ABSTRACT:   Sporophyll formation in two alariaceous plants, Undaria pinnatifida and Alaria crassifolia , was studied in relation to the nutrient requirements. The sporophylls of U. pinnatifida formed zoosporangia when they had N and P contents greater than 1.4 kgN/m3 and 0.74 kgP/m3. This indicates that these values are the critical nutrient levels for zoospore formation in U. pinnatifida . In the sporophytes of A. crassifolia , many sporophylls with zoosporangia showed nutrient contents higher than 6.25 kgN/m3 and 1.70 kgP/m3. These results suggest that the U. pinnatifida can form zoospores at lower levels of N and P contents than A. crassifolia . Both species often formed zoosporangial sori on the blades in the late period of each reproductive season. The fertile parts of the blade showed N and P contents higher than the critical levels. This phenomenon indicates that the blades have the capability to form zoosporangial sori if there is a sufficient accumulation of nutrients for zoospore formation. The zoospore formation on the blade seems to be accomplished by an overflow of excess nutrients from sporophylls into the blade, or by accumulating sufficient nutrients to form sori even if the sporophylls are not formed.  相似文献

IgE-reactive beef components were examined by an immunoblot analysis using a serum from a dog with food hypersensitivity against beef. The immunoblot analysis revealed a distinct band at approximately 66 kDa and a faint band at approximately 50 kDa. The immunoblot analysis for serum IgE reactivity to bovine serum albumin (BSA) also revealed a positive band at 66 kDa. Serum IgE reactivity to the 66-kDa protein of beef was diminished by pre-incubating the serum sample with BSA. Furthermore, a positive reaction to BSA was detected in intradermal testing in the dog. These results clearly indicated that BSA was an IgE-reactive beef component in the dog with food hypersensitivity against beef.  相似文献

Triploidization is an interesting tool to produce sterile fish. In the yellowtail tetra, Astyanax altiparanae, this can be applied for aquaculture and surrogate technologies. In this study, we compared the efficacy of cold (2 C) or heat shock (38 C, 40 C, and 42 C) on triploid induction in the yellowtail tetra. The eggs were treated with cold or heat shock, 2 min postfertilization (30 min in cold shock or 2 min in heat shock). Intact embryos served as the control group. Ploidy status was confirmed by karyotyping, flow cytometry, and nuclear diameter of erythrocytes. The hatching rate decreased after cold shock (12.69 ± 15.76%) and heat shock at 42 C (0.35 ± 0.69%) in comparison with the control group (63.19 ± 16.82%). At 38 C and 40 C, hatching rates (61.29 ± 17.73% and 61.75 ± 22.1%, respectively) were not decreased. Only one triploid arose at 38 C (1/80). At 40 C, a high number of triploids arose (72/78). At 42 C, very few embryos developed into the hatching stage. A large number of haploid individuals arose after cold shock (61/75), with only one triploid. Our results indicate that heat shocking of embryos at 40 C is optimum for triploid production in the yellowtail tetra.  相似文献

To investigate the effects of processing Chinese quince fruit on the denaturation of phenolics and their food functions, fruit phenolic extracts were heated together with organic acid for up to 12 h. Chinese quince phenolic (mostly procyanidins) solution subjected to heat treatment changed from almost colorless, pale yellow, to a reddish color. Before heat treatment, the absorption spectra of polymeric procyanidins were observed only around 280 nm; after heat treatment, absorption occurred between 400 and 600 nm, which is related to the reddish color appearance. Thioacidolysis of denatured reddish phenolics showed that (-)-epicatechin subunits decreased during heat treatment and, in contrast, cyanidin increased. In addition, novel substances that could not be degraded by thioacidolysis were formed. Meanwhile, antioxidant activities, assessed by linoleic acid peroxidation, 1,1-diphenyl-2-picrylhydrazyl (DPPH), Folin-Ciocalteu, and FRAP methods, increased during heat treatment. The antiinfluenza viral activity of denatured reddish phenolics was inferior to that of intact fruit phenolics; however, they retained moderate activity. These results indicate that red coloration of fruit products of Chinese quince was mainly due to the spectral (i.e. structural) changes of procyanidins accompanied with formation of cyanidin. Increasing the length of heat treatment increased the antioxidant capacity of phenolics, and the resultant reddish phenolics retained moderate antiinfluenza viral activity.  相似文献

The spread of coronavirus disease 2019 (COVID-19) and subsequent lockdown measures have impacted economies and industries worldwide. The fisheries industry witnessed a sharp decline in demand and a slump in fish prices due to its dependence on the food service industry. It is important to quantitatively assess those fish species affected most and the extent of the pandemic’s impact on them, to take specific countermeasures. We propose a time-series analysis as an alternative to the current practice of using ad hoc year-on-year comparisons. Although the pandemic makes it difficult to construct a counterfactual approach due to the lack of an appropriate control group, we use time-series forecasting to simulate normal conditions using pre-pandemic data. In Tokyo, the unit price of fish species that were negatively impacted by the food services industry dropped by 12.65% to 14.64%, and by 26.08% to 28.22% after the declaration of a state of emergency. Seasonality, short weekly cycles, and short-term market trends are factors that affect the price of fish. Species-specific impact estimates related to the COVID-19 pandemic can allow policymakers to implement recovery measures in a more targeted and effective manner. The results of our analysis can increase fishers’ and policymakers’ awareness of the usefulness of economic analyses and incentivize them to release data to establish a system to accumulate and analyze data strategically for urgent and appropriate interventions in the fisheries industry.

A restriction fragment length polymorphism (RFLP) – based linkage map of Chinese cabbage (Brassica rapa ssp. pekinensis) (2n=20) including two agronomic traits, clubroot resistance and orange-yellow pigmentation, was constructed using doubled haploid parents. The total linkage distance was 735 cM; 63 loci were distributed into ten linkage groups. Clubroot resistance of the parental line T136-8 to the current pathotype, race 2, was predominantly controlled by a single dominant gene that originated from European turnip. The locus for clubroot resistance by the dominant major gene (CRa) was mapped on linkage group 3, and RFLP loci HC352b and HC181 were located 3 cM and 12 cM from it, respectively. The locus HC352b was identified by a 4.4 Kb Eco R I fragment, which segregated for null allele. The absence of an allelic fragment in HC352b could be interpreted by deletion in the resistance source; homozygotes for CRa could be efficiently selected by detecting null types for the marker. Orange-yellow pigmentation expressed in head inner leaves and petals was governed by a single recessive gene. The locus (Oy) for the pigmentation was mapped on linkage group 1, being located 17–19 cM from three RFLP loci that were closely linked to each other. The linkage analysis for clubroot resistance and unique pigmentation revealed some informative RFLP markers. Identification of molecular markers for clubroot resistance and other agronomically important traits would provide useful information in breeding programs of Chinese cabbage. The brown planthopper (BPH) is one of the most destructive insect pests of rice. Resistant varieties have proved to be one of the most economic and effective measures for BPH management. In this study, an indica rice ‘DV85’ showed resistance to biotype 2 of BPH by bulked seedling test, and a recombinant inbred line (RIL) population derived from a cross between a susceptible rice ‘Kinmaze’ and ‘DV85’ was phenotyped to map genetic factors conferring BPH resistance in ‘DV85′. Composite interval mapping revealed that one quantitative trait locus (QTL) with a LOD score of 10.1 was detected between XNpb202 and C1172 on chromosome 11. This QTL was designated as Qbph11. Qbph11 explained 68.4% of the phenotypic variance of BPH resistance in this population. The allele from the resistant parent ‘DV85’ at Qbph11 reduced the damage caused by BPH feeding and would be very useful in breeding resistant rice varieties via marker‐assisted selection.  相似文献

Verticillium longisporum and V. dahliae, causal agents of Verticillium wilt, are spreading through the cabbage fields of Gunma Prefecture. Using the V. longisporum-specific intron within the 18S rDNA and differences between ITS 5.8S rDNA sequences in Japanese isolates of V. longisporum and V. dahliae, we developed three quantitative nested real-time (QNRT) PCR assays. The QNRT-PCR quantification of V. longisporum or V. dahliae in cabbage field soil was consistent with the severity of Verticillium wilt disease in those fields. In field trials of resistant cultivar YR Ranpo grown for three seasons in soil infested with the pathogen, disease severity and pathogen density in the soil were significantly reduced in a field moderately contaminated by V. dahliae, but only slightly reduced in a highly contaminated field. These results suggest that continuous cultivation of a resistant cultivar is an effective way to reduce the pathogen population. QNRT-PCR assays provide a powerful analytical tool to evaluate the soil population dynamics of V. longisporum and V. dahliae for disease management.  相似文献

In 2019, several aquarium-reared fish died at a sea life park in Japan. Necropsy revealed micronodules on the spleen in the dotted gizzard shad (Konosirus punctatus). Seven of 16 fish exhibited microscopic multifocal granulomas associated with acid-fast bacilli in the spleen, kidney, liver, alimentary tract, mesentery, gills, and/or heart. Bacterial cultures yielded isolates from the dotted gizzard shad and a Japanese sardine (Sardinops melanostictus). Microbiological and molecular biological examinations revealed the isolates as Mycobacterium pseudoshottsii. To our knowledge, this is the first isolation of M. pseudoshottsii from aquarium-reared fish.  相似文献
