Objective-To evaluate adverse effects and survival times in dogs with osteosarcoma that received a single SC infusion of carboplatin as adjunctive chemotherapeutic treatment following limb amputation or limb-sparing surgery. Design-Retrospective case series. Animals-17 client-owned dogs with spontaneously occurring osteosarcoma. Procedures-Medical records of dogs that underwent limb amputation or limb-sparing surgery followed by a single continuous SC infusion of carboplatin (total dose, 300 mg/m(2) infused over 3, 5, or 7 days) were evaluated. Signalment, tumor location, type of surgery (amputation or limb-sparing), duration of carboplatin infusion, results of hematologic and serum biochemical analyses, and adverse effects were recorded. Kaplan-Meier survival analysis was performed. Results-Median survival time for all dogs was 365 days. Nine dogs had adverse bone marrow-related (hematologic) effects, 1 had adverse gastrointestinal effects, and 7 had infections at the surgical site. No significant differences were detected in survival times of dogs grouped according to tumor location, type of surgery, duration of carboplatin infusion, or development of postoperative infection. Conclusions and Clinical Relevance-Median survival time and adverse effects in dogs with osteosarcoma that received a single SC infusion of carboplatin over a 3-, 5-, or 7-day period as adjunctive treatment following limb amputation or limb-sparing surgery were comparable to those of previously reported chemotherapy protocols requiring IV drug administration over several weeks. Further investigation is needed to evaluate the efficacy of this protocol as adjunctive treatment for osteosarcoma and other tumors in dogs.  相似文献

Our lack of understanding of the immune microenvironment in canine osteosarcoma (cOSA) has limited the identification of potential immunotherapeutic targets. In particular, our ability to utilize readily available tissue from a dog's primary tumour to predict the type and extent of immune response in their pulmonary metastatic lesions is unknown. We, therefore, collected 21 matched pairs of primary tumours and pulmonary metastatic lesions from dogs with OSA and performed immunohistochemistry to quantify T‐lymphocyte (CD3), FOXP3+ cell, B‐lymphocyte (Pax‐5), and CD204+ macrophage infiltration. We found that T‐lymphocytes and FOXP3+ infiltrates in primary tumours positively correlated with that of metastatic lesions (ρ = 0.512, P = 0.038 and ρ = 0.698, P = 0.007, respectively), while a strong trend existed for CD204+ infiltrates (ρ = 0.404, P = 0.087). We also observed T‐ and B‐lymphocytes, and CD204+ macrophages to be significantly higher in a dog's pulmonary metastasis compared to their primary tumour (P = 0.018, P = 0.018, P = 0.016, respectively), while FOXP3+ cells were only significantly higher in metastases when all primary tumour and metastasis lesions were compared without pairing (P = 0.036). Together, these findings suggest that the metastatic immune microenvironment may be influenced by that of the primary cOSA, and that primary tumour immune biomarkers could potentially be applied to predict immunotherapeutic responses in gross metastatic disease. We, therefore, provide a rationale for the treatment of cOSA pulmonary metastases with immunotherapeutics that enhance the anti‐tumour activity of these immune cells, particularly in dogs with moderate to high immune cell infiltration in their primary tumours.  相似文献

Immunotherapeutic strategies have shown promise for the treatment of canine osteosarcoma (cOSA). Very little is known about the immune microenvironment within cOSA, however, limiting our ability to identify potential immune targets and biomarkers of therapeutic response. We therefore prospectively assessed the disease‐free interval (DFI) and overall survival time (ST) of 30 dogs with cOSA treated with amputation and six doses of adjuvant carboplatin. We then quantified lymphocytic (CD3+, FOXP3+) and macrophage (CD204+) infiltrates within the primary tumours of this cohort using immunohistochemistry, and evaluated their association with outcome. Overall, the median DFI and ST were 392 and 455 days, respectively. The median number of CD3+ and FOXP3+ infiltrates were 45.8 cells/mm2 (4.6‐607.6 cells/mm2) and 8.5 mm2 (0‐163.1 cells/mm2), respectively. The median area of CD204+ macrophages was 4.7% (1.3%‐23.3%), and dogs with tumours containing greater than 4.7% CD204+ macrophages experienced a significantly longer DFI (P = 0.016). Interestingly, a significantly lower percentage of CD204+ macrophages was detected in cOSA arising from the proximal humerus compared to other appendicular bone locations (P = 0.016). Lymphocytic infiltrates did not appear to correlate with outcome in cOSA. Overall, our findings suggest that macrophages may play a role in inhibiting cOSA progression, as has been suggested in human osteosarcoma.  相似文献

Macrotermes termitaria are conspicuous features of savannah ecosystems in the Sudanian and Sahelian zones of West Africa. The mounds, alive or abandoned, are a major source of heterogeneity in the landscape. The purpose of the present study was to assess the impact of termitaria on tree community in a state forest of the Sudanian regional centre (Tiogo forest, Burkina Faso), under controlled burning and grazing experiments. A comparative inventory was carried out in a split-plot experiment (16 subplots of 2500 m2): 8 subplots where fire regime and grazing were controlled and 8 subplots exposed to grazing and with annual prescribed fire since 1992. All tree individuals (≥1.5 m) were recorded, both on termitaria and outside and their basal area at stump level was measured. A total of 61 observed (or 65.7 ± 2.4 estimated) tree species were recorded on 28 Macrotermes subhyalinus mounds (54 observed species or 60.8 ± 3.3 estimated), the immediate surroundings (44 observed and 59.0 ± 0.0 estimated species) and the rest of subplots (56 observed and 63.6 ± 0.0 estimated). Specific density was higher on mounds in comparison with the surroundings (P < 0.05). Results showed that termitaria played a key role in maintaining higher species diversity as compared to their surroundings (P < 0.05). Differences in species diversity between termitaria and immediate surroundings appeared more pronounced in disturbed plots (submitted to both fire and grazing). Some species, such as Tamarindus indica, Boscia senegalensis, Cadaba farinosa, Capparis sepiaria and Maerua angolensis are found solely on termitaria. Besides, the density of trees was significantly higher on termitaria compared to the surrounding (P < 0.05), as well as total basal area per unit of 100 m2 area (P < 0.05). We concluded that Macrotermes termitaria play an important role as a source of heterogeneity in this Sudanian savannah woodland ecosystem. This role is particularly important in ecosystems under stresses. Termitaria acted as refuge for tree vegetation. The density and dynamics of M. subhyalinus termitaria should, therefore, be taken into account in the global strategy of the forest resources management and conservation.  相似文献

Essential oil from the leaves of Chloroxylon swietenia DC. was obtained by hydrodistillation and cold extraction, and the chemical composition was determined by gas chromatography and gas chromatography-mass spectrometry. The major identified components were limonene, geijerene, pregeijerene, germacrene D and trans-beta-ocimene. Laboratory bioassays of the essential oil and four constituents of essential oil isolates were evaluated for insecticidal, antifeedant and oviposition deterrent effects on tobacco cutworm, Spodoptera litura (F.). Toxicity was determined by topical application of the isolates at varying concentrations. Pure oil, geijerene and pregeijerene were found to be more toxic, with LD50 values of 28.6, 35.4 and 40.7 microg per larva respectively. Maximum feeding deterrence was noted for geijerene and pregeijerene, with DC(50) values of 82.5 and 95.1 microg cm(-2) respectively. Furthermore, pure oil, geijerene and pregeijerene displayed oviposition deterrence, even at low concentrations. These results indicate that these natural products may lead to useful, biodegradable, environmentally safe insect control agents.  相似文献

Peste des petits ruminants (PPR) is an acute, highly contagious disease of small ruminants caused by a morbillivirus, Peste des petits ruminants virus (PPRV). The disease is prevalent in equatorial Africa, the Middle East, and the Indian subcontinent. A live attenuated vaccine is in use in some of the countries and has been shown to provide protection for at least three years against PPR. However, the live attenuated vaccine is not robust in terms of thermotolerance. As a step towards development of a heat stable subunit vaccine, we have expressed a hemagglutinin-neuraminidase (HN) protein of PPRV in peanut plants (Arachis hypogea) in a biologically active form, possessing neuraminidase activity. Importantly, HN protein expressed in peanut plants retained its immunodominant epitopes in their natural conformation. The immunogenicity of the plant derived HN protein was analyzed in sheep upon oral immunization. Virus neutralizing antibody responses were elicited upon oral immunization of sheep in the absence of any mucosal adjuvant. In addition, anti-PPRV-HN specific cell-mediated immune responses were also detected in mucosally immunized sheep.  相似文献

A two-dimensional finite element model of solute transport in a tile — drained soil — aquifer system has been applied to study the effects of the depth of impervious layer and quality of irrigation water on salt distribution during drainage of an initially highly saline soil. The model assumes steady state water movement through partially saturated soil and to drains in the saturated zone. The exact in time numerical solution yields explicit expressions for concentration field at any future time without having to compute concentrations at intermediate times. The model facilitates predictions of long-term effects of different irrigation and drainage practices on concentration of drainage effluent and salt distribution in the soil and groundwater. The model results indicated that the depth of impervious layer from drain level, dI, does not significantly influence the salt distribution in the surface 1 m root zone of different drain spacings (drain spacing (2S)=25, 50, 75 m; drain depth (dd)=1.8 m), its effect in the aquifer becomes dominant as drain spacing increases. It was also observed that dI significantly governs the quality of drainage effluent. The salinity of drainage water increases with increasing dI in all drain spacings and this effect magnifies with time. The model was also applied to study the effects of salinity of irrigation water in four drain spacing-drain depth combinations: (2S=48 m, dd=1.0 m; 2S=67 m, dd=1.5 m; 2S=77 m, dd=2.0 m; 2S=85 m, dd=2.5 m). The results indicated that a favorable salt balance can be maintained in the root zone even while irrigating with water up to 5 dS/m salinity in drains installed at 48 to 67 m spacing and 1.0 to 1.5 m depth. Further, irrespective of the quality of irrigation water, the deep, widely spaced drains (dd=2.5 m, 2S=85 m) produced much saline drainage effluent during the initial few years of operation of the drainage system than the more shallow, closely spaced drains, thus posing a more serious effluent disposal problem.
Résumé Considérant les conséquences potentiellement sérieuses de la pollution du sol et de l'eau souterraine dans l'agriculture irriguée, il est devenu absolument nécessaire de développer des modèles de simulation en vue d'évaluer les effects à long terme des méthodes agricoles modernes. Un modèle d'éléments finis à deux dimensions du transport en solution dans un système de sol aquifère drainé au moyen de tuyaux a été développé et validé sur le terrain (Kamra et al. 1991 a, b). Le modèle assume le mouvement de l'eau à régime constant à travers un sol partiellement saturé et jusqu'aux drains dans la zone saturée. La solution numérique exacte dans le temps produit des expressions explicites pour le champ de concentration à un temps future quelconque sans avoir à calculer les concentrations aux temps intermédiares. Le modèle facilite les prédictions des effets à long terme des diverses méthodes d'irrigation et de drainage sur la concentration des effluents de drainage et sur la distribution de la salinité dans le sol et dans l'eau souterraine. Les résultats du modèle relatifs aux effets de la profondeur de la couche imperméable et de la qualité de l'eau d'irrigation sur la distribution de la salinité lors du drainage d'un sol fortement salé à l'origine sont mentionnés dans la présente communication.Les résultats du modéle ont indiqué que la profondeur de la couche imperméable depuis le niveau du drain, dI, n'influence pas d'une façon significative la distribution de la salinité dans la zone superficielle radiculaire de 1 m des divers écartements de drains (écartement de drains, 2S=25, 50, 75 m; profondeur des drains, dd=1.8 m); son effet dans l'aquifère devient dominant à mesure que l'écartement de drains augmente. On a aussi constaté que le niveau du drain dI influence d'une manière significative les effluents du drainage. La salinité de l'eau de drainage augmente à mesure que dI augmente dans tous les écartements de drains et cet effet s'amplifie avec le temps. Le modèle a été aussi appliqué pour étudier les effets de la salinité de l'eau d'irrigation dans le cas de quatre conbinaisons d'écartement de drain et de profondeur de drain: (2S=48 m, dd=1,0 m; 2S=67 m, dd=1,5 m; 2S=77 m, dd=2,0 m; 2S=85 m, dd=2,5 m). Les résultats ont indiqué qu'un bilan de salinité favorable peut être maintenu dans la zone radiculaire même en irrigant avec de l'eau d'une salinité de 5 dS/m dans des drains installés à un écartement de 48 à 67 m et une profondeur de 1,0 à 1,5 m. De plus, indépendamment de la qualité de l'eau d'irrigation les drains profonds à grand écartement (dd=2,5 m, 2S=85 m) produisaient une grande quantité d'effluents salés de drainage durant les quelques premières années de l'exploitation du système de drainage par rapport aux drains peu profonds à écartement serré, posant ainsi un problème plus sérieux d'évacuation des effluents.Les résultats du développement et de l'évaluation du modèle on montré qu'il peut être utilement employé en vue d'une évaluation judicieuse de la variation de temps escomptée dans la salinité des effluents de drainage lors de la mise en valeur des sols salins et peut ainsi aider à formuler son règlement plus sûr du point de vue environnement et les projects d'évacuation.

Allometric models are necessary for estimating biomass in terrestrial ecosystems. Generalized allometric relationship exists for many tropical trees, but species- and region-specific models are often lacking. We developed species-specific allometric models to predict aboveground biomass for 11 native tree species of the Sudanian savanna-woodlands. Diameters at the base and at breast height, with species means ranging respectively from 11 to 28 cm and 9 to 19 cm, and the height of the trees were used as predictor variables. Sampled trees spanned a wide range of sizes including the largest sizes these species can reach. As a response variable, the biomass of the trees was obtained through destructive sampling of 4 754 trees during wood harvesting. We used a stepwise multiple regression analysis with backward elimination procedure to develop models separately predicting, total biomass of the trees, stem biomass, and biomass of branches and twigs. All species-specific regression models relating biomass with measured tree dimensions were highly significant (p < 0.001). The biomass of branches and twigs was less predictable compared to stem biomass and total biomass, although their models required fewer predictors and predictor interactions. The best-fit equations for total above-ground biomass and stem biomass had R 2 > 0.70, except for the Acacia species; for branches including twig biomass, R2-values varied from 0.749 for Anogeissus leiocarpa to 0.183 for Acacia macrostachya. The use of these equations in estimating available biomass will avoid destructive sampling, and aid in planning for sustainable use of these species.  相似文献
