horizontal shear occurred under the span/depth ratio range wider than that applicable for the asymmetric four-point loading of the specimen with a rectangular cross-section. The maximum shear stress was stable in a certain span/depth ratio range and the value of the maximum shear stress is effective as a parameter for comparing the shearing strength of materials with each other.     

A case report of traumatic neuroma of the cervical spinal cord in a dog

Traumatic neuroma of the cervical spinal cord was diagnosed in a 14-year-old male mixed-breed dog. A gross view showed two intradural extramedullary masses, measuring 1 and 0.6 cm in length and 0.7 and 0.4 cm in diameter, attached to the left side of the spinal cord at the level of the sixth and seventh cervical vertebrae. Microscopically, the cervical spinal masses comprised interlacing fascicles of axons and Schwann cells surrounded by collagenous stroma. Immunohistochemically, the fascicles were stained positively for neurofilament and S-100 proteins. Ultrastructurally, variably sized myelinated fibers and onion bulb-like structures were observed. To our knowledge, this is the first report of a traumatic neuroma in the cervical spinal cord of a dog.
