Inhibitors of cyclin‐dependent kinases, as roscovitine, have been used to prevent the spontaneous resumption of meiosis in vitro and to improve the oocyte developmental competence. In this study, the interference of oil overlay on the reversible arrest capacity of roscovitine in sheep oocytes as well as its effects on cumulus expansion was evaluated. For this, cumulus‐oocyte complexes (COCs) were cultured for 20 h in TCM 199 with 10% foetal bovine serum (Control) containing 75 μm roscovitine (Rosco). Subsequently, they were in vitro matured (IVM) for further 18 h in inhibitor‐free medium with LH and FSH. The culture was performed in Petri dishes under mineral oil (+) or in 96 well plates without oil overlay (?) at 38.5°C and 5% CO2. At 20 and 38 h, the cumulus expansion and nuclear maturation were evaluated under stereomicroscope and by Hoechst 33342 staining, respectively. No group presented cumulus expansion at 20 h. After additional culture with gonadotrophins, a significant rate of COCs from both Control groups (+/?) exhibited total expansion while in both Rosco groups (+/?) the partial expansion prevailed. Among the oocytes treated with roscovitine, 65.2% were kept at GV in the absence of oil overlay while 40.6% of them reached MII under oil cover (p < 0.05). This meiotic arrest was reversible, and proper meiosis progression also occurred in the Control groups (+/?). So, the culture system without oil overlay improved the meiotic inhibition promoted by roscovitine without affecting the cumulus expansion rate or the subsequent meiosis progression.  相似文献

Hooked epidermal appendages (trichomes) on leaves of field bean cultivars effectively capture nymph and adult leafhoppers. Frequency of capture and capture mortality are highly correlated with trichome density. Hooked trichomes inserted at angles less than 30 degrees are ineffective in capture.  相似文献

Debris flows have deposited inorganic laminae in an alpine lake that is 75 kilometers east of the Pacific Ocean, in Ecuador. These storm-induced events were dated by radiocarbon, and the age of laminae that are less than 200 years old matches the historic record of El Nino events. From about 15,000 to about 7000 calendar years before the present, the periodicity of clastic deposition is greater than or equal to 15 years; thereafter, there is a progressive increase in frequency to periodicities of 2 to 8.5 years. This is the modern El Nino periodicity, which was established about 5000 calendar years before the present. This may reflect the onset of a steeper zonal sea surface temperature gradient, which was driven by enhanced trade winds.  相似文献

Tetralobulate (Type A) glandular trichomes are present on foliage of many wild as well as cultivated species of potatoes. These trichomes confer resistance to insect pests if their density, gland size and biochemical products are similar as those ofSolanum berthaultti Hawkes andS. polyadenium Greenm. Hybrids ofS. tuberosum L. ×S. berthaultii have been screened for resistance by measurement of phenolic oxidation activity in their Type A trichomes by use of an enzymic browning assay. Ovarian development and fecundity of Colorado potato beetles reared on the resistant potato species,Solanum berthaultii were reduced compared to females reared on the susceptible potato,S. tuberosum. Striking changes in the appearance of the digestive tract and large quantities of undigested plant material in the midgut of females reared onS. berthaultii suggest that ovarian development might be impaired by poor nutritional quality ofS. berthaultii foliage. Alternatively, enlargement of the midgut by undigested plant material might physically limit ovarian development and subsequent fecundity. These results suggest that a major impact ofS. berthaultii on CPB performance occurs in the physiology of digestion and reproduction.  相似文献

The inheritance of trichome density and droplet size was investigated in diploid and tetraploid crosses. In haploidTuberosum ×Solanum berthaultii and (S.phureja ×haploid Tuberosum) × S.berthaultii crosses, droplet size on type B trichomes was small or droplets were lacking on F1 hybrids. Parental phenotypes were recovered in the F2 and backcross generations. Trichome density was more variable within a clone that was droplet size. Density of both type A and type B trichomes appears to be under the control of small numbers of genes, as parental phenotypes were recovered in the F2 and backcross generations. The variation observed for each of the three traits studied could not be explained by hypotheses based on the segregation of one or two loci. The insect resistance ofSolanum berthaultii appears to be due to the interaction of several chemical and physical factors, thus potato breeders should handle it as a quantitatively inherited trait.  相似文献

Polyphenoloxidase (PPO) activity in the glandular trichomes of 32Solanum tuberosum ×S. berthaultii F3 hybrid clones differentially resistant to the green peach aphid,Myzus persicae (Sulzer) was determined. Within this population of segregating clones, aphid infestation levels varied 8-fold, while a 5-fold difference in glandular trichome PPO activity was observed. PPO activity and aphid resistance at three sample dates were correlated as follows: r = 0.38 (P = 0.05), r = 0.31 (P = 0.10), and r = 0.55 (P = 0.01), respectively. The total browning potential test (TBP), a modification of the PPO assay, was developed for use in screening for aphid resistance. The TBP procedure is rapid, inexpensive, and highly effective in identifying aphid resistant clones.  相似文献

Morphological differences between old-growth trees and saplings of Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) may extend to differences in needle anatomy. We used microscopy with image analysis to compare and quantify anatomical parameters in cross sections of previous-year needles of old-growth Douglas-fir trees and saplings at the Wind River Canopy Crane site in Washington and at three sites in the Cascade Mountains of Oregon. We also compared needle anatomy across a chronosequence of 10-, 20-, 40- and 450-year-old Douglas-fir trees from the Wind River site. Anatomy differed significantly between needles of old-growth trees and saplings at all sites, suggesting a developmental change in needle anatomy with increasing tree age. Compared with needles of old-growth trees, needles of saplings were longer and had proportionately smaller vascular cylinders, larger resin canals and few hypodermal cells. Astrosclereids, which sequester lignin in their secondary cell walls and occupy space otherwise filled by photosynthetic cells, were scarce in needles of saplings but abundant in needles of old-growth trees. Needles of old-growth trees had an average of 11% less photosynthetic mesophyll area than needles of saplings. The percentage of non-photosynthetic area in needles increased significantly with increasing tree age from the chronosequence of 10-, 20-, 40- and 450-year-old trees at the Wind River site. This reduction in photosynthetic area may contribute to decreased growth rates in old trees.  相似文献

Pinto bean (Phaseolus vulgaris L.) and soybean [Glycine max (L.) Merv.] were exposed to O3 to determine the interactions of growth temperature, exposure temperature, K nutrition and doses of O3 on their foliar sensitivity. Pinto bean developed more foliar injury than soybean. Pinto bean were most sensitive when grown and exposed at 28°C. Growth and exposure temperature interacted in the development of foliar injury on pinto bean, but only growth temperature influenced the amount of foliar injury on soybean. Both species developed more foliar injury when grown with low K nutrition. There was no relationship between foliar injury and reducing sugars or sucrose content of the leaves.  相似文献

The impact of acid fog on foliar leaching and root uptake of Ca, Mg, and K by Douglas-fir (Pseudotsuga menziesii) seedlings was examined. In a factorial experiment, 1-year old seedlings were grown in solution culture at two levels of nutrient availability (low and moderate) and exposed twice a week (4 hr per event) for 12 weeks to fog at pH 5.6 or pH 3.1. Throughfall enrichment of Ca, Mg and K was determined from drip collectors at the base of each seedling and root uptake rates for trees under the moderate nutrient regime were evaluated by monitoring nutrient solution depletion. Throughfall enrichment was higher in the pH 3.1 fog than the pH 5.6 fog but much of the enrichment appeared to be wash off of precipitate from previous fogs. The amounts of nutrients coming off of the foliage with the low pH fog were small relative to the daily uptake rates. Foliar concentrations of K and Mg at the end of the exposures were lower under the low nutrient regime but were not affected by fog pH. Comparisons of wax weight and examinations of epicuticular wax by electron microscopy did not indicate a significant impact from exposure to the low pH fog.  相似文献
