Background: Lipase enzymes are of great importance in various industries. Currently, extensive efforts have been focused on exploring new lipase producer microorganism as well as genetic and protein engineering of available lipases to improve their functional features. Methods: For screening lipase-producing lactobacilli, isolated strains were inoculated onto tributyrin agar plates. Molecular identification of lipase-producing Lactobacilli was performed by sequencing the 16Sr DNA gene, and a phylogenetic tree was constructed. The LAF_RS05195 gene, encoding lipase protein in L. fermentum isolates, was identified using specific primers, amplified by PCR (918 bp) and cloned into the pET28a (+) vector. The recombinant proteins were expressed 2, 4, 6, and 12 hours after induction with IPTG and assessed using the SDS-PAGE. Enzymatic activity of the purified recombinant protein was measured at 410 nm in the presence of ρ-NPA and ρ-NPP. Results:Among five identified native lipase-producing isolates, one isolate showed 98% similarity with Enterococcus species. The other four isolates indicated 98% similarity to L. fermentum. After purification steps with Ni-NTA column, a single protein band of about 34 kDa was detected on SDS- PAGE gel. The enzymatic activity of purified recombinant protein alongside ρ-NPA and ρ-NPP was measured to be 0.6 U/ml and 0.2 U/ml, respectively. Conclusion:In the present research, a novel lipase/esterase from L. fermentum was cloned and expressed. The novel lipase/esterase has the merit to be further studied due to its substrate specificity. Phytobiotics include a large number of active components which potentially have a growth‐promoting effects and antioxidant properties. This study was conducted to investigate the protective effects of Shirazi thyme and vitamin E on growth and plasma biochemical parameters of common carp exposed to cadmium. Fish (34 ± 3 g) were divided into four groups and fed four distinct diets including commercial diet without any additive (for control and metal only group) and supplemented with either 1% ground Shirazi thyme or 100 mg kg?1 vitamin E for 45 days. At the end of the feeding trial, all treatments except control group were exposed to sub‐lethal concentration of waterborne cadmium for 15 days and sampling was done on days 0, 7 and 15 after the metal exposure. According to the results, dietary inclusion of thyme prevented the harmful effects of cadmium and also improved fish growth and nutritional indices including weight gain, feed conversion ratio, specific growth rate and condition factor. Results also revealed that Shirazi thyme was more effective than vitamin E. Similarly, no changes in the hepatosomatic, viscerosomatic and bile somatic indices were observed. Plasma enzymes (AST, ALT, ALP and LDH) and metabolites were not altered due to thyme supplementation compared to the control. In conclusion, dietary supplementation of 1% ground Shiraz thyme improved the growth and health status of fish and showed better hepatorenoprotective properties than vitamin E during waterborne cadmium exposure in common carp juveniles. It might be conceivable to consider Shirazi thyme as a potential phytobiotic for incorporation in fish diet. 相似文献

A study was conducted to investigate the effects of skid trail slope and traffic levels on soil disturbances at two soil depths
(0–10 and 10–20 cm). The treatments were set at four traffic levels (2, 7, 12 and 20), two slope classes (<20% and >20%) and
two soil depths (0–10 and 10–20 cm). Results show that skidder traffic, longitudinal slope and soil depth have significant
effect on soil bulk density in skid trail. Comparison of average soil bulk density in different traffic levels shows that
there are significant differences in average bulk density between different traffic levels and control (p<0.05). The average bulk densities in different slopes and soil depths are significantly increased with increase in traffic
levels, maximized at 12 passes (p<0.05), but there are no significant differences between 12 and 20 passes. The interaction effects between traffic and soil
depth are significant (F
0.05,3=0.109, p<0.001). For all traffic treatments, there are significant differences in soil moisture content between the two slope classes
and the two depths (p<0.001). However, the interaction effects between traffic levels and slope classes are not significant (p >0.05), although skidder traffic and slope affected soil moisture content. 相似文献

- In order to increase their catches, longliners targeting sharks in the Gulf of Gabes, Tunisia, have increased the number of hooks per basket and have included new bait types instead of mackerel (Scomber scombrus). This paper reports the effects of these changes on catch composition, catch rates, size distribution, life‐stage captures, and mortality at haulback.
- Data from 48 and 96 longline sets, carried out during the shark fishing seasons of 2007 and 2008, with two hooks per basket, and 2016 and 2017, with five hooks per basket, respectively, were examined to assess the effect of gear change. Moreover, the effects of bait types were inspected based on 33 fishing sets using whole mackerel, 19 using salema (Sarpa salpa) halves, and 27 using pieces of stingray (Dasyatis spp.), sampled during 2016 and 2017.
- The species composition indicated that longliners expand the vertical distribution of their hooks to operate from the surface to the bottom. The catches with both longline designs were dominated by sandbar shark (Carcharhinus plumbeus). Despite gear changes, the catch rate of C. plumbeus decreased by 42.21%, suggesting population decline. The size composition indicated a shift towards larger specimens, mainly adult females, which could jeopardize the shark population. Moreover, the mortality of the main species at haulback seems to be affected by the design of the longline.
- The bait used, mainly pieces of stingray, significantly increased the catch rate of sandbar shark, which suggests a greater attractiveness of the new bait; however, fish size and mortality rates at haulback were unchanged in response to bait variation.
- The fishery operates in shark nursery grounds, which exposes these fish (principally C. plumbeus) to considerable exploitation pressure. The new fishing practices intensified the pressure and thus the risk of a rapid depletion of populations. To preserve the shark species in the area, the use of new gear and attractive bait should be banned.

- This study evaluated the circle hook use as a tool for shark management in the pelagic longline fishery in the Gulf of Gabès.
- The usual J‐hook No. 2 with 10° offset, which has been traditionally used by the fishery, was compared to the 18/0 non‐offset circle hook in an alternating fashion along the main line. In total, 22 experimental longline sets were deployed through the shark fishing seasons of 2016 and 2017 to examine the effects of hook types on the catch composition, the catch rates, the hooking location, and the status at haulback.
- The catch composition differed significantly among hook types. Moreover, an overall increase in catch rates for the main species, the sandbar shark Carcharhinus plumbeus, and the shortfin mako shark Isurus oxyrinchus, was observed when using circle hooks.
- There was no size‐selective effect of circle hooks for the common species. The circle hooks were not effective at reducing at‐haulback mortality of sandbar shark. Conversely, shortfin mako and smooth‐hound shark Mustelus mustelus showed significantly lower relative mortality at haulback with circle hooks than with J‐hooks. Furthermore, circle hooks were more frequently hooked externally than the J‐hooks for the three shark species.
- Results demonstrated that the use of 18/0 non‐offset circle hooks in the pelagic shark longline fishery can reduce mortality at haulback for some species without any benefit for the dominant species, the sandbar shark.
- Overall, it is difficult to promote the adoption of the use of circle hooks as a management measure in this specialized fishery. Management measures focusing on fishing effort controls, fishing closures in critical habitats, and size limits could have significant benefits for the conservation of shark species and may help to improve the sustainability of the shark fishery in the Gulf of Gabès.
