Over the past decades, the general trend towards shade reduction and intensification of cacao management has led to biodiversity losses. In the Peruvian Amazon, the regional government is heavily promoting crop conversion to shift from regionally marketed foods towards cacao (Theobroma cacao) and copoazu (Theobroma grandiflorum). While this shift is already visibly impacting the farming landscape and the lives of many smallholder farmers, little is known about the reasons that drive farmers to choose certain types of Theobroma species or cacao varieties over others. In this paper, we addressed how cacao farmers perceive and manage specific and varietal Theobroma diversity. We interviewed cacao farmers (n?=?20) during a seed-exchange fair and adapted a version of the four-square analysis to explore which Theobromas are currently adopted by farmers and why. The native cacao variety (cacao chuncho) was the one cultivated by most farmers, followed by the more industrial clonal varieties. The source of seeds and seedlings for the most cultivated varieties was a mix of donations by public institutions (for clonal varieties) and informal exchange among farmers (mainly for native and criollo varieties, and species i.e. macambo (Theobroma bicolor) and cacauillo (Theobroma speciosum)). The cacao varieties incentivized by public institutions were the least desired for future investment. The motivations for farmers to plant Theobromas was mainly based on the perceived current and potential market, but their desire to invest in a given species or variety was also based on pest resistance, traditional use, farm diversification and taste. Copoazu and CCN cacao varieties are produced by many households in large areas, while macambo, cacauillo and the cacao varieties TSH and porcelana are produced by few households in small areas. Our study suggests that agroforestry systems that include alternative Theobroma species are multi-strata and more diverse than cacao-based systems. It highlights the risks of agrobiodiversity loss associated with the promotion of industrial cacao varieties, and the importance of seed and seedling access for the development of diverse farming systems. We recommend the expansion of local seed-sharing networks and the extension of public cacao-donation campaigns to neglected and underutilized Theobromas with socio-economic and environmental benefits, in order to make the local farming systems more diverse and resilient.

The twinning of techniques from biophysics and molecular biology has led to remarkable progress in understanding the molecular mechanisms of synaptic transmission. Here we review the current picture of Ca++-triggered exocytosis, which has emerged from studies of a simple cellular model, the adrenal chromaffin cell. We discuss the molecular players that have been assigned a specific role in a particular step of this process and give a brief outlook on what these insights might tell us about mechanisms of short-term plasticity at classical synapses.  相似文献

Neurotransmitter release is triggered by calcium ions and depends critically on the correct function of three types of SNARE [soluble N-ethylmaleimide-sensitive factor attachment protein (SNAP) receptor] proteins. With use of the large calyx of Held presynaptic terminal from rats, we found that cleavage of different SNARE proteins by clostridial neurotoxins caused distinct kinetic changes in neurotransmitter release. When elevating calcium ion concentration directly at the presynaptic terminal with the use of caged calcium, cleavage of SNAP-25 by botulinum toxin A (BoNT/A) produced a strong reduction in the calcium sensitivity for release, whereas cleavage of syntaxin using BoNT/C1 and synaptobrevin using tetanus toxin (TeNT) produced an all-or-nothing block without changing the kinetics of remaining vesicles. When stimulating release by calcium influx through channels, a difference between BoNT/C1 and TeNT emerged, which suggests that cleavage of synaptobrevin modifies the coupling between channels and release-competent vesicles.  相似文献

Reports of disease suppression by compost are inconsistent likely because there are no established standards for feedstock material, maturity age for application, and application rate. The overall goal of the study was to evaluate a suite of biological indicators for their ability to predict disease suppression. Indicators included both commercial available methods for compost stability (Solvita?, respiration) and metrics of soil ecology not yet adopted by the compost industry (e.g., ecoenzymes, nematode community index). Damping-off by Rhizoctonia solani on radish was chosen as a model system given its global importance, competitiveness affected by carbon quality, and lack of disease management options for organic production. Biological indicators were evaluated for their ability to consistently differentiate among curing process, maturity, and feedstock material as a function of disease severity of a seedling bioassay and a compost extract assay to test competition with R. solani growth. Compost processed as vermicompost and anaerobic digestate were more suppressive against R. solani than windrow or aerated static pile. Mature composts were more suppressive than immature components. Feedstocks containing dairy manure and/or hardwood bark tended to have suppressive qualities. In contrast, poultry manure-based components were conducive to disease. Microbial ecoenzymes active on chitin and cellulose and nematode community indices were better predictors of disease suppressiveness than microbial respiration. These indicators are quicker than plant bioassays and could be adopted as tools to certify commercial products.  相似文献

The only known case of an avian digestive system with active foregut fermentation is reported for the hoatzin (Opisthocomus hoazin), one of the world's few obligate folivorous (leaf-eating) birds. Hoatzins are one of the smallest endotherms with this form of digestion. Foregut fermentation in a flying bird may be explained by increased digestive efficiency by selection of highly fermentable and extremely patchy resources, coupled with microbial nutritional products and secondary compound detoxification. This unexpected digestive system gives a new perspective to the understanding of size limitations of vertebrate herbivores and to the evolution of foregut fermentation.  相似文献

Frequent hydration and drying of soils in arid systems can accelerate desert carbon and nitrogen mobilization due to respiration, microbial death, and release of intracellular solutes. Because desert microinvertebrates can mediate nutrient cycling, and the autotrophic components of crusts are known to be sensitive to rapid desiccation due to elevated temperatures after wetting events, we studied whether altered soil temperature and frequency of summer precipitation can also affect the composition of food web consumer functional groups. We conducted a two-year field study with experimentally-elevated temperature and frequency of summer precipitation in the Colorado Plateau desert, measuring the change in abundance of nematodes, protozoans, and microarthropods. We hypothesized that microfauna would be more adversely affected by the combination of elevated temperature and frequency of summer precipitation than either effect alone, as found previously for phototrophic crust biota. Microfauna experienced normal seasonal fluctuations in abundance, but the effect of elevated temperature and frequency of summer precipitation was statistically non-significant for most microfaunal groups, except amoebae. The seasonal increase in abundance of amoebae was reduced with combined elevated temperature and increased frequency of summer precipitation compared to either treatment alone, but comparable with control (untreated) plots. Based on our findings, we suggest that desert soil microfauna are relatively more tolerant to increases in ambient temperature and frequency of summer precipitation than the autotrophic components of biological soil crust at the surface.  相似文献
