This study compared the amino acid (AA) profile of five residues (original forage, borate-phosphate buffer residue (BPR), neutral detergent fiber residue with (NDF+) and without (NDF-) sodium sulfite, and acid detergent fiber residue (ADF). Fourteen grasses and legumes from tropical and temperate regions were used in this study. The use of sodium sulfite did not affect the NDF concentration, but the NDF insoluble protein was lower (P < 0.05) in the NDF+ than in the NDF- (3.9 vs 4.5% DM, respectively). For all of the amino acids tested, the amino acid content, expressed as a percentage of CP, was lower in the ADF residue than in the original forage. There were no differences in the amino acid concentrations of the NDF- and NDF+ extracts (P > 0.05). Only in the case of methionine was there a difference in the amount of amino acid when the original forage was compared with the BPR (1.84 vs 1.45 % CP). When the AA profile of each residue was corrected for the AA content of the ADF, no difference was observed between the AA profile of the original forage and of the BPR (P > 0.05). Similar to the result without correction for the amino acids in ADF, the AA profiles of the NDF+ and NDF- fractions were similar (P > 0.05). From this result, we infer that the sodium sulfite had similar effects on all AA in the NDF residue that we tested. There were differences in amino acid concentrations in the original forage and the NDF residues for several amino acids (Met, Cys, Lys, Thr, Arg, Ile, Leu, and Phe) (P < 0.05). When the amino acid values of the original forage and the BPR were used with animal data in the Cornell Net Carbohydrate and Protein System model, few differences in animal predicted performance were evident. These findings suggest that the AA profile of the original forage can be used to predict the AA profile of the undegraded intake protein instead of using the borate-phosphate buffer residue for amino acid analyses. This would simplify obtaining feed amino acid values for use in the Cornell Net Carbohydrate and Protein System.  相似文献

In Sweden, 90% of ammonia (NH3) emissions to the atmosphere originate from agriculture, predominantly from animal manure handling. It is well known that incorporation of manure into soil can reduce NH3 emissions after spreading. However, there is a risk of increased nitrous oxide (N2O) and methane (CH4) emissions caused by bacterial activity and limited oxygen availability under these conditions. A full‐scale injector was developed and evaluated in a field experiment on grassland. Cattle slurry was either injected in closed slots 5 cm below ground or band spread on the soil surface above the crop canopy at a rate of 25 t ha?1. In a control treatment, no slurry was applied. During a 5‐day period after application, NH3 emissions were measured using an equilibrium concentration method. Gas samples for estimating CH4 and N2O emissions were also collected during 7 weeks following slurry application. Injection in closed slots resulted in no detectable NH3 emissions. After band spreading, however, NH3 emissions corresponded to nearly 40% of the total ammoniacal nitrogen in the applied slurry. The injection of slurry gave rise to a broad peak of N2O emissions during the first 3 weeks after application. In total, for the measuring period, N2O emissions corresponded to 0.75 kg N ha?1. Band spreading resulted in only a very small N2O release of about 0.2 kg N ha?1 during the same period. Except for the first sampling occasion, the soil was predominantly a sink for CH4 in all the treatments. The use of the injector without slurry application reduced grass yield during unfavourable growing conditions. In conclusion, shallow injection in closed slots seems to be a promising technique to reduce negative environmental impacts from NH3 emissions with a limited release of N2O and CH4.  相似文献

The Cornell Net Carbohydrate and Protein System (CNCPS), a mechanistic model that predicts nutrient requirements and biological values of feeds for cattle, was modified for use with sheep. Published equations were added for predicting the energy and protein requirements of sheep, with a special emphasis on dairy sheep, whose specific needs are not considered by most sheep-feeding systems. The CNCPS for cattle equations that are used to predict the supply of nutrients from each feed were modified to include new solid and liquid ruminal passage rates for sheep, and revised equations were inserted to predict metabolic fecal N. Equations were added to predict fluxes in body energy and protein reserves from BW and condition score. When evaluated with data from seven published studies (19 treatments), for which the CNCPS for sheep predicted positive ruminal N balance, the CNCPS for sheep predicted OM digestibility, which is used to predict feed ME values, with no mean bias (1.1 g/100 g of OM; P > 0.10) and a low root mean squared prediction error (RMSPE; 3.6 g/100 g of OM). Crude protein digestibility, which is used to predict N excretion, was evaluated with eight published studies (23 treatments). The model predicted CP digestibility with no mean bias (-1.9 g/100 g of CP; P > 0.10) but with a large RMSPE (7.2 g/100 g of CP). Evaluation with a data set of published studies in which the CNCPS for sheep predicted negative ruminal N balance indicated that the model tended to underpredict OM digestibility (mean bias of -3.3 g/100 g of OM, P > 0.10; RMSPE = 6.5 g/100 g of OM; n = 12) and to overpredict CP digestibility (mean bias of 2.7 g/100 g of CP, P > 0.10; RMSPE = 12.8 g/100 g of CP; n = 7). The ability of the CNCPS for sheep to predict gains and losses in shrunk BW was evaluated using data from six studies with adult sheep (13 treatments with lactating ewes and 16 with dry ewes). It accurately predicted variations in shrunk BW when diets had positive N balance (mean bias of 5.8 g/d; P > 0.10; RMSPE of 30.0 g/d; n = 15), whereas it markedly overpredicted the variations in shrunk BW when ruminal balance was negative (mean bias of 53.4 g/d, P < 0.05; RMSPE = 84.1 g/d; n = 14). These evaluations indicated that the Cornell Net Carbohydrate and Protein System for Sheep can be used to predict energy and protein requirements, feed biological values, and BW gains and losses in adult sheep.  相似文献

East Coast fever (ECF) is a highly fatal lymphoproliferative disease of cattle caused by Theileria parva, a tick-borne intracellular apicomplexan parasite. Parasite antigens that are targets of protective cytotoxic T lymphocyte (CTL) responses are required to formulate a sub-unit vaccine against ECF. A number of CTL target antigens have recently been identified and initial evaluation has shown their vaccine potential. This study aimed to evaluate whether these antigens were recognised by CTL obtained from six genetically diverse Zebu cattle immunized with a cocktail of T. parva stocks. T. parva Muguga specific polyclonal CD8(+) CTL lines were generated and confirmed to specifically lyse autologous infected cells. CTL recognition of autologous skin fibroblasts (iSF) transduced with recombinant modified vaccinia virus Ankara strain (MVA) expressing previously identified T. parva Muguga vaccine candidate antigens was evaluated using an IFN-gamma ELISpot assay. CTL lines from one of the four calves, BY120, responded specifically to cells infected with MVA expressing the antigen Tp2 and synthetic peptides were employed to map a new CTL epitope on this antigen. Immunoscreening of the T. parva genome with these CTL lines should identify novel antigens that will constitute valuable additions to the vaccine candidates currently being evaluated.  相似文献

Greywater is a potential resource of water that can be improved to meet the quality needed for irrigation. This study evaluated the performance of bark, activated charcoal, polyurethane foam and sand filters in removing biochemical oxygen demand (BOD5), surfactants, phosphorus, nitrogen and microbial indicators from greywater during start-up and steady state. In column experiments, 0.6?m high filters (diameter 20?cm) were fed for 113?days with artificial greywater at a hydraulic loading rate of 0.032?m3?m?2?day?1 and an organic loading rate of 0.014?kg BOD5 m?2?day?1. Bark and activated charcoal efficiently reduced the concentrations of organics (BOD5), surfactants (methylene blue active substances??MBAS), total phosphorus (Tot-P) and total thermotolerant coliform numbers, while sand and foam were less efficient. Bark, activated charcoal, foam and sand reduced influent BOD5 by 98, 97, 37 and 75?%; MBAS by >99, >99, 73 and 96?%; Tot-P by 97, 91, 36 and 78?%; and total nitrogen by 19, 98, 13 and 5?%, respectively. BOD5 and MBAS were efficiently reduced directly from start-up by bark and activated charcoal, while foam needed 30?days to achieve about 50?% reduction in BOD5. Bark was the most efficient filter in reducing thermotolerant faecal coliforms (2.4 log10), while foam achieved the lowest reduction (0.5 log10). Overall, bark and activated charcoal filters appeared to be the most suitable filters for improving greywater quality to reach irrigation quality in terms of organic matter reduction. Performance of these filters under higher and fluctuating loadings and the long-term sustainability of the filter materials need further investigation.  相似文献
