Attack of tea plant (Camellia sinensis (L.) O. Kuntze) leaves by the tea mosquito bugHelopeltis theivora (Hemiptera: Miridae) was positively correlated to temperature and rainfall, and partially to humidity, as determined in 12 varieties during the period 2000–2002. The insect attack was severe during the months of May–September, which had high temperature and rainfall, and led to severe loss of biomass due to curling and drying up of the leaves. The biochemical response of these 12 varieties of tea to attack by the insect was determined with special reference to oxidative enzymes and flavonoid flavor components. Insect attack led to an increase in the activities of the oxidative enzymes peroxidase, ascorbate peroxidase and polyphenol oxidase. Activities of phenyl alanine ammonia lyase generally decreased as a result of insect attack, which was significant in the United Planters Association of South India (UPASI) varieties. A significant decrease in polyphenols was also obtained in UPASI varieties. HPLC analysis of catechins revealed a decrease in some of the catechins in the infected leaves. Ergot, caused by Claviceps africana , has emerged as a serious threat to sorghum hybrid seed production worldwide. In the absence of gene-for-gene-based qualitative resistance in commercial cultivars, varieties with high pollen production that can escape ergot infection are preferred. Recent demonstration of differences in ergot susceptibility among male-sterile lines has indicated the presence of partial resistance. Using chitin-specific fluorescin-isothiocyanate-conjugated wheat germ agglutin and callose-specific aniline blue, this study investigated the process of sorghum ovary colonization by C. africana . Conidia germinated within 24 h after inoculation (a.i.); the pathogen was established in the ovary by 79 h a.i., and at least half of the ovary was converted into sphacelial tissue by 120 h a.i. Changes in fungal cell wall chitin content and strategic callose deposition in the host tissue were associated with penetration and invasion of the ovary. The rate of ovary colonization differed in three male-sterile lines that also differed in ergot susceptibility. This work demonstrates a possible histological basis for partial resistance in male-sterile sorghum lines that could lay the foundation for variety improvement through further breeding and selection.  相似文献

ABSTRACT

The aim of this study was to prepare the refined oil derived from Sardinella longiceps by a step-wise procedure of degumming, bleaching, and deodorization. The refined sardine oil showed greater susceptibility toward oxidation than the crude oil, but the addition of ethyl acetate fractions of the red seaweeds Hypnea musciformis, Kappaphycus alvarezii, and Jania rubens with great antioxidant potential significantly increased the oxidative stability of the refined oil. The refined oil supplemented with the ethyl acetate fraction of these seaweeds indicated more noteworthy oxidative stability indices (≥ 0. 51 h) than α-tocopherol. The induction time was greater for refined oil added with H. musciformis (1.26 h) than butylated hydroxytoluene (1.04 h) and α-tocopherol (0.4 h). No traces of aromatic (δ 6.7–7.5 ppm), hydroperoxides, and aldehydes (δ 9–10 ppm) in the 1H-NMR spectra of the refined oil supplemented with seaweed extracts at the baseline and after the accelerated storage study demonstrated that the seaweed extracts were able to prevent the formation of secondary oxidation products in the unsaturated system during storage. This study demonstrated the potential of seaweeds as natural alternatives to the synthetic antioxidants to prevent the rancidity of the refined fish for use in food and pharmaceutical industries.  相似文献

ABSTRACT

A protein hydrolysate was prepared from proteins of tuna dark muscle by-product. The hydrolysis conditions (time, temperature, pH, and enzyme concentration) using Alcalase was optimized by response surface methodology (RSM). The regression coefficient close to 1.0, observed during experimental and validation runs, indicated the validity of the model. The hydrolysate produced under the optimum conditions determined by RSM has a low rate of peptide fraction of molecular weight of 4–1 kDa. Meanwhile, the results obtained by hydrolysis under optimal conditions determined by a complementary study (temperature 55°C, time 60 min, 1% enzyme concentration, and pH 8.5) show that the hydrolysate produced has a height rate of the peptide fraction of molecular weight of 4–1 kDa. The amino acid composition of the protein hydrolysate prepared proved to have the potential for application as an ingredient in balanced fish diets and as a source of nitrogen in microbial growth media.  相似文献

The everted gut sac technique has been used to investigate the effect of Vibrio vulnificus on water and electrolyte (Na+, K+, Cl, HCO3 ) transport on the intestine of sea bream (Sparus aurata L.). Both the anterior and the posterior intestine were incubated in a medium containing 108 V. vulnificus cells ml−1 at 25°C for 2 h. The presence of V. vulnificus resulted in a significant reduction (P < 0.05) of water absorption in the anterior intestine, while sodium absorption in the anterior (P < 0.01) and posterior (P < 0.05) intestine was elevated. Chloride absorption was increased, but the changed was not significant, while potassium absorption decreased significantly (P < 0.05), but only in the posterior intestine. Incubation the sea bream intestine with V. vulnificus did not affect carbonate secretion in the anterior segment, whereas high secretion was stimulated in the posterior segment (P < 0.01). Histological evaluations demonstrated damage in the anterior intestine of sea bream that was characterized by the detachment of degenerative enterocytes, alterations in the microvilli, and the presence of a heterogenous cell population, indicating inflammation. Based on our results, we conclude that V. vulnificus caused cell damage to the intestine of sea bream and that the anterior intestine is more susceptible than the posterior part of the intestine. Several hypotheses are suggested to explain our observations, such as the presence of higher numbers of villosities in the anterior intestine than in the posterior one and/or the presence of endogenous bacteria in the posterior intestine which may have a protector role.  相似文献

The studied soils were collected from a 24-year long-term fertilizer experiment with rice (Oryza sativa L.) grown on tropical Typic Endoaquepts during monsoon season only. The organic carbon (OC) and total nitrogen (TN) status of the soils improved with the N100P22K42 treatment than the initial values at the start of the experiment. The available P status of the soils depleted in the N100P11K22 and N0P0K0 treatments than the initial value. Soil microbial biomass carbon (MBC) and basal soil respiration (BSR), activities of urease, acid and alkaline phosphatases as well as fluorescein diacetate hydrolyzing activity (FDHA) were the highest in the N100P22K42 treatment. The highest percentage of MBC in OC in the N100P22K42 treatment revealed the accumulation of organic matter with an overall improvement in nutrient status of soil, since MBC is a labile pool of plant nutrients. Long-term application of fertilizers (N100P22K42) had no deleterious effect on microbial and biochemical soil quality parameters. Rice grain yields were inconsistent with N100P0K0 and N0P0K0 while that of N100P22K42, N100P11K22 and N100P22K0 increased gradually during the course of the experiment for each block period of six years. The grain yield of rice showed high significant correlation with the microbial and biochemical soil parameters.  相似文献

Proper doses of nitrogenous fertilizer are most important for rice production system because a large part of the nitrogen may be lost if it is not applied judiciously. A study was conducted covering five blocks of Balasore and two blocks of Bhadrak districts. Soil samples were collected randomly, and field visit was conducted during peak vegetative stage of rice. Two approaches have been used in this study for estimating the site-specific nitrogen (N) requirement in the study area. In one approach, geostatisical analysis and kriging was used to develop the soil test–based N recommendation map by which a minimum of 72 kg N ha?1 and maximum of 94 kg N ha?1 were recommended. In a second approach, remote sensing was used and N recommendation map was developed using the moderate-resolution imaging spectroradiometer (MODIS) leaf area index (LAI) and normalized difference vegetation index (NDVI) satellite data, and a minimum requirement of 60 kg N ha?1 and maximum of 120 kg N ha?1 was estimated through this approach.  相似文献

The present study was conducted to assess the effect of soil salinity on yield attributes as well as nutrient accumulation in different plant parts of seven Brassica cultivars from two different species raised in pot culture experiment with two levels of salinity treatments along with control corresponding to soil electrical conductivity (EC) values of 1.65 (S0), 4.50 (S1) and 6.76 (S2) dS m?1. The experiment was consisted of twelve replications in a completely randomized design. Imposition of salinity stress affected various yield attributing characters including plant height, which ultimately led to severe yield reduction. However, tolerant cultivars, CS 52 and CS 54 performed better under salt treatment showing lesser yield loss. Salinity stress reduced the nitrogen (N) content in leaves of the Brassica plants, which reflected in decreased seed protein content. Reduced accumulation of iron (Fe), manganese (Mn) and zinc (Zn) was observed in leaf, stem and root at flowering and post-flowering stages, while CS 52 and CS 54 showed less reduction than susceptible cultivars under salinity stress.  相似文献
