Helicobacter pylori infects the stomachs of nearly a half the human population, yet most infected individuals remain asymptomatic, which suggests that there is a host defense against this bacterium. Because H. pylori is rarely found in deeper portions of the gastric mucosa, where O-glycans are expressed that have terminal alpha1,4-linked N-acetylglucosamine, we tested whether these O-glycans might affect H. pylori growth. Here, we report that these O-glycans have antimicrobial activity against H. pylori, inhibiting its biosynthesis of cholesteryl-alpha-D-glucopyranoside, a major cell wall component. Thus, the unique O-glycans in gastric mucin appeared to function as a natural antibiotic, protecting the host from H. pylori infection.  相似文献

We examined the effects of deep percolation on dissolved inorganic nitrogen (DIN) exports from two adjacent weathered granite headwater catchments with different deep percolations of water in the Kiryu Experimental Watershed (KEW), Japan. The DIN output in streamflow was estimated from a regression equation between stream discharge and the DIN load, determined from both monthly sampling data and event-based sampling data. The range of deep percolation of DIN was estimated by multiplying observed DIN concentrations in bedrock groundwater by the deep percolation of water estimated from an annual water budget analysis. We found that the deep percolation of DIN corresponded to 34–76 % and ?18 to 8 % of the total DIN output in catchments where the deep percolation of water was 37–45 % and ?6 to 3 % of annual precipitation, respectively. This means that the deep percolation of DIN is not negligible when estimating the total output of DIN in the former catchment. Moreover, the fact that deep percolating water from upper catchments discharged into a lower catchment in KEW suggests that deep percolation of DIN affects downstream N budgets. Therefore, it is important to account for the deep percolation of DIN when evaluating N budgets in forested headwater catchments as well as downstream catchments.  相似文献

Granulocyte colony-stimulating factor (G-CSF) is widely used to mobilize peripheral blood stem cells, and expected to restore cardiac function for patients with coronary artery diseases as a consequence of progression of atherosclerosis. Safety issues related to the administration of G-CSF to these patients, however, are still under study. The animal model for atherosclerosis was produced by feeding miniature swine a high-cholesterol diet for 3 months. G-CSF (5 or 10 microg/kg/day) was given to the animal model by daily subcutaneous injections for 10 days and 20 main arteries were evaluated pathologically. In addition, the general toxicological effects were studied on clinical signs, body weight, hematology, blood chemistry and pathology. In the G-CSF-treated groups, a variety of changes related to the major pharmacological activity of G-CSF including an increase in white blood cell (WBC) counts were observed. In many arteries, atherosclerotic lesions similar to Type I-V of the proposed classification by the American Heart Association were observed. No effects of the G-CSF treatment were seen on the histopathological findings, incidence, severity or distribution of atherosclerotic lesions. In addition, no infiltration of neutrophils to the lesions was observed. These findings suggest that the administration of G-CSF causes neither exacerbation or modification of atherosclerotic lesions nor adverse changes despite that a sufficient increase in WBC counts could be achieved in the peripheral blood.  相似文献
